The object of this study was to investigate the changes in physical parameters produced during an in-season resistance training (RT) and detraining (DT, or RT cessation) in 16 high level team handball players (THPs). Apart from normal practice sessions, THPs underwent 12 weeks of RT. Subjects performed 3 sets of 3-6 reps with a load of 70-85% concentric 1 repetition maximum bench press (1RMBP), 3 sets of 3-6 reps with a load of 70-95% of 4 repetition maximum parallel squats (4RMPS), plus vertical jumps and sprints. The 1RMBP, 4RMPS, speed over 30 m (S30), jump (countermovement jump height [CMJ]; CMJ with additional weights [20kg and 40kg], and ball throw velocity (BTv) were tested before the experimental period (T1), after 6 weeks (T2), and after the 12-week experimental period (T3). Immediately after these 12 weeks, THPs started a 7-week DT period, maintained normal practices. The CMJ and the BTv were the only parameters evaluated during DT. The most important gains (p < 0.001) in S30 were obtained between T1-T2 and T1-T3. The BTv improved significantly (p < 0.001) only between T1-T2 and T1-T3. The most relevant increases (p < 0.001) in jumping performance took place between T1-T2 and T1-T3. The 1RMBP showed significant increases (p < 0.001) only between T1-T2 and T1-T3. The 4RMPS increased significantly between all testing trials. After the DT, THPs showed no significant losses in CMJ performance. However, they declined significantly in BTv (p = 0.023). The results suggest that elite THPs can optimize important physical parameters over 12 weeks in-season and that 7 weeks of DT, although insufficient to produce significant decreases in CMJ, are sufficient to induce significant decreases in BTv. It is concluded that after RT cessation THPs reduced BTv performance.  相似文献

Energy budget of juvenile fat snook Centropomus parallelus fed live food   总被引:1,自引:0,他引:1  
The fat snook Centropomus parallelus is a tropical estuarine species with importance for recreational and commercial fisheries and further aquaculture potential. Considering ingested energy (C) is channeled into growth (P), metabolic expenditure (R), excretion (U) and feces (F), a balanced energy budget was established for isolated and grouped juvenile C. parallelus (5.18 to 10.25 g wet mass) by experimental quantification of each of these parameters. Fish were fed live prawn (Macrobrachium sp.) at 25 degrees C and 20 per thousand for 19 days and daily energy budgets could be calculated. Energy content of food (live prawn), fish whole body and feces were 17.7, 14.5 and 6.1 KJ g(-1) dry mass (DW), respectively. Mass-specific rates of oxygen consumption and ammonia-N excretion were 0.271 and 0.0082 mg of O(2) or NH(3)-N g(-1) wet mass (WW) h(-1), respectively, resulting in O:N=23.4. Daily ingestion (C) was dependent on the amount of food offered and ranged between 4.9% to 7.4% of initial wet weight. Growth (P) was positively correlated with initial mass varying from 0.008 to 0.104 g day(-1). Feces release also correlated to fish mass and averaged 9.53 mg dry mass day(-1). The components of energy budget showed mean values of 2.39 (C), 0.24 (P), 1.96 (R), 0.11 (U) and 0.06 (F) KJ ind(-1) day(-1). As percentage of ingested energy, C. parallelus channeled 10% in growth (P), 82% in metabolism (R), 4.6% in excretion (U) and 2.5% in feces (F). Gross (K(1)) and net (K(2)) growth efficiencies were 9.2% and 9.9%, respectively. On the course of this period of juvenile development, K(1) and K(2) increased significantly. Partitioning of ingested energy in P, R, U and F was significantly different in individually and group maintained fish (P<0.05). Energy budget of C. parallelus fed a highly digestible diet (live prawn) revealed poor growth and growth efficiencies (K(1) and K(2)) possibly associated with an elevated metabolic demand and a high channeling of metabolized energy (P+R) into metabolism (R) in both isolated and grouped fish. Data can be applied to ecosystem modeling and may contribute to identify species potential to aquaculture.  相似文献

