Conclusion The ethnic Hungarians and the Romanians share a number of myths about their ancestors and about their homeland, Transylvania. I fully agree with Anthony D. Smith, who states that the core of ethnic identity is made up of a myth-symbol complex, consisting of myths, symbols, historical memory and key values. According to Smith, myths and symbols guarantee the preservation and the passing down to future generations of ethnic identity.25 The main problem with the ethnic Hungarians and the Romanians of Transylvania is that their convictions are not compatible, they are even contradictory. In the ethnic Hungarians' view, Transylvania is in fact Hungarian, but it was taken from them by Romania in 1920. According to the Romanians, Transylvania is Romanian but for centuries mistakenly considered as Hungarian by the Hungarians. The historical myths are part of the Hungarian as well as the Romanian collective consciousness. They strengthen cohesion within these groups but, as the convictions are incompatible, they widen the gap between them. In my view, these contradictory views are an additional cause of the current mutual oversensitivity, the mutual suspicion and mutual ignorance which characterize the relation between the two ethnic groups. I think that the success of the Romanian nationalist parties such as the PUNR and România Mare 26 among the Romanian villagers of Transylvania 27is partly due to their exploitation of these contradictory myths.However, the differing oral traditions in the villages do not cause open conflicts. That everyone in his own group passes on his own version helps to explain this. In mixed villages like Dumbrava and Mànàstireni, the sensitive issues are not discussed in ethnically mixed groups. Virtually everything to do with history is suppressed in the day-to-day life of the village. Nevertheless, that does not stop one ethnic group from gossiping about the other. Thus, mutual distrust persists. Such a sense of fear is a dangerous breeding-ground because it can easily be exploited by nationalist parties. When anything occurs in politics which appeals to these feelings of fear and distrust, two fronts are lined up in no time. This actually happened as a result of the controversy about the archaeological excavations in Cluj.Fortunately, there is a sufficiently large number of people who realize that this is pointless and who see through the extremist nationalist party leaders' malice. Bearing in mind the tragic example of the former Yugoslavia, they advocate understanding and peaceful interethnic co-existence.I have touched on only one of several conditions for ethnic survival and revival: the shared historical myths. The western flower thrips [Frankliniella occidentalis (Pergande) (Thysanoptera: Thripidae)] is a major pest in pepper cultivation. Therefore, host plant resistance to thrips is a desirable trait. The objectives of this study were to determine the effect of resistance on the development of thrips and to identify metabolite compounds related to the resistance. Three highly resistant, three medium resistant, and three susceptible pepper accessions were used in this study. Adult and pre‐adult survival, developmental time, and oviposition rate were assessed. Gas chromatography‐mass spectrometry was used to identify compounds that correlate with the level of resistance to thrips. Our results show that resistance of pepper accessions has a significant effect on oviposition rate and larval mortality. Seven compounds were identified that correlate with resistance to thrips and six compounds were identified that correlate with susceptibility to thrips. Some of these compounds, such as tocopherols, were previously shown to have an effect on insects in general. Also, some specific secondary metabolites (alkanes) seem to be more abundant in susceptible accessions and were induced by thrips infestation.  相似文献

Paget’s disease of bone (PDB) is one of the most frequent metabolic bone disorders (1–5%), next to osteoporosis, affecting individuals above age 55. Sequestosome1 mutations explain a part of the PDB patients, but still the disease pathogenesis in the remaining PDB patients is largely unknown. Therefore, association studies investigating the relationship between genetic polymorphisms and sporadic PDB have been performed to find the genetic risk variants. Previously such studies indicated a role of the OPG and RANK gene. The latter was recently confirmed in a genome-wide association study (GWAS) which also indicated the involvement of chromosomal regions harbouring the CSF1 and OPTN gene. In this study, we sought to replicate these findings in a Belgian and a Dutch population. Similar significant results were obtained for the single nucleotide polymorphisms and the haplotypes. The most significant results are found in the CSF1 gene region, followed by the OPTN and TNFRSF11A gene region (p values ranging from 1.3 × 10?4 to 3.8 × 10?8, OR = 1.523–1.858). We next obtained significant association with a polymorphism from the chromosomal region around the TM7SF4 gene (p = 2.7 × 10?3, OR = 1.427), encoding DC-STAMP which did not reach genome-wide significance in the GWAS, but based on its function in osteoclasts it can be considered a strong candidate gene. After meta-analysis with the GWAS data, p values ranged between 2.6 × 10?4 and 8.8 × 10?32. The calculated cumulative population attributable risk of these four loci turned out to be about 67% in our two populations, indicating that most of the genetic risk for PDB is coming from genetic variants close to these four genes.  相似文献

