We report the identification and characterization of p33, the product of Kaposi's sarcoma-associated herpesvirus (KSHV) open reading frame 69 (ORF69), a positional homolog of the conserved herpesvirus protein UL31. p33 is expressed upon induction of viral lytic cycle with early kinetics. Immunofluorescence analysis revealed that in infected cell lines, the protein is localized in the nucleus, both in dotted spots and along the nuclear membrane. Nuclear fractionation experiments showed that p33 partitions with the nuclear matrix, and both immunoblotting of purified virions and immunoelectron microscopy indicated that the novel protein is not a component of the mature virus. Following ectopic expression in KSHV-negative cells, the protein was never associated with the nuclear membrane, suggesting that p33 needs to interact with additional viral proteins to reach the nuclear rim. In fact, after cotransfection with the ORF67 gene, the KSHV positional homolog of UL34, the p33 intranuclear signal changed and the two proteins colocalized on the nuclear membrane. A similar result was obtained when ORF69 was cotransfected with BFRF1, the Epstein-Barr virus (EBV) positional homolog of UL34 and ORF67. Finally, upon cotransfection, ORF69 significantly increased nuclear membrane reduplications induced by BFRF1. The above results indicate that KSHV p33 shares many similarities with its EBV homolog BFLF2 and suggest that functional cross-complementation is possible between members of the gammaherpesvirus subfamily. 相似文献

The vitelline coat (VC) surrounding coelomic eggs of the frog, Rana japonica , comprises bundles of filaments running both parallel and perpendicular to the egg surface. The coat gives little or no staining reaction with PA-CrA-Silver methenamine. In contrast, in the VC of uterine eggs the filament bundles are less conspicuous. and the interstices between the filament bundles stain strongly for carbohydrate. This alteration occurs during passage of the eggs down the first 1/20 th of the oviduct, the pars recta. The epithelium of the p. recta contains secretory cells, which contain electron-dense granules distinct from those in the jelly-secreting cells in more caudal portions of the oviduct. Treatment of coelomic eggs with an extract of p. recta followed by exposure to a sperm suspension resulted in marked swelling and softening of the VC. These results indicate that the contents of the granules secreted from the epithelial cells in the p. recta are deposited in the VC to increase its susceptibility to a fertilizing sperm. 相似文献

The effect of temperature and photoperiod on the rate of predation of nymphs and adults of the predator Macrolophus pygmaeus was studied using Myzus persicae prey feeding on egg-plant and pepper plants. The experiments were conducted at three photoperiods (16L:8D, 12L:12D and 8L:16D), three temperatures (20, 25 and 30 °C), and at 65% r.h. The rate of predation increased with temperature. Predation rate was affected by photoperiod on pepper but not on egg-plant. Females and fifth instar nymphs were the most voracious stages followed by third and fourth instar nymphs and males. First and second instar nymphs consumed far fewer aphids. Predation rate was higher on leaves of pepper than egg-plant, especially at 30 °C. The predominance of secondary forest‐species in Brazilian subtropical forests highlights the importance of understanding the ecology of these taxa, and dendrochronology provides valuable information about the growth and climate response of tree species. The wide distribution of Alchornea triplinervia (Spreng.) Mull. Arg. (Euphorbiaceae), and the presence of growth rings in its wood, leds to its selection for this study. Samples were collected from 34 trees growing in rainy dense forest fragments in the cities of Blumenau and Brusque, in southern Brazil, and subjected to standard dendrochronological techniques. The local chronology spanned from 1889 to 2013 and the age of the trees ranged from 32 to 125 years. The mean age was 73 years and annual increment was 1.13 mm year?1; the diameter and age showed a weak correlation. The results suggest that A. triplinervia growth responded negatively to hot summers and increased rainfall in the previous spring and winter. The high perdiodicity trend characteristic of ENSO activity was reflected in wavelet power spectra of A. triplinervia chronology, affecting positively its growth after strong ENSO events (1970–2009). Negative influences occurred before 1970 during normal ENSO events. Also, there is evidence of the existence of a connection between Pacific and Atlantic sea surface temperatures influencing on tree growth. 相似文献

We present here characterization data for seven new microsatellite markers designed from new microsatellite loci isolated from a microsatellite‐enriched DNA library from Vitis vinifera. The observed heterozygosity varied from 0.73 up to 0.93 and the number of alleles per locus ranged from 12 to 26. This high polymorphism makes these new markers interesting for use in genotyping studies and completing the set of microsatellite markers already available for V. vinifera. Additionally these seven new markers appear to be conserved in four other Vitis species and 15 Vitis hybrids used as rootstocks for V. vinifera cultivation. 相似文献
