The mitochondrial redox (NAD+/NADH) state can be used as a reflection of oxygen availability within the mitochondrion. Previous studies using isolated muscle preparations suggest that active muscle is not hypoxic during lactate production, whereas experiments with humans come to the opposite conclusion. Six men exercised for 5 min at 75% maximal O2 consumption (VO2max) and then at 100% VO2max to exhaustion. Ammonia, oxoglutarate (alpha-ketoglutarate), and glutamate, as well as lactate, were measured in biopsies (vastus lateralis) taken at the end of each exercise. The three former metabolites were used to determine the mass action ratio of glutamate dehydrogenase and thus were used as an estimate of the mitochondrial redox state. Muscle lactate increased (P less than 0.05) to 14.5 and 24.5 mmol/kg wet wt after 75 and 100% VO2max, respectively. At both exercise intensities, muscle ammonia rose (P less than 0.05), glutamate fell (P less than 0.05) to only 30-35% of rest levels, and oxoglutarate declined (P less than 0.05). Despite the high levels of muscle lactate accumulation, the estimated mitochondrial redox rate rose 300% (P less than 0.05) in both exercise bouts. This response should increase the activity of key oxidative enzymes and promote increased VO2. Furthermore the data do not support the concept that muscle lactate is formed because of tissue hypoxia.  相似文献

An image understanding machine vision system for histological diagnoses is based on three interacting expert systems: a diagnostic expert system utilizing terms familiar to pathologists, an interpretive expert system relating human diagnostic concepts to computable histometric features and a scene segmentation expert system which extracts the diagnostic information from the imagery. The control software for the image understanding system resides on a multiprocessor computer. This article details measures to maintain system efficiency and to accommodate the requirements of interprocess communication and processing task scheduling.  相似文献

A radioimmunoassay has been developed for prostaglandin E2 (PGE2) using methyl oxime (MOX) derivatisation and a novel 125Iodine radiolabel. PGE2-methyl oxime (PGE2-MOX) is coupled through an imide linkage to proline in a pro-gly-tyr or similar peptide rather than through the conventional amide linkage to histamine or tyrosine methyl ester. The main advantage of this method is that the imide linkage in the label does not resemble the amide link used in the original antigen and the conjugate is therefore readily displaced by the natural PGE2. This overcomes the traditional difficulty encountered in hapten RIAs where the antiserum has a higher affinity for the label than it has for the compound to be measured. The assay that has been developed using these modifications and a solid-phase second antibody separation step, is both sensitive (with a lower detection limit of 0.5 pg/tube), reliable and simple and has the advantage that methyl oximation of the sample protects the PGE from degrading prior to and during the assay.  相似文献

Summary Human recombinant interleukin-1 (HrIL-1) inhibited WEHI-3B cell growth in a dose-related manner (10–10000 U/ml). Prostaglandin E2 at high concentrations (1000 ng/ml) also inhibited cell growth. When added together, HrIL-1 and prostaglandin E2 inhibited WEHI-3B growth in a synergistic manner (HrIL-1 concentrations of 10–10000 U/ml and prostaglandin E2 concentrations of 10–1000 ng/ml). In contrast to the effects of the cyclooxygenase metabolite of arachidonic acid leukotriene C4, a lipoxygenase metabolite, reversed the cytostatic action of HrIL-1.  相似文献

Abstract Carbon metabolism in Bradyrhizobium japonicum bacteroids is reviewed. Additionally, the bacteroid tricarboxylic acid (TCA) cycle and its regulation under oxygen-limited conditions is considered, with emphasis on possible sites of TCA cycle rate-limiting reactions. Furthermore, we consider other adaptive pathways that may be employed by these organisms while in symbiosis. These pathways include: (1) a poly-β-hydroxy-butyrate shunt, (2) a malate-aspartate shuttle, (3) an α-ketoglutarate-glutamate shunt, (4) a modified dicarboxylic acid cycle, and (5) fermentation pathways leading to lactate, acetaldehyde and ethanol. The effects of oxygen limitation on host carbon metabolism are also considered briefly.  相似文献

Summary Recombinant plasmids carrying ruvA, ruvB, or both were constructed and used to investigate the genetic defects in a collection of UV-sensitive ruv mutants. The results revealed that efficient survival of UV-irradiated cells depends on both ruvA and ruvB, and on a third gene, ruvC, located upstream of the ruvAB operon. Southern blotting analysis was used to locate insertions in ruv and to examine putative deletion mutants. Two Tn10 insertions were located to the region encoding ruvA. Since these insertions caused a deficiency in the activities of both ruvA and ruvB, we concluded that they must exert a polar effect on ruvB. Two putative ruv deletion mutants were shown to be the result of deletion-inversion events mediated during imprecise excision of Tn10. The relevant inversion breakpoints in these mutants were located to ruvA and ruvC.  相似文献
