Accurate identification of human‐induced injuries that lead to death or interfere with reproduction is important for marine mammal management, as deaths exceeding established limits can lead to restrictions on fisheries or vessel operations. The fates of cetaceans last seen swimming with attached gear, particularly in pelagic fisheries, or with vessel strike lacerations, have been difficult to predict. Survival and reproduction data from long‐term research on resident common bottlenose dolphins near Sarasota, Florida were examined relative to consequences of fishing gear ingestion, line entanglements, vessel strikes, and amputations of unknown origins. Fishing hooks embedded in the throat, goosebeak, or esophagus, or line wrapped around the goosebeak, generally lead to death. Multiple, constrictive line wraps around fin insertions can lead to amputation, blood loss, impaired mobility, or infection. Dolphins with ingested gear or severe entanglements may swim away with the gear, but likely die later. Propeller injuries involving only soft tissue were often survivable. Some dolphins survived amputations of the distal ends of fins, and continued to reproduce. As a precautionary approach, dolphins with ingested gear or severe constrictive entanglements should be considered mortalities, but extrapolations of findings from coastal bottlenose dolphins to other cetaceans and different gear must be done with caution.  相似文献

Phosphorylation of the nicotinic acetylcholine receptor (nAChR) is believed to play a critical role in its nicotine-induced desensitization and up-regulation. We examined the contribution of a consensus PKC site in the α4 M3/M4 intracellular loop (α4S336) on the desensitization and up-regulation of α4β2 nAChRs expressed in oocytes. Position α4S336 was replaced with either alanine to abolish potential phosphorylation at this site or with aspartic acid to mimic phosphorylation at this same site. Mutations α4S336A and α4S336D displayed a threefold increase in the ACh-induced response and an increase in ACh EC50. Epibatidine binding revealed a three and sevenfold increase in surface expression for the α4S336A and α4S336D mutations, respectively, relative to wild-type, therefore, both mutations enhanced expression of the α4β2 nAChR. Interestingly, the EC50’s and peak currents for nicotine activation remained unaffected in both mutants. Both mutations abolished the nicotine-induced up-regulation that is normally observed in the wild-type. The present data suggest that adding or removing a negative charge at this phosphorylation site cannot be explained by a simple straightforward on-and-off mechanism; rather a more complex mechanism(s) may govern the functional expression of the α4β2 nAChR. Along the same line, our data support the idea that phosphorylation at multiple consensus sites in the α4 subunit could play a remarkable role on the regulation of the functional expression of the α4β2 nAChR.  相似文献

Escherichia coli is a rod-shaped bacterium that divides at its midplane, partitioning its cellular material into two roughly equal parts. At the appropriate time, a septum forms, creating the two daughter cells. Septum formation starts with the appearance of a ring of FtsZ proteins on the cell membrane at midplane. This Z-ring causes an invagination in the membrane, which is followed by growth of two new endcaps for the daughter cells. Invagination occurs against a cell overpressure of several atmospheres. A model is presented for the shape of the cell as determined by the tension in the Z-ring. This model allows the calculation of the force required for invagination. Then three possible models to generate the force necessary to achieve invagination are presented and analyzed. These models are based on converting GTP-bound FtsZ polymeric structures to GDP-bound FtsZ structures, which then leave the polymer. Each model is able to generate the force by relating the hydrolyzation to an irreversible molecular binding event, resulting in a net motion of putative anchors for the structures. All three models show that cross-linking the FtsZ protofilaments into a polymer structure allows the removal of GDP-FtsZ without interrupting the structure during force generation, as would happen for a simple polymeric chain. In this long-term study, we examined the invasion by the exotic shrub glossy buckthorn (Rhamnus frangula L.) and the response of co-occurring plants in a large, undisturbed wetland. We first sampled the vegetation in 1991 and repeated the sample 15 years later using the same, permanently located sample units (n = 165). Despite dramatic increases in the abundance of buckthorn, the invasion elicited little apparent response by the resident plant community. Species richness and cover in the herbaceous plant stratum had no apparent relationship with change in buckthorn cover. The number of shrub species other than buckthorn showed no relationship with change in buckthorn cover, but the cover of other shrubs decreased as buckthorn cover increased. Species composition changed independently of changes in buckthorn cover. These results show that dramatic increases in the abundance of an invasive species do not necessarily cause large changes in the native plant community and suggest disturbance history influences community response to invasion.  相似文献

