Many developing systems obey the principle of continuity: a morphogenetic field, when perturbed, tends to restore the normal local pattern of structures in its organ district. We have investigated physical field theories for a morphogenetic field, seeking constraints which would make a field theory produce the principle of continuity. We assume that during embryonic (ontogenetic) development a leg develops a pattern of positional values and a length which extremize a time-independent functional—the integral, over the length of the leg, of a function of positional values and position. For a single state variable which represents positional value, if a unique extremizing solution for the ontogenetically generated pattern and the length exists, and if no position-dependent functions other than the state variable appear in the integrand, then the principle of continuity is valid: in any regenerated leg the state variable is continuous and each region is locally identical to a region of the ontogenetically generated leg. This proposition is applied to three simple examples. For an exponential gradient and a Jacobi elliptic function there is a set of parameter values and boundary values for which a functional is minimized and the ontogenetically generated leg has an optimal length. Thus a leg which meets these constraints will obey the principle of continuity. However, a functional which when extremized gives a sinusoidal pattern does not in general provide a unique extremal length. Mathematical conditions are discussed under which an ontogenetically generated limb or a regenerated limb represents an asymptotically stable steady state. For a specific model of the transient dynamics in the exponential gradient case, the steady state gradient is asymptotically stable.  相似文献

The use of dogs in military work environments has always aroused great interest in the general population and determining the stress levels they go through is extremely important to maintain their welfare. The aim of this research was to evaluate if the work shifts in military working dogs leads to stress conditions and if this working influences on the reproductive performance and life quality. The study was conducted at the Military Police Kennel located at Campinas, Sao Paulo, Brazil. Eight male dogs of four different breeds (German Shepherd, Belgian Malinois Shepherd, Doberman, and Rottweiler) were evaluated during two different shifts: Working Shifts: animals working 12 hours a day with 2 hour-interval; and Control Shifts: animals that were on their day off (36 hours). Saliva samples were collected for cortisol analysis at the control and working shifts. The study was carried out over 60 days and analyzed behavior, physiology, and reproduction quality. Saliva samples, behavior observation of stereotyping, resting and moving activities and semen analysis were collected by digital stimulation (for combined second and third fractions). The salivary cortisol levels during the control and working shifts were between 0.361–0.438 and 0.312–0.592 µg/dL, respectively; the highest values were found at the end of working shifts. The animals were resting during most of the observation period, but few showed stereotypic behaviors. The testicular consistency was firm and semen parameters were within the normal values in German Shepherd, Belgian Malinois Shepherd, and Doberman dogs. However, Rottweiler dogs had a higher rate of sperm abnormalities, higher salivary cortisol levels, and more stereotypic behaviors. Nevertheless this work highlights the importance of further research relating reproduction and cortisol levels in military dogs.  相似文献

S100B is a Ca2+-modulated protein of the EF-hand type expressed in high abundance in a restricted set of cell types including certain neuronal populations. S100B has been suggested to participate in cell cycle progression, and S100B levels are high in tumor cells, compared with normal parental cells. We expressed S100B in the neuronal cell line PC12, which normally does not express the protein, by the Tet-Off technique, and found the following: (i) proliferation was higher in S100B+ PC12 cells than in S100B- PC12 cells; (ii) nerve growth factor (NGF), which decreased the proliferation of S100B- PC12 cells, was less effective in the case of S100B+ PC12 cells; (iii) expression of S100B made PC12 cells resistant to the differentiating effect of NGF; and (iv) interruption of S100B expression did not result in an immediate restoration of PC12 cell sensitivity to the differentiating effect of NGF. Expression of S100B in PC12 cells resulted in activation of Akt; increased levels of p21WAF1, an inhibitor of cyclin-dependent kinase (cdk) 2 and a positive regulator of cdk4; increased p21WAF1-cyclin D1 complex formation; and increased phosphorylation of the retinoblastoma suppressor protein, Rb. These S100B-induced effects, as well as the reduced ability of S100B+ PC12 cells to respond to NGF, were dependent on Akt activation because they were remarkably reduced or abrogated in the presence of LY294002, an inhibitor of the Akt upstream kinase phosphatidylinositol 3-kinase. Thus, S100B might promote cell proliferation and interfere with NGF-induced PC12 cell differentiation by stimulating a p21WAF1/cyclin D1/cdk4/Rb/E2F pathway in an Akt-mediated manner.  相似文献

