The endothelial isoform of nitric-oxide synthase (eNOS) is a key determinant of vascular tone. eNOS, a Ca(2+)/camodulin-dependent enzyme, is also regulated by a variety of agonist-activated protein kinases, but the role and regulation of the protein phosphatase pathways involved in eNOS dephosphorylation are much less well understood. Treatment of endothelial cells with vascular endothelial growth factor (VEGF), a potent eNOS agonist, leads to the activation of calcineurin, a Ca(2+)/camodulin-dependent protein phosphatase. In these studies, we used a phosphorylation state-specific antibody to show that VEGF promotes dephosphorylation of eNOS at serine residue 116 in cultured endothelial cells. Cyclosporin A, an inhibitor of calcineurin, completely blocks VEGF-induced eNOS dephosphorylation; under identical conditions, cyclosporin A also inhibits VEGF-induced eNOS activation. VEGF-induced eNOS dephosphorylation shows an EC(50) of 2 ng/ml and is maximal 30 min after agonist addition. eNOS phosphorylation at serine 116 is completely blocked by the protein kinase C inhibitor calphostin but is blocked by neither wortmannin (an inhibitor of phosphatidylinositide 3-kinase) nor the MAP kinase pathway inhibitor U0126. A phosphorylation-deficient mutant of eNOS in which serine 116 is changed to an alanine residue (S116A) shows significantly enhanced enzyme activity compared with the wild-type enzyme. Taken together, these findings indicated that VEGF-induced eNOS dephosphorylation at serine 116 leads to enzyme activation. Cyclosporin A is widely used as an immunosuppressive drug for which hypertension is an important dose-limiting side effect. Our results suggest that cyclosporin A-induced hypertension may involve, at least in part, the attenuation of endothelium-derived NO production through a calcineurin-sensitive pathway regulating eNOS dephosphorylation.  相似文献

The development of the cephalothecoid peridium of Aporothielavia leptoderma was examined using light and electron microscopy. Early stages in ascoma initiation were consistent with previous reports for other species in the Chaetomiaceae. However, as young cleistothecia increased in size, clusters of peridial cells in the outer textura angularis elongated in a radial pattern around a central cell or cell cluster to form rosettes of relatively thick-walled segments that marked the central areas of incipient cephalothecoid plates. The external flank along median portions of the radial cells became thin walled and swelled outwards so that each plate became concave and was separated from adjacent plates by a more or less circular to polygonal ridge of knuckle-shaped swellings. When dry, mature peridia split apart along some of the ridges demarcating individual plates but an internal mechanism for liberating ascospores from the confines of the ascoma was not observed. Physical disturbance of mature cleistothecia by beetles, when enclosed together in a Petri dish, shattered the peridia, liberating the ascospores. Smaller insects were unable to cause disarticulation of the cephalothecoid plates. Because of the presence of an apical germ pore in the ascospores and morphological similarity to Chaetomidium arxii, the new combination Chaetomidium leptoderma (syn. Thielavia leptoderma) comb. nov. is proposed.  相似文献

Forty crossbred barrows (Camborough 15 Line female×Canabred sire) weighing an average of 79.6±8.0?kg were used in a factorial design experiment (5 barleys×2 enzyme levels) conducted to determine the effects of phytase supplementation on nutrient digestibility in low-phytate barleys fed to finishing pigs. The pigs were assigned to one of 10 dietary treatments comprised of a normal 2-rowed, hulled variety of barley (CDC Fleet, 0.26% phytate) or 2 low-phytate hulled genotypes designated as LP422 (0.14% phytate) and LP635 (0.09% phytate). A normal, hulless barley (CDC Dawn, 0.26% phytate) and a hulless genotype designated as LP422H (0.14% phytate) were also included. All barleys were fed with and without phytase (Natuphos 5000 FTU/kg). The diets fed contained 98% barley, 0.5% vitamin premix, 0.5% trace mineral premix, 0.5% NaCl and 0.5% chromic oxide but no supplemental phosphorus. The marked feed was provided for a 7-day acclimatization period, followed by a 3-day faecal collection. In the absence of phytase, phosphorus digestibility increased substantially (P<0.05) as the level of phytate in the barley declined. For the hulled varieties, phosphorus digestibility increased from 12.9% for the normal barley (0.26% phytate) to 35.3 and 39.8% for the two low-phytate genotypes (0.14 and 0.09% phytate respectively). For the hulless varieties, phosphorus digestibility increased from 9.2% for the normal barley (0.26% phytate) to 34.7% for the hulless variety with 54% of the normal level of phytate (0.14% phytate). In contrast, when phytase was added to the diet, there was little difference in phosphorus digestibility between pigs fed normal barley and those fed the low-phytate genotypes (significant barley×enzyme interaction, P=0.01). For the hulled varieties, phosphorus digestibility was 50.1% for the barley with the normal level of phytate (0.26% phytate) compared with 51.1 and 52.4% for the varieties with 54 and 35% of the normal level of phytate (0.14 and 0.09% phytate respectively). For the hulless varieties, phosphorus digestibility increased from 47.1% for the normal barley (0.26% phytate) to 54.4% for the hulless variety with 54% of the normal level of phytate (0.14% phytate). In conclusion, both supplementation with phytase and selection for low-phytate genotypes of barley were successful in increasing the digestibility of phosphorus for pigs. Unfortunately, the effects did not appear to be additive. Whether or not swine producers will choose low-phytate barley or supplementation with phytase as a means to improve phosphorus utilization, will likely depend on the yield potential of low-phytate barley and the additional costs associated with supplementation with phytase.  相似文献