Regression tree analysis, a non-parametric method, was undertaken to identify predictors of the serum concentration of polychlorinated biphenyls (sum of marker PCB 1 ABBREVIATIONS: BMI: body-mass index, CV: cross validation, ln: natural logarithm, ns: not significant, PCAHs: polychlorinated aromatic hydrocarbons, PCBs: polychlorinated biphenyls, R2 a: adjusted coefficient of determination, VIF: variance inflation factor. View all notes 138, 153, and 180) in humans. This method was applied on biomonitoring data of the Flemish Environment and Health study (2002–2006) and included 1679 adolescents and 1583 adults. Potential predictor variables were collected via a self-administered questionnaire, assessing information on lifestyle, food intake, use of tobacco and alcohol, residence history, health, education, hobbies, and occupation. Relevant predictors of human PCB exposure were identified with regression tree analysis using ln-transformed sum of PCBs, separately in adolescents and adults. The obtained results were compared with those from a standard linear regression approach. The results of the non-parametric analysis confirm the selection of the covariates in the multiple regression models. In both analyses, blood fat, gender, age, body-mass index (BMI) or change in bodyweight, former breast-feeding, and a number of nutritional factors were identified as statistically significant predictors in the serum PCB concentration, either in adolescents, in adults or in both. Regression trees can be used as an explorative analysis in combination with multiple linear regression models, where relationships between the determinants and the biomarkers can be quantified.  相似文献

Background

Over the last decade several phase III microbicides trials have been conducted in developing countries. However, laboratories in resource constrained settings do not always have the experience, infrastructure, and the capacity to deliver laboratory data meeting the high standards of clinical trials. This paper describes the design and outcomes of a laboratory quality assurance program which was implemented during a phase III clinical trial evaluating the efficacy of the candidate microbicide Cellulose Sulfate 6% (CS) [1].

Methodology

In order to assess the effectiveness of CS for HIV and STI prevention, a phase III clinical trial was conducted in 5 sites: 3 in Africa and 2 in India. The trial sponsor identified an International Central Reference Laboratory (ICRL), responsible for the design and management of a quality assurance program, which would guarantee the reliability of laboratory data. The ICRL provided advice on the tests, assessed local laboratories, organized trainings, conducted supervision visits, performed re-tests, and prepared control panels. Local laboratories were provided with control panels for HIV rapid tests and Chlamydia trachomatis/Neisseria gonorrhoeae (CT/NG) amplification technique. Aliquots from respective control panels were tested by local laboratories and were compared with results obtained at the ICRL.

Results

Overall, good results were observed. However, discordances between the ICRL and site laboratories were identified for HIV and CT/NG results. One particular site experienced difficulties with HIV rapid testing shortly after study initiation. At all sites, DNA contamination was identified as a cause of invalid CT/NG results. Both problems were timely detected and solved. Through immediate feedback, guidance and repeated training of laboratory staff, additional inaccuracies were prevented.

Conclusions

Quality control guidelines when applied in field laboratories ensured the reliability and validity of final study data. It is essential that sponsors provide adequate resources for implementation of such comprehensive technical assessment and monitoring systems.

