Biodiversity is declining from unprecedented land conversions that replace diverse, low-intensity agriculture with vast expanses under homogeneous, intensive production. Despite documented losses of species richness, consequences for β-diversity, changes in community composition between sites, are largely unknown, especially in the tropics. Using a 10-year data set on Costa Rican birds, we find that low-intensity agriculture sustained β-diversity across large scales on a par with forest. In high-intensity agriculture, low local (α) diversity inflated β-diversity as a statistical artefact. Therefore, at small spatial scales, intensive agriculture appeared to retain β-diversity. Unlike in forest or low-intensity systems, however, high-intensity agriculture also homogenised vegetation structure over large distances, thereby decoupling the fundamental ecological pattern of bird communities changing with geographical distance. This ~40% decline in species turnover indicates a significant decline in β-diversity at large spatial scales. Macrocyclic pyrrolobenzodiazepine dimers were designed and evaluated for use as antibody-drug conjugate payloads. Initial structure–activity exploration established that macrocyclization could increase the potency of PBD dimers compared with non-macrocyclic analogs. Further optimization overcame activity-limiting solubility issues, leading to compounds with highly potent (picomolar) activity against several cancer cell lines. Desensitization induced by chronic nicotine exposure has been hypothesized to trigger the up-regulation of the alpha4beta2 neuronal nicotinic acetylcholine receptor (nAChR) in the central nervous system. We studied the effect of acute and chronic nicotine exposure on the desensitization and up-regulation of different alpha4beta2 subunit ratios (1alpha:4beta, 2alpha:3beta, and 4alpha:1beta) expressed in Xenopus oocytes. The presence of alpha4 subunit in the oocyte plasmatic membrane increased linearly with the amount of alpha4 mRNA injected. nAChR function and expression were assessed during acute and after chronic nicotine exposure using a two-electrode voltage clamp and whole-mount immunofluorescence assay along with confocal imaging for the detection of the alpha4 subunit. The 2alpha4:3beta2 subunit ratio displayed the highest ACh sensitivity. Nicotine dose-response curves for the 1alpha4:4beta2 and 2alpha4:3beta2 subunit ratios displayed a biphasic behavior at concentrations ranging from 0.1 to 300 microm. A biphasic curve for 4alpha4:1beta2 was obtained at nicotine concentrations higher than 300 microm. The 1alpha4:4beta2 subunit ratio exhibited the lowest ACh- and nicotine-induced macroscopic current, whereas 4alpha4:1beta2 presented the largest currents at all agonist concentrations tested. Desensitization by acute nicotine exposure was more evident as the ratio of beta2:alpha4 subunits increased. All three alpha4beta2 subunit ratios displayed a reduced state of activation after chronic nicotine exposure. Chronic nicotine-induced up-regulation was obvious only for the 2alpha4: 3beta2 subunit ratio. Our data suggest that the subunit ratio of alpha4beta2 determines the functional state of activation, desensitization, and up-regulation of this neuronal nAChR. Antisera were raised to a 70-kD (kilodalton) soybean (Glycine max) protein encoded by a 2,4-dichlorophenoxyacetic acid (2,4-D) inducible mRNA, GH3. These antisera have been used to probe protein blots to study the kinetics and specificity of the GH3 induction response as well as the species specificity and intracellular location of the protein. Detectable levels of the GH3 protein are induced by 2,4-D within 2 h in elongating hypocotyl sections, root sections, and etiolated plumules, and within 30–60 min in soybean suspension cells. Synthesis of the GH3 protein is induced by a variety of auxins. Other plant hormones such as gibberellic acid, cytokinin and ethylene added with or whithout 2,4-D do not alter the level of GH3 protein induction. The GH3 protein is found only in the S100 fraction and is not associated with the nucleus or cell wall. This antiserum also reacts with a 2,4-D-inducible 70-kD protein in other dicots. 相似文献

The boll weevil, Anthonomus grandis, entered the United States of America in the early twentieth century and became a major pest in cotton, Gossypium spp. Shortly after the passage of Tropical Storm Erin on 16 August 2007 through the South Texas/Winter Garden boll weevil eradication zone, over 150 boll weevils were captured in the Southern Rolling Plains (SRP) eradication zone that was essentially weevil-free since 2003. Pollen analyses were made of the SRP weevils and weevils collected in two suspected source zones, Cameron (Southern Blacklands eradication zone) and Uvalde (Winter Garden eradication zone). An additional examination of the palynological evidence and examination of additional pollen residue shed new light on this event and strengthens the conclusion that the Uvalde area was the source of the SRP weevils. A total of 192 pollen grains from 39 taxa were found in the SRP weevils: 1904 pollen grains from ten taxa from the Cameron weevils and 148 grains from 28 taxa in the Uvalde weevils. The SRP weevils shared 16 taxa, including Phermeranthus sp. (flameflower) with the Uvalde weevils and only five taxa with the Cameron weevils. Common taxa between SRP and Uvalde weevils and the lack of the dominant ‘low spine’ Asteraceae that occurred in all Cameron samples confirm that the SRP weevils originated from the South Texas/Winter Garden zone. Problems associated with this type of research are similar to those in forensic palynology. These problems include the unknown origin of the weevils, pollen contamination and care and storage of the samples. 相似文献