Herbivores are sensitive to the genetic structure of plant populations, as genetics underlies plant phenotype and host quality. Polyploidy is a widespread feature of angiosperm genomes, yet few studies have examined how polyploidy influences herbivores. Introduction to new ranges, with consequent changes in selective regimes, can lead to evolution of changes in plant defensive characteristics and also affect herbivores. Here, we examine how insect herbivores respond to polyploidy in Solidago gigantea, using plants derived from both the native range (USA) and introduced range (Europe). S. gigantea has three cytotypes in the US, with two of these present in Europe. We performed bioassays with generalist (Spodoptera exigua) and specialist (Trirhabda virgata) leaf-feeding insects. Insects were reared on detached leaves (Spodoptera) or potted host plants (Trirhabda) and mortality and mass were measured. Trirhabda larvae showed little variation in survival or pupal mass attributable to either cytotype or plant origin. Spodoptera larvae were more sensitive to both cytotype and plant origin: they grew best on European tetraploids and poorly on US diploids (high mortality) and US tetraploids (low larval mass). These results show that both cytotype and plant origin influence insect herbivores, but that generalist and specialist insects may respond differently.Key words: polyploidy, cytotype, Solidago gigantea, insect herbivore, herbivory, invasive plant, introduced plantPolyploidy, or the possession of more than two sets of homologous chromosomes, is a fundamental force in angiosperm evolution.1,2 Many plant species or species complexes consist of multiple cytotypes that may occur sympatrically;3 this is an important source of genetic structure in plant populations that is often overlooked.4 Possession of multiple genomes may confer advantages to polyploid plants such as increased heterozygosity, a decreased probability of inbreeding depression, or a greater gene pool available for selection; these traits contribute to the widespread success of polyploids and may make them prone to invasiveness.5,6 In a recent article,7 we examined the functional consequences of polyploidy for different cytotypes of Solidago gigantea Ait. (Asteraceae), collected from both its native range (North America) and its introduced range (Europe). In this addendum, we show how cytotype and continent of origin influence interactions of S. gigantea with insect herbivores. Interactions with herbivores are expected to vary with cytotype because of phenotypic changes associated with polyploidy, but this area has received little study (reviewed in refs. 811). Plant origin, from either the native range or an introduced range, should also influence herbivores. Plants may escape from their specialist natural enemies in the introduced range, thereby experiencing reduced herbivore pressure from an insect community dominated by generalists.12,13 Given sufficient time, plants from the introduced range may evolve to decrease investment in anti-herbivore defenses, particularly those effective against specialists.14 While a growing body of research has addressed whether plant defenses against herbivory are lower in the introduced range,12,15,16 few of these studies have also examined the influence of cytotype.17Three cytotypes of S. gigantea can be found in its native range in North America (diploid, tetraploid and hexaploid, 2n = 18, 36 and 54 respectively). These are morphologically indistinguishable and not generally treated as separate species.18 In Europe, where S. gigantea was introduced in the mid 18th century,19 tetraploids are the dominant cytotype but diploids also occur. S. gigantea supports a diverse array of insect herbivores in its native range, but has few natural enemies in its introduced range.20 We report here on experiments using both a generalist and a specialist leaf-chewing insect. The generalist, Spodoptera exigua (Lepidoptera: Noctuidae) is widely distributed and highly polyphagous, while the specialist Trirhabda virgata (Coleoptera: Chrysomelidae) feeds only on closely-related species within the genus Solidago. T. virgata is an outbreak insect that can be a major defoliator of S. gigantea and related species in North America.21 We grew plants originating from 10 populations in the US and 20 populations in Europe in common gardens at the University of Wisconsin-Milwaukee Field Station in Saukville, Wisconsin. There were five plant origin-cytotype combinations: three cytotypes from the US and two from Europe. Insects were reared on detached leaves from a single plant (Spodoptera) or on potted host plants (Trirhabda), for a set period of 21 d (Spodoptera) or until pupation (Trirhabda). We recorded insect survival and mass at the end of 21 d (Spodoptera) or at pupation (Trirhabda) (reviewed in ref. 22).Overall survival was much better for the specialist Trirhabda than for the generalist Spodoptera (91% vs. 72%). Spodoptera larvae are not generally found on S. gigantea in the field, and while they are able to complete development, we found that this plant was not an ideal host. Spodoptera larvae were more sensitive to differences among cytotype and plant origin than were Trirhabda larvae. Percent survival was particularly poor for Spodoptera larvae reared on diploids from the US, where slightly more than half of the caterpillars survived for 21 days (Fig. 1). Trirhabda pupal mass was remarkably consistent across the five ploidy-plant origin combinations. In contrast, Spodoptera larvae responded to both cytotype and continent of origin. Surviving Spodoptera larvae did particularly well on tetraploid plants from the introduced range (Europe), and particularly poorly on tetraploids from the US (Fig. 1). We have previously reported that Spodoptera grow better on plants from Europe;22 our current results reveal that this difference is due exclusively to better growth on tetraploid plants. However, our results also show that both diploids and tetraploids from the US were poor hosts for Spodoptera: diploids because they caused high mortality and tetraploids because they resulted in poor growth. Sample sizes for each cytotype-origin combination vary because cytotypes were not known at the time plants were collected; these distributions represent frequencies of cytotypes in our collections.Effects of the host plant on Spodoptera were probably driven, at least in part, by changes in secondary chemistry. We have previously shown that foliar terpenoids, chemicals known to influence insect herbivores,23,24 are affected by both cytotype and continent of origin.7 It is surprising that Trirhabda larvae were not more sensitive to these differences in secondary chemistry among the five ploidy-origin combinations, given that Trirhabda is known to respond to host-plant chemistry.23 We have previously reported that Trirhabda growth does not differ on European and US plants22 and show here that accounting for cytotype does not change this conclusion. In a recent study on the closely-related Solidago altissima, Halverson et al.11 reported that the effects of plant cytotype on 5 gall-making herbivores were complex and not easily characterized. All five herbivores responded to plant cytotype, but for four of the five insects the most preferred cytotype was not consistent across sites. It is possible in our study that Trirhabda were responding to cytotype at a finer scale than that examined here. There may be differences due to cytotype that shift among the populations that we sampled, and that are averaged out when examined at the continental scale. We lack sufficient replication of cytotypes within populations to test this possibility. Even so, our results reported here reveal that plant cytotype can be an important source of variation affecting insect herbivores, but that generalist and specialist insects may respond differently.  相似文献