The aim of the present study was to estimate the index and individual responses to selection for milk (MY), fat (FY) and protein (PY) yields for different breeding goals for two commercial buffalo milk production systems in S?o Paulo State characterized by: 1) all milk produced is sold to the industry (MILK) and 2) all milk produced is used in the mozzarella cheese-making process at the farm (MOZZARELLA). The current payment policy is based exclusively on milk volume. The mozzarella price refers to the wholesale selling price. Index responses to selection (IR) were calculated for three different breeding goals (BG): 1) MY exclusively (BG(1)); 2) FY + PY (BG(2)) and 3) MY + FY + PY (BG(3)). IR for the MILK system were 41.79 US dollars (BG(1)), 5.91 US dollars (BG(2)) and 38.22 US dollars (BG(3)). For the MOZZARELLA system, IR were 179.50 US dollars (BG(1)), 262.85 US dollars (BG(2)) and 402.41 US dollars (BG(3)). The results suggest that for the present circumstances, selection for milk components is not advantageous when milk is produced for sale to the industry. However, when mozzarella making is added to the system, the selection for components and milk volume is the most economically beneficial.  相似文献

A soluble fructose-1,6-bisphosphate aldolase enzyme has been purified 50.2-fold (2.36%) at the homogeneity from the electric organ of Electrophorus electricus by one step of DEAE-52 anion exchange chromatography followed by Superose-12 gel filtration-FPLC. Like other aldolase enzymes the E. electricus protein is a dimer with two identical subunits of 45 kDa. The N-terminal (20 residues) revealed a high homology with S. aurata (75%, goldfish), R. ratus and M. musculus (mouse, 80%) enzymes.  相似文献

Biocatalysis constitutes an important tool in organic synthesis, especially for the preparation of chiral molecules of biological interest. A series of aliphatic and aromatic aldehydes and two ketones were reduced using plant cell preparations from Manihot esculenta and Manihot dulcis roots. The reduced products were typically obtained in excellent yields (80-96%), and with excellent enantiomeric excess (94-98%), except for vanillin. Esters, a nitrile, and an amide were also examined, but were not reduced. Preliminary conversion rate studies are reported. This is the first attempt to perform the biotransformation of carbonyl compounds using Manihot species.  相似文献

More than 100 transthyretin (TTR) variants are associated with hereditary amyloidosis. Approaches for TTR amyloidosis that interfere with any step of the cascade of events leading to fibril formation have therapeutic potential. In this study we tested (−)-epigallocatechin-3-gallate (EGCG), the most abundant catechin of green tea, as an inhibitor of TTR amyloid formation. We demonstrate that EGCG binds to TTR “in vitro” and “ex vivo” and that EGCG inhibits TTR aggregation “in vitro” and in a cell culture system. These findings together with the low toxicity of the compound raise the possibility of using EGCG in a therapeutic approach for familial amyloidotic polyneuropathy, the most frequent form of hereditary TTR amyloidosis.

