A novel amphiphilic fluorescent probe (Fluorazophore-L) with a strongly dipolar, nonionic azoalkane as headgroup and a palmitoyl tail has been synthesized and characterized. Pure Fluorazophore-L was found to be sufficiently amphiphilic to form stable air-water monolayers. An analysis of the surface pressure versus area suggests an area per molecule of about 34+/-2 A(2) at 29 mN m(-1). The partitioning into a lipid membrane model was quantified at the air-water interface by spreading 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine (POPC) monolayers. Measurements with different molar fractions of Fluorazophore-L revealed a small but significant reduction of the mean area in the mixed monolayer. The excess free energy of mixing (-0.5+/-0.1 kT) indicated a weakly attractive interaction slightly above thermal energy, suggesting a good miscibility of the fluorescent probe within the lipid monolayer without major structural modifications. Spectroscopic measurements confirmed the incorporation of Fluorazophore-L into POPC vesicles. The fluorescence lifetime was very long (125+/-5 ns under air) with monoexponential fluorescence decays.  相似文献

Accelerated solvent extraction (ASE) is an alternative sample extraction procedure for fumonisins in corn and corn products. ASE gave results comparable to that of a draft CEN method, but required less extraction time. Background

There is accumulating evidence that autoimmune components, such as autoantibodies and autoantibody depositions, play a role in the pathogenesis of neurodegenerative diseases like Alzheimeŕs disease or Multiple Sclerosis. Due to alterations of autoantibody patterns in sera and aqueous humor, an autoimmune component is also assumed in the pathogenesis of glaucoma, a common reason for irreversible blindness worldwide. So far there has been no convincing evidence that autoantibodies are accumulated in the retina of glaucoma patients and that the local immune homeostasis might be affected.

Methods and Results

Six human glaucomatous donor eyes and nine samples from donors with no recorded ocular disease were included. Antibody microarrays were used to examine the patterns of pro-inflammatory proteins and complement proteins. Analysis of TNF-α and interleukin levels revealed a slight up-regulation exclusively in the glaucomatous group, while complement protein levels were not altered. IgG autoantibody accumulations and/or cellular components were determined by immunohistology (n = 4 per group). A significantly reduced number of retinal ganglion cells was found in the glaucomatous group (healthy: 104±7 nuclei/mm, glaucoma: 67±9 nuclei/mm; p = 0.0007). Cell loss was accompanied by strong retinal IgG autoantibody accumulations, which were at least twice as high as in healthy subjects (healthy: 5.0±0.5 IgG deposits/100 cells, glaucoma: 9.4±1.9 IgG deposits/100 cells; p = 0.004). CD27+ cells and CD27+/IgG+ plasma cells were observed in all glaucomatous subjects, but not in controls.

Conclusion

This work provides serious evidence for the occurrence of IgG antibody deposition and plasma cells in human glaucomatous retina. Moreover, the results suggest that these IgG deposits occurred in a pro-inflammatory environment which seems to be maintained locally by immune-competent cells like microglia. Thereby, glaucoma features an immunological involvement comparable to other neurodegenerative diseases, but also shows a multifactorial pathomechanism, which diverges and might be linked to the specific nature of both eye and retina.  相似文献

Hematopoietic stem cells in myeloproliferative diseases mostly retain the potential to differentiate but are characterized by hyper-responsiveness to growth factors, as well as partial factor-independent growth. The V617F activating point mutation in Jak2 has recently been associated with myeloproliferative disorders. Using various cell line models, mechanisms that contribute to Jak2V617-mediated signaling were investigated. Treatment of the Jak2V617F mutant-expressing erythroid leukemia cell line HEL with a small molecule Jak2 inhibitor was associated with a dose-dependent G(1) cell cycle arrest. This inhibition correlated with decreased expression of cyclin D2 and increased expression of the cell cycle inhibitor p27(Kip). Inhibition of Jak2V617F with a Jak2-targeted small interfering RNA approach resulted in a similar phenotype. Mechanisms leading to altered p27(Kip) and cyclin D2 likely involve inhibition of STAT5, a major target of Jak2 in hematopoietic cells, because a constitutively active form of STAT5 reduced p27(Kip) and increased cyclin D2 expression. Jak2V617F and constitutively active STAT5 also induced high levels of reactive oxygen species, which are sufficient to promote G(1)/S phase transition. In contrast, treatment of HEL cells with the antioxidant N-acetylcysteine decreased cell growth or expression of cyclin D2 and increased expression of p27(Kip). Similar results were obtained in BaF3 cells transfected with Jak2V617F, but these cells required coexpression of the erythropoietin receptor for optimal signaling. These results suggest that regulation of cyclin D2 and p27(Kip) in combination with redox-dependent processes promotes G(1)/S phase transition downstream of Jak2V617F/STAT5 and therefore hint at potential novel targets for drug development that may aid traditional therapy.  相似文献