Seventeen synthetic 1-substituted 1,2,4-triazoles exerted a significant effect on the bacteriaB. subtilis, S. aureus, E. coli andP. aeruginosa. The least sensitive to the effects of the triazoles wasS. aureus. With all triazole derivatives and their combinations,B. subtilis andP. aeruginosa exhibited IC50 and MIC values several times higher than with ampicillin. The most effective triazoles have a N-phenyl ring or benzimidinoyl ring substituted with one or several chlorine atoms. The role of membrane depolarization in the regulation of expression of a neuron specific protein was evaluated by culturing superior cervical ganglia from neonatal rats in defined medium and manipulating neuronal activity by depolarizing agents. P65 is an integral membrane protein of synaptic vesicles and can be used as a marker for general neuronal maturation. P65 antigen levels were quantified by indirect radioimmunoassay, using monoclonal antibodies. The expression of p65 in ganglion explants increased by 40-100% when the cultures were treated with the depolarizing agents, veratridine or high potassium. The veratridine effect could be blocked by simultaneous treatment with the sodium channel blocker, tetrodotoxin (TTX). The rise in p65 was not evident until 36 h after depolarizing treatment had begun and reached peak levels after 48 h, with no further increases observed with sustained treatment. After removal of the depolarizing treatment, p65 levels returned to control values after 24 h. P65 joins a growing number of molecules whose expression is regulated by membrane depolarization.  相似文献

The African freshwater oyster Etheria elliptica, which is of great economic importance throughout the continent, is facing overharvesting in many fisheries in West Africa. Its reproductive traits (sex ratio, size at sexual maturity, oocyte diameter and fecundity) were studied at four stations located along the Pendjari River, northern Benin, in April 2013. Histological techniques were implemented to identify sex and gonad development stages. Oocyte sizes were measured based on the histological images and mean oocyte diameter was 38 µm. Fecundity, estimated by counting the developing eggs, averaged 106 724 eggs and increased significantly with shell size. The average sex ratio was approximately 1:1. Hermaphrodites were rare. Males reached sexual maturity at 57.6 mm dorsoventral height, earlier than females at 64.7 mm. The majority of the oyster specimens were in ripe-spawning stages, indicating that reproductive activities partly took place towards the end of the dry season. Both mature and immature individuals were subjected to harvesting at all the investigated fishing sites. The mean size of exploited oysters was 60.6 mm, lower than the size of females at maturity. Management strategies must enforce the minimum size of 65 mm required for harvesting, particularly in open access sites where juveniles are mostly harvested.  相似文献

The central part of the tin ore deposit Ehrenfriedersdorf/Erzgebirge, which was exploited from the 13th century to 1990, was flooded from 1994 to 1996. Since that time mine waters have flown through the gallery “Tiefer Sauberger Stolln” to the creek Wilisch in the Elbe river catchment area. The water at the mine portal shows high concentrations of arsenic and heavy metals. The average arsenic concentration is about 0.5 mg/L. Approximately two thirds of arsenic are transported dissolved. Where the mine water ascends from deeper levels, arsenic concentrations of about 0.4 mg/L were found. Here arsenic occurs predominantly particular. The mining gallery “Tiefer Sauberger Stolln” provides the unique opportunity of subsurface sampling for the identification of the arsenic sources under different hydrological conditions (normal and high water level). The sources of dissolved arsenic in the gallery part between the raise and the portal were determined and analyzed. Between these two monitoring points, many inflows of infiltration water were detected. The concentration of As in the infiltration water reaches up to 1.8 mg/L, which varies depending on the location in the gallery and the hydrological situation. The first part of the gallery was straightened, heightened and partly concreted with modern mining technique. The arsenic concentrations can decrease owing to high precipitation rates and snow melt events. The last part of the gallery was preserved due to low coverage. Here the arsenic concentrations in the infiltration waters increase with the surface water inflow. At a normal water level, 1 kg arsenic per day leaves the raise and 2.1 kg the gallery portal, which means that 50 % of the arsenic load comes from the infiltration water. At a high water level, 2.5 kg arsenic per day are transported through the raise and 8.2 kg per day through the gallery portal, which means that about 70 % of the arsenic load comes from infiltration water. The area of Ehrenfriedersdorf is characterized by a superposition of anthropogenic soil pollution over the geogenic inventory. There is a close connection between ancient soil contaminations by high amounts of water‐soluble arsenic compounds, e.g. arsenic trioxide formed by roasting the ores during ancient tin smelting, and high concentrations of dissolved arsenic in the infiltration water. The contamination of surface water and river sediments by arsenic is originating from an anthropogenic pollution of soils by ancient tailings via infiltration of water rich in arsenic into the mine gallery.  相似文献