Maternal overnutrition during suckling period is associated with increased risk of metabolic disorders in the offspring. We aimed to assess the effect of Vitis vinifera L. grape skin extract (ACH09) on cardiovascular and metabolic disorders in adult male offspring of rats fed a high-fat (HF) diet during lactation. Four groups of female rats were fed: control diet (7% fat), ACH09 (7% fat plus 200 mg kg?1 d?1 ACH09 orally), HF (24% fat), and HF+ACH09 (24% fat plus 200 mg kg?1 d?1 ACH09 orally) during lactation. After weaning, all male offspring were fed a control diet and sacrificed at 90 or 180 days old. Systolic blood pressure was increased in adult offspring of HF-fed dams and ACH09 prevented the hypertension. Increased adiposity, plasma triglyceride, glucose levels and insulin resistance were observed in offspring from both ages, and those changes were reversed by ACH09. Expression of insulin cascade proteins IRS-1, AKT and GLUT4 in the soleus muscle was reduced in the HF group of both ages and increased by ACH09. The plasma oxidative damage assessed by malondialdehyde levels was increased, and nitrite levels decreased in the HF group of both ages, which were reversed by ACH09. In addition, ACH09 restored the decreased plasma and mesenteric arteries antioxidant activities of superoxide dismutase, catalase and glutathione peroxidase in the HF group. In conclusion, the treatment of HF-fed dams during lactation with ACH09 provides protection from later-life hypertension, body weight gain, insulin resistance and oxidative stress. The protective effect ACH09 may involve NO synthesis, antioxidant action and activation of insulin-signaling pathways.  相似文献

We have investigated the fusion of phospholipid vesicles induced by lysozyme and glyceraldehyde-3-phosphate dehydrogenase (GAPDH). Vesicles were composed of dimyristoylphosphatidylcholine/dioleoylphosphatidylethanolamine/ cholesterol (DMPC:DOPE:Chol, 2:1:1). Small unilamellar vesicles (SUV, diameter ca. 30 nm) obtained by extensive sonication or large unilamellar vesicles (LUV, diameters ranged from 100 to 400 nm) obtained by extrusion methods were used. Fusion of LUV induced by lysozyme and GAPDH was drastically decreased when the diameter of the vesicles increased over a value of 100 nm. Lysozyme effect was stopped at the aggregation step while GAPDH effect was stopped at the fusion (lipid mixing) step. Fusion of heterogeneous vesicle populations (SUV with LUV) was observed only with GAPDH and this happened only when the lipids were in the liquid-crystalline state.  相似文献

Activation of brain mitochondrial phospholipase(s) A(2) (PLA(2)) might contribute to cell damage and be involved in neurodegeneration. Despite the potential importance of the phenomenon, the number, identities, and properties of these enzymes are still unknown. Here, we demonstrate that isolated mitochondria from rat brain cortex, incubated in the absence of respiratory substrates, release a Ca(2+)-dependent PLA(2) having biochemical properties characteristic to secreted PLA(2) (sPLA(2)) and immunoreacting with the antibody raised against recombinant type IIA sPLA(2) (sPLA(2)-IIA). Under identical conditions, no release of fumarase in the extramitochondrial medium was observed. The release of sPLA(2) from mitochondria decreases when mitochondria are incubated in the presence of respiratory substrates such as ADP, malate, and pyruvate, which causes an increase of transmembrane potential determined by cytofluorimetric analysis using DiOC(6)(3) as a probe. The treatment of mitochondria with the uncoupler carbonyl cyanide 3-chlorophenylhydrazone slightly enhances sPLA(2) release. The increase of sPLA(2) specific activity after removal of mitochondrial outer membrane indicates that the enzyme is associated with mitoplasts. The mitochondrial localization of the enzyme has been confirmed by electron microscopy in U-251 astrocytoma cells and by confocal laser microscopy in the same cells and in PC-12 cells, where the structurally similar isoform type V-sPLA(2) has mainly nuclear localization. In addition to sPLA(2), mitochondria contain another phospholipase A(2) that is Ca(2+)-independent and sensitive to bromoenol lactone, associated with the outer mitochondrial membrane. We hypothesize that, under reduced respiratory rate, brain mitochondria release sPLA(2)-IIA that might contribute to cell damage.  相似文献