A new singly bridging complex [Cu(dach)(μ-NCS)(NCS)]n (dach=1,4-diazacycloheptane) has been synthesised and its crystal structure determined. There are many examples of double NCS bridged polymeric chains, but fewer singly bridged ones. IR, ESR and temperature variable magnetic studies are described but no magnetic interaction was found between the copper centres. [Cu(dach)2(N3)]ClO4 has also been characterised by IR, ESR spectra and magnetic studies. The crystal structure determination shows that it is a penta-coordinated monomeric species with an axially coordinated azide linked to the perchlorate counterion by hydrogen bonding.  相似文献

Three seriesof Rh(I) complexes of the type Tp3R,5RRh(LL), with LL = 2 CO (1), norbornadiene (NBD) (2) and 1,5-cyclooctadiene (COD) (3) and the tris (pyrazolyl)borate (Tp) ligands 3R=5R=Me (a), 3R=CF35R=Me (b); and 3R=5R=CF3 (c) were synthesized and fully characterized by IR and multinuclear NMR spectroscopy. Three isomeric forms were identified in solutions of these complexes: two square-planar isomers with a κ2-Tp3R,5R ligand, the uncoordinated pyrazolyl ring occupying either an equatorial position (type A), or an axial position (type B), and a five-coordinate species with a κ3, Tp3R,5R ligand (type C). In the carbonyl complexes 1 the dynamic equilibria between these isomers are solvent dependent. Interestingly, solutions of complex 1c contained all three isomers simultaneously. 103Rh and 13C NMR spectral studies indicate that the NBD compounds, 2, preferentially form square-planar complexes when TpCF3,Me and TpCF3,CF3 are present, while for the COD complexes, 3, square-planar complexes are preferred for all three Tp-type ligands. The X-ray structure of TpCF3,MeRh(CO)2 (1b) was determined (spacce group C2/ c,a = 21.271(9), B = 11.004(3), C = 21.563(9) Å, β = 114.93(3)°, V=4577(3) Å3, Z = 8, R = 3.41, Rw = 4.70). Its structure is of type B, with the third pyrazolyl ring axially placed, the N(4) being almost directly above the Rh atom but exerting only a weak Rh-N interaction.  相似文献

Determination of the nickel (Ni) distribution in tissues of hyperaccumulator plants aids in understanding the strategies and mechanisms used by these plants to take up Ni from soils. Commonly used methods for measuring Ni distribution in plant tissues require expensive equipment and complex sample preparation. We tested a suite of staining methods consisting of dimethylglyoxime (DMG) dissolved in a range of solvents for the mapping of Ni distribution in the Ni hyperaccumulator Berkheya coddii Roessler. The best solution was DMG (10 g l−1) dissolved in borax (25 mM) and KOH (30 mM). Plant tissue cross-sections were imaged under a microscope immediately after DMG application. A Karhunen-Loeve transformation was applied to the images to minimize interference from colours of other origin, e.g. from chlorophyll. The distribution of Ni could be determined at the cellular level and consistent patterns were obtained for replicates. Staining of Ni dissolved in agar at various concentrations was used to calibrate the method. Concentrations as low as 50 mg kg−1 (fresh weight) could be detected. Averaged over several cross-sections the DMG method systematically gave lower concentrations than ICP-OES analysis of the respective plant part, indicating that not all Ni in the tissue reacted with DMG, but only Ni that is readily available. The DMG method may be used in conjunction with spectroscopic methods to resolve biologically active Ni.  相似文献