Wildfire promotes dominance of invasive giant reed (Arundo donax) in riparian ecosystems

Widespread invasion of riparian ecosystems by the large bamboo-like grass Arundo donax L. has altered community structure and ecological function of streams in California. This study evaluated the influence of wildfire on A. donax invasion by investigating its relative rate of reestablishment versus native riparian species after wildfire burned 300 ha of riparian woodlands along the Santa Clara River in southern California in October 2003. Post-fire A. donax growth rates and productivity were compared to those of native woody riparian species in plots established before and after the fire. Arundo donax resprouted within days after the fire and exhibited higher growth rates and productivity compared to native riparian plants. Arundo donax grew 3–4 times faster than native woody riparian plants—up to a mean of 2.62 cm day−1—and reached up to 2.3 m in height less than 3 months after the fire. One year post-fire, A. donax density was nearly 20 times higher and productivity was 14–24 times higher than for native woody species. Three mechanisms—fire-adapted phenology, high growth rate, and growth response to nutrient enrichment—appear to promote the preemption of native woody riparian species by A. donax after fire. This greater dominance of A. donax after wildfire increased the susceptibility of riparian woodlands along the Santa Clara River to subsequent fire, potentially creating an invasive plant-fire regime cycle. Moreover, A. donax infestations appear to have allowed the wildfire to cross the broad bed of the Santa Clara River from the north, allowing thousands of acres of shrubland to the south to burn.  相似文献

Coronavirus particles are enveloped and pleomorphic and are thus refractory to crystallization and symmetry-assisted reconstruction. A novel methodology of single-particle image analysis was applied to selected virus features to obtain a detailed model of the oligomeric state and spatial relationships among viral structural proteins. Two-dimensional images of the S, M, and N structural proteins of severe acute respiratory syndrome coronavirus and two other coronaviruses were refined to a resolution of approximately 4 nm. Proteins near the viral membrane were arranged in overlapping lattices surrounding a disordered core. Trimeric glycoprotein spikes were in register with four underlying ribonucleoprotein densities. However, the spikes were dispensable for ribonucleoprotein lattice formation. The ribonucleoprotein particles displayed coiled shapes when released from the viral membrane. Our results contribute to the understanding of the assembly pathway used by coronaviruses and other pleomorphic viruses and provide the first detailed view of coronavirus ultrastructure.  相似文献

Human cytomegalovirus (HCMV) pathogenesis is dependent on the hematogenous spread of the virus to host tissue. While data suggest that infected monocytes are required for viral dissemination from the blood to the host organs, infected endothelial cells are also thought to contribute to this key step in viral pathogenesis. We show here that HCMV infection of endothelial cells increased the recruitment and transendothelial migration of monocytes. Infection of endothelial cells promoted the increased surface expression of cell adhesion molecules (intercellular cell adhesion molecule 1, vascular cell adhesion molecule 1, E-selectin, and platelet endothelial cell adhesion molecule 1), which were necessary for the recruitment of na?ve monocytes to the apical surface of the endothelium and for the migration of these monocytes through the endothelial cell layer. As a mechanism to account for the increased monocyte migration, we showed that HCMV infection of endothelial cells increased the permeability of the endothelium. The cellular changes contributing to the increased permeability and increased na?ve monocyte transendothelial migration include the disruption of actin stress fiber formation and the decreased expression of lateral junction proteins (occludin and vascular endothelial cadherin). Finally, we showed that the migrating monocytes were productively infected with the virus, documenting that the virus was transferred to the migrating monocyte during passage through the lateral junctions. Together, our results provide evidence for an active role of the infected endothelium in HCMV dissemination and pathogenesis.  相似文献

DNA helicases are essential for DNA metabolism; however, at the molecular level little is known about how they assemble or function. Therefore, as a model for a eukaryotic helicase, we are analyzing T antigen (T-ag) the helicase encoded by simian virus 40. In this study, nuclear magnetic resonance (NMR) methods were used to investigate the transit of single-stranded DNA (ssDNA) through the T-ag origin-binding domain (T-ag OBD). When the residues that interact with ssDNA are viewed in terms of the structure of a hexamer of the T-ag OBD, comprised of residues 131 to 260, they indicate that ssDNA passes over one face of the T-ag OBD and then transits through a gap in the open ring structure. The NMR-based conclusions are supported by an analysis of previously described mutations that disrupt critical steps during the initiation of DNA replication. These and related observations are discussed in terms of the threading of DNA through T-ag hexamers and the initiation of viral DNA replication.  相似文献
