Ryanodine receptor (RyR)-mediated Ca(2+) dysregulation is associated with Alzheimer's disease (AD) neuropathology. Using 2-photon Ca(2+) imaging and patch clamp recordings in brain slice preparations from young 3xTg-AD and NonTg control mice, we recently demonstrated that RyR-mediated Ca(2+) -induced Ca(2+) release (CICR) is substantially increased within dendrites from AD neurons, such that synaptic stimulation alone is sufficient to generate aberrant CICR. We also observed supra-additive Ca(2+) release upon coincident RyR activation with synaptic stimulation in 3xTg-AD mice. Here, we describe an additional observed phenomenon: generation of patterned Ca(2+) oscillations in the spines and dendrites from AD neurons upon coincident RyR and synaptic stimulation. As the temporal entrainment of Ca(2+) signals influences many downstream cellular and synaptic functions, these abnormal oscillatory patterns may be associated with the structural and functional breakdown of synapses in AD. 相似文献

The toxic metal ion cadmium (Cd2+) induces pleiotropic effects on cell death and survival, in part through effects on cell signaling mechanisms and cytoskeletal dynamics. Linking these phenomena appears to be calmodulin‐dependent activation of the Ca2+/calmodulin‐dependent protein kinase II (CaMK‐II). Here we show that interference with the dynamics of the filamentous actin cytoskeleton, either by stabilization or destabilization, results in disruption of focal adhesions at the ends of organized actin structures, and in particular the loss of vinculin and focal adhesion kinase (FAK) from the contacts is a result. Low‐level exposure of renal mesangial cells to CdCl2 disrupts the actin cytoskeleton and recapitulates the effects of manipulation of cytoskeletal dynamics with biological agents. Specifically, Cd2+ treatment causes loss of vinculin and FAK from focal contacts, concomitant with cytoskeletal disruption, and preservation of cytoskeletal integrity with either a calmodulin antagonist or a CaMK‐II inhibitor abrogates these effects of Cd2+. Notably, inhibition of CaMK‐II decreases the migration of FAK‐phosphoTyr925 to a membrane‐associated compartment where it is otherwise sequestered from focal adhesions in a Cd2+‐dependent manner. AbstractThe inactivation of Pseudomonas aeruginosa biofilms grown on glass under high shear stress and exposed to a range of dissolved ozone concentrations (2, 5 and 7?ppm) at 10 and 20?min was investigated. The regression equation, log reduction (biofilm)?=?0.64?+?0.59×(C – 2)?+?0.33×(T – 10), described the dependence of biofilm inactivation on the dissolved ozone concentration (C, ppm) and contact time (T, min). The predicted D-values were 11.1, 5.7 and 2.2?min at 2, 5 and 7?ppm, respectively. Inactivation of biofilms grown on various surfaces was tested at a single dissolved ozone concentration of 5?ppm and a single exposure time of 20?min. Biofilms grown on plastic materials showed inactivation results similar to that of biofilms on glass, while biofilms grown on ceramics were statistically significantly more difficult to inactivate, suggesting the importance of utilizing non-porous materials in industrial and clinical settings. 相似文献

The diol constituent of Rhus cotinus leaf epicuticular wax has been identified as nonacosane-5,10-diol from chemical investigations of the free compound, the TMSi ether and the nonacosane-5,10-dione prepared from the diol by oxidation. The form and distribution of the crystalline waxes changed as the leaves expanded, dense clusters of short tubes covering the thin ribbons formed during the initial stages of growth. Extracts from marine sources exhibit antimicrobial and antioxidant properties in vitro and there has been great interest within the food industry to move towards natural methods of food preservation. Natural extracts from seaweeds could potentially have a multiple functionality within the food industry to increase safety and enhance the quality of food products. The present study is aimed to assess the antimicrobial activity of a hydrophilic extract from the fucoid brown alga Himanthalia elongata in model food systems. Carbohydrate and protein model food systems (CMFS and PMFS, respectively) were studied at varying concentrations (1 %, 5 % and 10 %) and bacterial inhibition of the extract was investigated against Salmonella abony and Listeria monocytogenes. The extract provided up to 100 % inhibition of the bacteria with a bactericidal effect in CMFS, while a bacteriostatic effect was seen in PMFS. In general, there was a significant difference (P?<?0.05) between the efficacies of the extract against S. abony as compared to L. monocytogenes with higher inhibition for S. abony. In terms of antioxidants; the extract had a total phenolic content of 34.0 mg GAE g?1 of extract and a DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity of 139.8 mg AAE g of extract. The results of the present study are promising as it provides an insight for the inclusion of seaweed extracts into real food systems. 相似文献

Microclimate was found to affect the daily flight activity ofSimulium arcticum in a pasture situation in central Alberta, Canada. Vapour pressure, air temperature, light intensity, and wind gust velocity had the greatest correlation to flight activity. These variables were correlated to support the concept that they influence the energy and water balances of a black fly.
