Head orientation in the sagittal plane was assessed before and the second and sixth days after a prolonged microgravity exposure (6 months) in 7 astronauts, which were instructed to execute various locomotor movements (stair ascent, jump down and level walk). Crewmembers systematically flexed down the head when they executed the locomotion the second day post-flight compared to the pre-flight walk. Cephalic orientation was normally positioned when movements was performed the sixth day after Earth re-entry. This head flexion may reflect a strategy used by cosmonauts to increase the vestibular sensibility, and then improve the control of the dynamic equilibrium during locomotor movements. The high rate of re-adaptation indicated that cosmonauts rapidly recovered their normal walking capacities despite the prolonged microgravity exposure.  相似文献

Natural selection is a central tenet of evolutionary theory, yet the estimation of the direction and intensity of selection remains problematic. Here, we assess the strength of selection on the early expression of a secondary sexual ornament, bill colour, in male European blackbirds (Turdus merula) using 5 years of capture-mark-recapture (CMR) data. The best-fitting model consisted of a quadratic relationship between survival rate and bill colour, indicating stabilizing natural selection on the early expression of a secondary sexual trait. There was no evidence for sexual selection acting on bill colour in the first year. We suggest that the consideration of early selection and the adoption of refined statistical methods may reveal patterns of selection in the wild that have, as yet, remained undetected.  相似文献

Infection of cells with poliovirus induces a massive intracellular membrane reorganization to form vesicle-like structures where viral RNA replication occurs. The mechanism of membrane remodeling remains unknown, although some observations have implicated components of the cellular secretory and/or autophagy pathways. Recently, we showed that some members of the Arf family of small GTPases, which control secretory trafficking, became membrane-bound after the synthesis of poliovirus proteins in vitro and associated with newly formed membranous RNA replication complexes in infected cells. The recruitment of Arfs to specific target membranes is mediated by a group of guanine nucleotide exchange factors (GEFs) that recycle Arf from its inactive, GDP-bound state to an active GTP-bound form. Here we show that two different viral proteins independently recruit different Arf GEFs (GBF1 and BIG1/2) to the new structures that support virus replication. Intracellular Arf-GTP levels increase approximately 4-fold during poliovirus infection. The requirement for these GEFs explains the sensitivity of virus growth to brefeldin A, which can be rescued by the overexpression of GBF1. The recruitment of Arf to membranes via specific GEFs by poliovirus proteins provides an important clue toward identifying cellular pathways utilized by the virus to form its membranous replication complex.  相似文献

A common paradigm holds that during cell-to-cell transmission, viruses behave as lone soldiers. Recently, we discovered not only that enteroviruses are transmitted via vesicles as populations of viral particles but also that this type of transmission enhances their infection efficiency (Y. H. Chen et al., Cell 160:619–630, 2015). This mechanism could be advantageous for the overall fitness of the viral population, promoting genetic interplay by enabling viral quasispecies to collectively infect a susceptible host cell. Here, we discuss these findings in the context of viral pathogenesis and also propose that this novel type of vesicular transmission is widespread among different virus families and includes populations of both viral particles and naked viral genomes.  相似文献

The independent effects of herbivores and neighbors on plants are generally negative, and therefore the combined effects of these interactions are generally assumed to have additive or multiplicative negative effects on plant growth. However, because herbivores can stimulate the growth of plants (compensation). and neighbors can facilitate each other, the combined effects of herbivory and plant-plant interactions can be highly variable and poorly predicted by current competition and plant-herbivore theory. In some cases in North America, Festuca species appear to facilitate invasive Centaurea species and enhance their compensatory responses in controlled greenhouse conditions. We explored the interactions between herbivory and neighbor effects in the French Alps by testing the effect of the neighbor, Festuca paniculata L., on the compensatory growth response of defoliated Centaurea uniflora L. over two growing seasons. Seventy percent of aboveground C. uniflora biomass was clipped at each of seven times throughout two growing seasons in the presence or absence of F. paniculata. Centaurea uniflora compensated for severe damage in the first year, but was negatively affected by defoliation in the second year. Defoliating C. uniflora reduced final aboveground biomass by 44% and flower number by 64%, but did not affect survival. Unlike observations for other Centaurea and Festuca species, F. paniculata had significant competitive effects on C. uniflora. Festuca paniculata neither enhanced compensatory responses of C. uniflora nor increased the negative effects of defoliation. Our results show that compensatory responses can weaken over time, but that neighboring plants do not necessarily increase the negative effects of defoliation.  相似文献

Ghrelin, a 28 residues acylated peptide, is the natural ligand of the growth-hormone secretagogue receptor (GHS-R), which also interacts with small synthetic peptides. We investigated the importance of each of the first 14 N-terminal residues by Ala replacement (Ala-scan) and also of the N-terminal positive charge, on the recombinant GHS-R expressed in HEK293 or CHO cells by binding, IP and Ca(2+) assays. Nearly all of the replacements had no significant effect on the ligand binding or IP(3)/Ca(2+) stimulation. Exceptions were the modification of the N-terminal residue to [A(1)]- or N(alpha)-acetyl-ghrelin (1-14), confirming the requirement for the positive charge at the amino-terminus. Mutation of [F(4)]- to [A(4)]- or [Y(4)]-ghrelin (1-14), were detrimental suggesting direct interaction with the GHS-R. [A(8)] and [Y(8)] were more potent than ghrelin (1-14), implying that the naturally occurring Glu(8) residue may not be the optimal.  相似文献