Medicine Lake is a small (165 ha), relatively shallow (average 7.3 m), intermediate elevation (2,036 m) lake located within the summit caldera of Medicine Lake volcano, Siskiyou County, California, USA. Sediment cores and high-resolution bathymetric and seismic reflection data were collected from the lake during the fall of 1999 and 2000. Sediments were analyzed for diatoms, pollen, density, grain size (sand/mud ratio), total organic carbon (TOC), and micro-scale fabric analysis. Using both 14C (AMS) dating and tephrochronology, the basal sediments were estimated to have been deposited about 11,400 cal year BP, thus yielding an estimated average sedimentation rate of about 20.66 cm/1,000 year. The lowermost part of the core (11,400–10,300 cal year BP) contains the transition from glacial to interglacial conditions. From about 11,000–5,500 cal year BP, Medicine Lake consisted of two small, steep-sided lakes or one lake with two steep-sided basins connected by a shallow shelf. During this time, both the pollen (Abies/Artemisia ratio) and the diatom (Cyclotella/Navicula ratio) evidences indicate that the effective moisture increased, leading to a deeper lake. Over the past 5,500 years, the pollen record shows that effective moisture continued to increase, and the diatom record indicates fluctuations in the lake level. The change in the lake level pattern from one of the increasing depths prior to about 6,000 cal year BP to one of the variable depths may be related to changes in the morphology of the Medicine Lake caldera associated with the movement of magma and the eruption of the Medicine Lake Glass Flow about 5,120 cal year BP. These changes in basin morphology caused Medicine Lake to flood the shallow shelf which surrounds the deeper part of the lake. During this period, the Cyclotella/Navicula ratio and the percent abundance of Isoetes vary, suggesting that the level of the lake fluctuated, resulting in changes in the shelf area available for colonization by benthic diatoms and Isoetes. These fluctuations are not typical of the small number of low-elevation Holocene lake records in the region, and probably reflect the hydrologic conditions unique to Medicine Lake.  相似文献

The present work aims to gain insights into the role of peptide-lipid interactions in the mechanisms of cellular internalization and endosomal escape of the S4(13)-PV cell-penetrating peptide, which has been successfully used in our laboratory as a nucleic acid delivery system. A S4(13)-PV analogue, S4(13)-PVscr, displaying a scrambled amino acid sequence, deficient cell internalization and drug delivery inability, was used in this study for comparative purposes. Differential scanning calorimetry, fluorescence polarization and X-ray diffraction at small and wide angles techniques showed that both peptides interacted with anionic membranes composed of phosphatidylglycerol or a mixture of this lipid with phosphatidylethanolamine, increasing the lipid order, shifting the phase transition to higher temperatures and raising the correlation length between the bilayers. However, S4(13)-PVscr, in contrast to the wild-type peptide, did not promote lipid domain segregation and induced the formation of an inverted hexagonal lipid phase instead of a cubic phase in the lipid systems assayed. Electron microscopy showed that, as opposed to S4(13)-PVscr, the wild-type peptide induced the formation of a non-lamellar organization in membranes of HeLa cells. We concluded that lateral phase separation and destabilization of membrane lamellar structure without compromising membrane integrity are on the basis of the lipid-driven and receptor-independent mechanism of cell entry of S4(13)-PV peptide. Overall, our results can contribute to a better understanding of the role of peptide-lipid interactions in the mechanisms of cell-penetrating peptide membrane translocation, helping in the future design of more efficient cell-penetrating peptide-based drug delivery systems.  相似文献

Trypanosoma cruzi is a rare example of an eukaryote that has genes for two threonine proteases: HslVU complex and 20S proteasome. HslVU is an ATP-dependent protease consisting of two multimeric components: the HslU ATPase and the HslV peptidase. In this study, we expressed and obtained specific antibodies to HslU and HslV recombinant proteins and demonstrated the interaction between HslU/HslV by coimmunoprecipitation. To evaluate the intracellular distribution of HslV in T. cruzi we used an immunofluorescence assay and ultrastructural localization by transmission electron microscopy. Both techniques demonstrated that HslV was localized in the kinetoplast of epimastigotes. We also analyzed the HslV/20S proteasome co-expression in Y, Berenice 62 (Be-62) and Berenice 78 (Be-78) T. cruzi strains. Our results showed that HslV and 20S proteasome are differently expressed in these strains. To investigate whether a proteasome inhibitor could modulate HslV and proteasome expressions, epimastigotes from T. cruzi were grown in the presence of PSI, a classical proteasome inhibitor. This result showed that while the level of expression of HslV/20S proteasome is not affected in Be-78 strain, in Y and Be-62 strains the presence of PSI induced a significantly increase in Hslv/20S proteasome expression. Together, these results suggest the coexistence of the protease HslVU and 20S proteasome in T. cruzi, reinforcing the hypothesis that non-lysosomal degradation pathways have an important role in T. cruzi biology.  相似文献