We have isolated and sequenced a cDNA clone that, as judged by the sequence of the homeobox region, encodes a sea urchin homolog of the homeobox containing the gene Abdominal-B of Drosophila. The total length of the cDNA is 3634 nucleotides and includes an open reading frame, which encodes a protein that is 32,321 Da. The N-terminal region of the homeodomain includes consensus sequences found in some of TgHBox4's Abdominal-B relatives. A genomic clone representing the 5' part of the message was also isolated. This clone and a previously isolated clone were found to represent the full-length cDNA sequence. We have also raised antibodies against a bacterially expressed portion of the TgHBox4 protein and used them to determine the location of TgHBox4 proteins during development. The protein displays ubiquitous expression early in development but becomes more restricted, to posterior regions, late in embryogenesis. Thus, in contrast to its Abd-B homologs in bilateral metazoans, TgHBox4 is probably not involved in pattern formation but may have a posterior-defining role late in embryogenesis.  相似文献

Background

The transition to retirement introduces a decline in total physical activity and an increase in TV viewing time. Nonetheless, as more time becomes available, early retirement is an ideal stage to implement health interventions. Therefore, knowledge on specific determinants of physical activity and sedentary time is needed. Former work-related physical activity has been proposed as a potential determinant, but concrete evidence is lacking. The aim of this study was to examine if former work-related sitting, standing, walking or vigorous activities predict physical activity and sedentary time during early retirement. Additionally, moderating effects of educational level and physical functioning were examined.

Methods

In total, 392 recently retired Belgian adults (>6 months, <5 years) completed the International Physical Activity Questionnaire, the SF-36 Health Survey and a questionnaire on sociodemographics and former work-related activities. Generalized linear regression analyses were conducted in R. Moderating effects were examined by adding cross-products to the models.

Results

More former work-related sitting was predictive of more screen time during retirement. Lower levels of former work-related vigorous activities and higher levels of former work-related walking were associated with respectively more cycling for transport and more walking for transport during retirement. None of the predictors significantly explained passive transportation, cycling and walking for recreation, and leisure-time moderate-to-vigorous physical activity during retirement. Several moderating effects were found, but the direction of the interactions was not univocal.

Conclusions

Former-work related behaviors are of limited importance to explain physical activity during early retirement, so future studies should focus on other individual, social and environmental determinants. Nonetheless, adults who previously had a sedentary job had higher levels of screen time during retirement, so this is an important subgroup to focus on during interventions. Because of the inconsistent moderating effects of educational level and physical functioning, no clear recommendations can be formulated.  相似文献

To progress the fields of tissue engineering (TE) and regenerative medicine, development of quantitative methods for non-invasive three dimensional characterization of engineered constructs (i.e. cells/tissue combined with scaffolds) becomes essential. In this study, we have defined the most optimal staining conditions for contrast-enhanced nanofocus computed tomography for three dimensional visualization and quantitative analysis of in vitro engineered neo-tissue (i.e. extracellular matrix containing cells) in perfusion bioreactor-developed Ti6Al4V constructs. A fractional factorial ‘design of experiments’ approach was used to elucidate the influence of the staining time and concentration of two contrast agents (Hexabrix and phosphotungstic acid) and the neo-tissue volume on the image contrast and dataset quality. Additionally, the neo-tissue shrinkage that was induced by phosphotungstic acid staining was quantified to determine the operating window within which this contrast agent can be accurately applied. For Hexabrix the staining concentration was the main parameter influencing image contrast and dataset quality. Using phosphotungstic acid the staining concentration had a significant influence on the image contrast while both staining concentration and neo-tissue volume had an influence on the dataset quality. The use of high concentrations of phosphotungstic acid did however introduce significant shrinkage of the neo-tissue indicating that, despite sub-optimal image contrast, low concentrations of this staining agent should be used to enable quantitative analysis. To conclude, design of experiments allowed us to define the most optimal staining conditions for contrast-enhanced nanofocus computed tomography to be used as a routine screening tool of neo-tissue formation in Ti6Al4V constructs, transforming it into a robust three dimensional quality control methodology.  相似文献
