Lehman Caves is an extensively decorated high desert cave that represents one of the main tourist attractions in Great Basin National Park, Nevada. Although traditionally considered a water table cave, recent studies identified abundant speleogenetic features consistent with a hypogenic and, potentially, sulfuric acid origin. Here, we characterized white mineral deposits in the Gypsum Annex (GA) passage to determine whether these secondary deposits represent biogenic minerals formed during sulfuric acid corrosion and explored microbial communities associated with these and other mineral deposits throughout the cave. Powder X-ray diffraction (pXRD), scanning electron microscopy with electron dispersive spectroscopy (SEM-EDS), and electron microprobe analyses (EPMA) showed that, while most white mineral deposits from the GA contain gypsum, they also contain abundant calcite, silica, and other phases. Gypsum and carbonate-associated sulfate isotopic values of these deposits are variable, with δ34SV-CDT between +9.7‰ and +26.1‰, and do not reflect depleted values typically associated with replacement gypsum formed during sulfuric acid speleogenesis. Petrographic observations show that the sulfates likely co-precipitated with carbonate and SiO2 phases. Taken together, these data suggest that the deposits resulted from later-stage meteoric events and not during an initial episode of sulfuric acid speleogenesis. Most sedimentary and mineral deposits in Lehman Caves have very low microbial biomass, with the exception of select areas along the main tour route that have been impacted by tourist traffic. High-throughput 16S rRNA gene amplicon sequencing showed that microbial communities in GA sediments are distinct from those in other parts of the cave. The microbial communities that inhabit these oligotrophic secondary mineral deposits include OTUs related to known ammonia-oxidizing Nitrosococcales and Thaumarchaeota, as well as common soil taxa such as Acidobacteriota and Proteobacteria. This study reveals microbial and mineralogical diversity in a previously understudied cave and expands our understanding of the geomicrobiology of desert hypogene cave systems. 相似文献

The mouse monoclonal antibody HNK-1 and the human monoclonal IgM antibody present in patients with polyneuropathy both recognize carbohydrate epitope(s) on human myelin-associated glycoprotein and P0. In the present study, the oligosaccharide structures that bear the antibody epitope(s) were investigated. The extracellular derivative of myelin-associated glycoprotein (dMAG) was purified by immunoaffinity chromatography. P0 was electroeluted from gel slices. Western blot analysis of whole glycoproteins demonstrated that the epitopes for HNK-1 and the human monoclonal IgM antibody were different. The glycopeptides obtained by proteolysis of purified dMAG and P0 were separated and characterized by affinity chromatography on concanavalin A-Sepharose. Both dMAG and P0 displayed heterogeneity in their oligosaccharide structures, i.e., they both contained mainly tri- and tetraantennary oligosaccharides (approximately 80%), although biantennary (10%) and high-mannose and/or hybrid (10%) oligosaccharides were present. The human monoclonal IgM antibody epitope was present on all types of isolated oligosaccharide structures from either dMAG and P0. The HNK-1 epitope was present on all types of oligosaccharide structures of dMAG, whereas it was present only on tri- and tetraantennary structures of P0. 相似文献

We examined the effects of exercise intensity and menstrual cycle phase on glucose flux rates during rest and exercise in rested and fed (3-h postabsorptive) women. Eight moderately active, eumenorrheic women were studied under conditions of rest (90 min) and exercise (60 min, leg ergometer cycling at 45 and 65% peak oxygen consumption) during follicular and luteal phases. In both menstrual phases, an effect of exercise intensity was evident with glucose rates of appearance and disappearance and metabolic clearance rates: rest < 45% intensity < 65% intensity (P < 0.05). In addition, we observed no significant effect of menstrual phase on glucose rates of appearance and disappearance and metabolic clearance rate during rest or exercise at either intensity. These results are interpreted to mean that in women fed several hours before study 1) glucose flux is directly related to exercise intensity, 2) menstrual cycle phase does not alter glucose flux during rest and exercise, and 3) the subtle effects of endogenous ovarian hormones on glucose kinetics are subordinate to the much larger effects of exercise and recent carbohydrate nutrition. 相似文献

fishes, with both Antarctic (Trematomus bernacchii, T. pennellii, Pagothenia borchgrevinki) and non-Antarctic (Notothenia angustata, Bovichtus variegatus) distributions, as well as non-notothenioid fish from the Antarctic (Lycodichthys dearborni, Family Zoarcidae) and New Zealand (Bellapiscis medius, Family Tripterygiidae), in an effort to better understand the effect that inhabiting a sub-zero environment has on maintaining
the integrity of the cellular protein pool. Overall, levels of Ub-conjugated proteins in cold-adapted Antarctic fishes were
significantly higher than New Zealand fishes in gill, liver, heart and spleen tissues suggesting that life at sub-zero temperatures

The Andes of South America contain one of the richest avifaunas in the world, but little is known about how this diversity arises and is maintained. Variation in mitochondrial DNA and morphology within the speckled hummingbird (Adelomyia melanogenys) was used to elucidate the phylogeographic pattern along an Ecuadorian elevational gradient, from the coastal cordillera to the inland Andean montane region. We examined sequence, climatic/remote sensing and morphological data to understand the effects of topography and ecology on patterns of variation. Populations on either side of the Andes are genetically divergent and were separated during a period that corresponds to the final stages of Andean uplift during the Pliocene. Despite isolation, these two populations were found to be morphologically similar suggesting a strong effect of stabilizing selection across ecologically similar Andean cloud forests, as assessed using climatic and remote sensing data. In contrast, little genetic divergence was found between coastal and west-Andean individuals, suggesting recent interruption of gene flow between these localities. However, coastal populations were found to inhabit different habitats compared to Andean populations as shown by climatic and remote sensing variables. Furthermore, coastal individuals had significantly longer bills compared to their montane relatives, indicative of differential directional selection and the influence of habitat differences in shaping phenotypic variation. Results highlight the role of both isolation and ecology in diversification in Ecuadorian montane regions, while suggesting the two may not always act in concert to produce divergence in adaptive traits. 相似文献
