The methodology for determination of the minimally required aerobic sludge retention time (SRTminaer) in biological phosphorus removal (BPR) systems is presented in this article. Contrary to normal biological conversions, the BPR process is not limited by a SRTmin resulting from the maximum growth rate of the organisms. This is because the aerobic SRT should be long enough to oxidize the amount of poly-hydroxy-alkanoates (PHA) stored in the anaerobic phase. This means that the SRTminaer will primarily depend on the PHA conversion kinetics and the maximal achievable PHA content in the cell (storage capacity). The model for the prediction of the minimally required aerobic SRT as a function of kinetic and process parameters was developed and compared with experimental data used to evaluate several operational aspects of BPR in a sequencing batch reactor (SBR) system. Rattlesnakes (Crotalus unicolor, C vegrandis) and Russell's vipers (Vipera russelli) at San Diego Zoo exhibited sustained high rates of tongue flicking after striking mice (Mus musculus) but not after seeing, smelling, and/or detecting thermal cues arising from mice. Called strike-induced chemosensory searching (SICS), this phenomenon contributes to poststrike trailing of envenomated prey. Because these zoo-raised snakes had always been offered dead rodents, and because these prey were usually ingested without first being struck, the present snakes had rarely exercised their innate predatory repertoire (ie, ambush tactics including striking, releasing, and trailing). Indeed, most specimens had never before struck a mouse and, hence, had never exhibited SICS. The occurrence of SICS in the present study clearly indicates that this important aspect of the predatory repertoire had not been degraded as a consequence of long-term captive husbandry. 相似文献

The objective of this study was to develop a rapid and efficient means of sexing murine preimplantation embryos at the 4- to 8-cell stage of development. To achieve this goal, a nested, multiplex polymerase chain reaction (PCR) was optimized using DNA from male and female mice and primers specific for X- (DXNds3)- and Y- (Sry,Zfy) gene sequences. Sensitivity of the assay was measured using groups of 4, 2, or 1 blastomere from dissociated embryos. Efficiency was evaluated using single blastomeres obtained by embryo biopsy. Accuracy of sexing was determined by comparing single-cell results with those of matched blastocysts. Robust amplification of male (XY) and female (XX) gene sequences was obtained in less than 6 hours. The percentage of male (3 bands) and female (1 band) reactions for groups of 4, 2, or 1 blastomere was 100% (6/6), 100% (15/15), and 94.4% (17/18), respectively. Assay efficiency for single, biopsied blastomeres from 4 to 8 cell embryos was 95.8% (207/216). For male and female embryos, sexing of single blastomeres accurately predicted results of matched blastocysts, 100% (10/10) and 100% (13/13), respectively. Simultaneous amplification of one X- and two Y-gene sequences ensured correct interpretation of sexing reactions. How does body size affect the structure and gas exchange capacities of insect tracheae? Do insects become more oxygen-limited as they grow? We addressed these questions by measuring the dimensions of two transverse tracheae within the abdomen of American locusts of different ages, and evaluating the potential for diffusion or convection to provide adequate gas exchange. The grasshopper abdomen has longitudinal tracheae that run along the midgut, heart, nerve cord, and lateral body wall. Transverse tracheae run from each spiracle to the longitudinal tracheae. Dorsal air sacs attach near each spiracle. In both transverse tracheae studied, diffusive capacities increased more slowly than metabolic rates with age, and calculated oxygen gradients necessary to supply oxygen by diffusion increased exponentially with age. However, surgical studies demonstrated that transport of gas through these transverse tracheae occurred by convection, at least in adults. Convective capacities paralleled metabolic rates with age, and the calculated pressure gradients required to sustain oxygen consumption rates by convection were independent of age. Thus, in growing grasshoppers, tracheal capacities matched tissue oxygen needs. Our morphological and physiological data together suggest that use of convection allows older grasshoppers to overcome potential limitations on size imposed by diffusion through tracheal systems. 相似文献

In search of a PDE5 inhibitor for erectile dysfunction, an SAR was developed from a PDE1/PDE5 purine series of leads, which had modest PDE5 potency and poor isozyme selectivity. A compound (41) with PDE5 inhibition and in vivo activity similar to sildenafil was discovered from this effort. In addition, purine 41 demonstrated superior overall PDE isozyme selectivity when compared to the approved PDE5 inhibitors sildenafil, vardenafil, and tadalafil, which may result in a more favorable side-effect profile. 相似文献

Summary In humans, the skin is a particularly sensitive target for 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) and certain halogenated analogs. Reported lesions include a thickening of the epidermis (acanthosis), hyperkeratosis,
and squamous metaplasia of the epithelial lining of the sebaceous glands. In this report we describe ongoing studies on the
actions of TCDD on cultured human epidermal cells. This system has been established as an in vitro model for interfollicular
epidermal hyperkeratinization. Treatment of newly confluent cultures with TCDD results in enhanced differentiation as judged
by histologic examination of the cultures, a decrease in the number of basal proliferating cells, and an increase in the number
of envelope competent (differentiating) cells and terminally differentiated cells with highly cross-linked cornified envelopes.
Changes in the differentiation program are preceded by a decrease in epidermal growth factor (EGF) binding. The concentration
dependence and stereospecificity for these responses suggest the involvement of theAh receptor. We propose that TCDD modulates normal patterns of epidermal differentiation through direct actions on proliferating
basal cells, modulating the responsiveness of these cells to growth factors such as EGF.

The motion after-effect (MAE) persists in crowding conditions, i.e., when the adaptation direction cannot be reliably perceived. The MAE originating from complex moving patterns spreads into non-adapted sectors of a multi-sector adapting display (i.e., phantom MAE). In the present study we used global rotating patterns to measure the strength of the conventional and phantom MAEs in crowded and non-crowded conditions, and when attention was directed to the adapting stimulus and when it was diverted away from the adapting stimulus. The results show that: (i) the phantom MAE is weaker than the conventional MAE, for both non-crowded and crowded conditions, and when attention was focused on the adapting stimulus and when it was diverted from it, (ii) conventional and phantom MAEs in the crowded condition are weaker than in the non-crowded condition. Analysis conducted to assess the effect of crowding on high-level of motion adaptation suggests that crowding is likely to affect the awareness of the adapting stimulus rather than degrading its sensory representation, (iii) for high-level of motion processing the attentional manipulation does not affect the strength of either conventional or phantom MAEs, neither in the non-crowded nor in the crowded conditions. These results suggest that high-level MAEs do not depend on attention and that at high-level of motion adaptation the effects of crowding are not modulated by attention. 相似文献