Two marine, Mn(II)-oxidizing bacterial cultures, BIII 45 and BIII 82, were examined spectrophotometrically at ambient temperature for their cytochrome complements. Membrane preparations from an ethylenediaminetetracetate-lysozyme treatment of 48-h cultures of both strains contained type b, c, and o cytochromes. No evidence for a type a cytochrome was noted. “Periplasmic” fractions of both strains also contained small amounts of cytochrome, including cytochrome o, but “intracellular” fractions did not. Type c cytochrome in membrane preparations of culture BIII 45 was consistently reduced by Mn(II) when the membranes were suspended in the periplasmic fraction of the culture. In the case of culture BIII 82, type c cytochrome in membrane preparations was consistently reduced by Mn(II) when the membranes were suspended in either periplasmic or intracellular fractions of the strain. Although, based on previous inhibitor studies, type b cytochrome was also expected to be reduced by Mn(II), no spectrophotometric evidence for its reduction was found, probably because not enough of it was reduced under the steady-state conditions of the experiments.  相似文献

Many studies show environmental enrichment is correlated with benefits to captive animals; however, one should not always assume this positive relationship given that enrichment increases the amount of resources that a territorial animal must defend and possibly affects its aggressive dynamics. In this study, we tested if environmental enrichment affects aggressive interactions in the aggressive fish Nile tilapia (Oreochromis niloticus). We compared fights staged between pairs of male tilapia of similar size (= matched in resource holding potential) in a novel arena that was either barren or enriched, to examine whether enrichment enhances territory value in line with theoretical predictions, with the potential for compromised welfare. We evaluated time elapsed until the first attack (latency), frequency of aggressive interactions and fight duration. We detected fight dynamic differences at the pair level. Higher resource value generated increased aggression but had no effect on fight duration or latency. This conclusion is in line with game theory predictions concerning resource value and contradicts the theory that enrichment of the environment will serve welfare purposes.  相似文献

Fungi of the genus Paracoccidioides are responsible forparacoccidioidomycosis. The occurrence of drug toxicity and relapse in this diseasejustify the development of new antifungal agents. Compounds extracted from fungalextract have showing antifungal activity. Extracts of 78 fungi isolated from rocks ofthe Atacama Desert were tested in a microdilution assay againstParacoccidioides brasiliensis Pb18. Approximately 18% (5) of theextracts showed minimum inhibitory concentration (MIC) values 125.0µg/mL. Among these, extract from the fungus UFMGCB 8030 demonstrated the bestresults, with an MIC of 15.6 µg/mL. This isolate was identified asAspergillus felis (by macro and micromorphologies, and internaltranscribed spacer, β-tubulin, and ribosomal polymerase II gene analyses) and wasgrown in five different culture media and extracted with various solvents to optimiseits antifungal activity. Potato dextrose agar culture and dichloromethane extractionresulted in an MIC of 1.9 µg/mL against P. brasiliensis and did notshow cytotoxicity at the concentrations tested in normal mammalian cell (Vero). Thisextract was subjected to bioassay-guided fractionation using analyticalC18RP-high-performance liquid chromatography (HPLC) and an antifungal assay usingP. brasiliensis. Analysis of the active fractions by HPLC-highresolution mass spectrometry allowed us to identify the antifungal agents present inthe A. felis extracts cytochalasins. These results reveal thepotential of A. felis as a producer of bioactive compounds withantifungal activity.  相似文献
