The interaction of simian virus 40 (SV40) T antigen (T-ag) with the viral origin has served as a model for studies of site-specific recognition of a eukaryotic replication origin and the mechanism of DNA unwinding. These studies have revealed that a motif termed the "beta-hairpin" is necessary for assembly of T-ag on the SV40 origin. Herein it is demonstrated that residues at the tip of the "beta-hairpin" are needed to melt the origin-flanking regions and that the T-ag helicase domain selectively assembles around one of the newly generated single strands in a manner that accounts for its 3'-to-5' helicase activity. Furthermore, T-ags mutated at the tip of the "beta-hairpin" are defective for oligomerization on duplex DNA; however, they can assemble on hybrid duplex DNA or single-stranded DNA (ssDNA) substrates provided the strand containing the 3' extension is present. Collectively, these experiments indicate that residues at the tip of the beta-hairpin generate ssDNA in the core origin and that the ssDNA is essential for subsequent oligomerization events.  相似文献

A new approach toward understanding marine ecosystems has emerged through the integration of ecological physiology and macroecology. This multidisciplinary approach, titled here marine macrophysiology, facilitates unique insight into the foundation of macro-scale ecological patterns, such as biogeographic distributions, via examination of functional attributes of marine organisms across large spatial scales. For example, these broad-scale physiological inquiries confer the ability to directly assess the abundant-center hypothesis (aka Brown's principle) which proposes that species have decreased performance toward their range edges. By extension, the marine macrophysiological perspective also stands to clarify our understanding of more complex macro-scale phenomena such as biological invasions, the design of marine protected areas, and species' responses to global climate change. In this article, we review recent marine macrophysiology research and offer insights into future directions for this emerging field.  相似文献

Paracetamol, sulfathiazole and L-glutamic acid are presented as examples of pharmaceutical crystal polymorphic systems. The effect of N-acylated sulfathiazole derivatives (3-6) on sulfathiazole crystallisation is discussed, and possible modes of action presented. Methods for the control of the crystal polymorphism of L-glutamic acid which utilise the principles of conformation mimicry and co-operative binding are presented. The preparation of a series of bis-amides of EDTA derived from sulfathiazole, 5-aminoisophthalic acid and 4-hydroxyaniline (i.e. compounds 9a-c) is presented, as is data on the effect of these compounds on the crystallisation of, respectively, sulfathiazole, L-glutamic acid and paracetamol.  相似文献

In species in which adults of the two sexes show long-term association, males often engage in acts of assistance (“services”) aimed at females and/or their young in various behavioural contexts that are not reciprocated. We conducted a quantitative review of the primate literature on sex differences in involvement in protection against predation. We found that males were more likely to be at the front of group progressions, especially in contexts of increased risk, were more vigilant, sometimes to the point of becoming sentinels, were more active in mobbing, and were far more likely to counter-attack felids and monkey-eating raptors than females did. Our evaluation showed that these services may be an expression of male parental care or aimed at bonded partners, but we also encountered many cases in which non-sires, unrelated to either females or immatures, provide them. We hence investigate explanations invoking group augmentation or costly signalling. Currently, available data on male anti-predation services in nonhuman primates do not allow us to distinguish among them, although costly signalling better explains data on birds and humans. We develop predictions that will allow more detailed tests of the various hypotheses for this understudied phenomenon.  相似文献

Life abounds with examples of conspecifics actively cooperating to a common end, despite conflicts of interest being expected concerning how much each individual should contribute. Mathematical models typically find that such conflict can be resolved by partial-response strategies, leading investors to contribute relatively equitably. Using a case study approach, we show that such model expectations can be contradicted in at least four disparate contexts: (i) bi-parental care; (ii) cooperative breeding; (iii) cooperative hunting; and (iv) human cooperation. We highlight that: (a) marked variation in contributions is commonplace; and (b) individuals can often respond positively rather than negatively to the contributions of others. Existing models have surprisingly limited power in explaining these phenomena. Here, we propose that, although among-individual variation in cooperative contributions will be influenced by differential costs and benefits, there is likely to be a strong genetic or epigenetic component. We then suggest that selection can maintain high investors (key individuals) when their contributions promote support by increasing the benefits and/or reducing the costs for others. Our intentions are to raise awareness in—and provide testable hypotheses of—two of the most poorly understood, yet integral, questions regarding cooperative ventures: why do individuals vary in their contributions and when does cooperation beget cooperation?  相似文献

A major trend in foster care in developed countries over the past quarter century has been a shift toward placing children with “kin” rather than with unrelated foster parents. This change in practice is widely backed by legislation and is routinely justified as being in the best interests of the child. It is tempting to interpret this change as indicating that the child welfare profession has belatedly discovered that human social sentiments are nepotistic in their design, such that kin tend to be the most nurturant alloparents. Arguably, however, the change in practice has been driven by demographic, economic, and political forces rather than by discovery of its benefits. More and better research is needed before we can be sure that children have actually benefitted.  相似文献
