Tilapia is one of most important foodfish species. The low omega-3 to omega-6 fatty acid ratio in freshwater tilapia meat is disadvantageous for human health. Increasing omega-3 content is an important task in breeding to increase the nutritional value of tilapia. However, conventional breeding to increase omega-3 content is difficult and slow. To accelerate the increase of omega-3 through marker-assisted selection (MAS), we conducted QTL mapping for fatty acid contents and profiles in a F2 family of saline tilapia generated by crossing red tilapia and Mozambique tilapia. The total omega-3 content in F2 hybrid tilapia was 2.5 ± 1.0 mg/g, higher than that (2.00 mg/g) in freshwater tilapia. Genotyping by sequencing (GBS) technology was used to discover and genotype SNP markers, and microsatellites were also genotyped. We constructed a linkage map with 784 markers (151 microsatellites and 633 SNPs). The linkage map was 2076.7 cM long and consisted of 22 linkage groups. Significant and suggestive QTL for total lipid content were mapped on six linkage groups (LG3, -4, -6, -8, -13, and -15) and explained 5.8–8.3% of the phenotypic variance. QTL for omega-3 fatty acids were located on four LGs (LG11, -18, -19, and -20) and explained 5.0 to 7.5% of the phenotypic variance. Our data suggest that the total lipid and omega-3 fatty acid content were determined by multiple genes in tilapia. The markers flanking the QTL for omega-3 fatty acids can be used in MAS to accelerate the genetic improvements of these traits in salt-tolerant tilapia.  相似文献

Astrocytes are known to play multi-functional roles in support of many homeostatic mechanisms in the central nervous system including host defense mechanisms. Despite the ability of cytokines to alter gene expression and cellular activity, their effect on receptor-mediated poly-phosphoinositide (poly-PI) signaling pathway has not been examined in detail. In this study, an immortalized astrocyte cell line (DITNC) was used to test the effect of IL-1 exposure on the poly-PI signaling pathway. Similar to primary astrocytes, DITNC cells exhibit P2-purinergic receptor response to ATP and UTP leading to transient increases in inositol 1,4,5-trisphosphate [Ins(1,4,5)P3] and intracellular calcium concentration, [Ca2+]i. Upon exposure of DITNC cells to IL-1 (100U/ml) for 24 hrs, an increased response to the poly-PI agonists was observed. The increase in ATP-mediated Ins(1,4,5)P3 release could not be attributed to a shift in the ATP dose or an alteration of the time profile for the release of Ins(1,4,5)P3. Since the increase in response required a lag time of 4 hr after IL-1 exposure, it is unlikely that this effect was due to a direct interaction of IL-1 with the purinergic receptor. On the other hand, an increase in ATP response could be observed in DITNC cells exposed to conditioned medium obtained after IL-1 treatment. It can be concluded that exposure of astrocytes to cytokines may lead to an increase in receptor-mediated poly-PI signaling activity and this may involve compounds secreted into the culture medium, e.g., the secretory phospholipase A2.  相似文献

Many current theories of community function are based on the assumption that disturbances such as herbivory act to reduce the importance of neighbor interactions among plants. In this study, we examined the effects of herbivory (primarily by nutria, Myocastor coypus) on neighbor interactions between three dominant grasses in three coastal marsh communities, fresh, oligohaline, and mesohaline. The grasses studied were Panicum virgatum, Spartina patens, and Spartina alterniflora, which are dominant species in the fresh, oligohaline, and mesohaline marshes, respectively. Additive mixtures and monocultures of transplants were used in conjunction with exclosure fences to determine the impact of herbivory on neighbor interactions in the different marsh types. Herbivory had a strong effect on all three species and was important in all three marshes. In the absence of herbivores, the impact of neighbors was significant for two of the species (Panicum virgatum and Spartina patens) and varied considerably between environments, with competition intensifying for Panicum virgatum and decreasing for Spartina patens with increasing salinity. Indications of positive neighbor effects (mutualisms) were observed for both of these species, though in contrasting habitats and to differing degrees. In the presence of herbivores, however, competitive and positive effects were eliminated. Overall, then, it was observed that in this case, intense herbivory was able to override other biotic interactions such as competition and mutualism, which were not detectable in the presence of herbivores.  相似文献

Cell transplantation has potential benefits for tissue replacement in the the enhancement of tissue regeneration and as cell-mediated gene therapy for systemic diseases. The transplantation of myoblasts into skeletal muscle also allows gene transfer into cells of the host since myoblasts fuse with host fibers thereby forming hybrid myofibers. The success of myoblast transplantation can be determined by a variety of measures, such as the percentage of myoblasts that fuse, the number of hybrid myofibers formed, or the level of transgene expression. Each measure is a reflection of the fate of the transplanted cells. In order to compare different measures of transplantation efficacy, we followed the fate of transplanted myoblasts expressing the marker enzyme β-galactosidase (β-gal) in two different assays. Two weeks after transplantation, the number of hybrid myofibers was determined histochemically, whereas transgene (β-gal) expression was measured biochemically. To control for variabilities of transplantation among different animals, we obtained both measurements from each muscle by using alternate cryosections in the two assays. Within each individual muscle, both hybrid fiber number and/β-gal expression were maximal at the site of implantation and diminished in parallel with distance from the site. However, for determining the success of transplantation among groups of muscles, these two measures of efficacy yielded discordant results: the transplants with the highest number of hybrid fibers were not the transplants with the greatest β-gal activity. Such discrepancies are likely due to regional variations at the transplantation site that arise when cells are introduced into a solid tissue. These results demonstrate the importance of multiple measures of cell fate and transplantation efficacy for studies of cell trans-plantation and for the application of such studies to cell therapy and cell-mediated gene therapy.  相似文献

Abstract: Autoradiographic studies have shown that σ receptors are enriched in the locus coeruleus, the origin of noradrenergic projections to the cerebellum, as well as in the Purkinje, molecular, and granular layers and the interpositus cerebellar nucleus of the cerebellum itself. In contrast, the cerebellum is relatively poor in phencyclidine (PCP) binding sites, which have been historically confused with σ sites. The high ratio of σ to PCP receptors in cerebellum is advantageous for discriminating σ-mediated physiological effects. σ agonists and antagonists have been shown to regulate N -methyl- d -aspartate (NMDA)-stimulated norepinephrine release in hippocampus, which is innervated by locus coeruleus projections. We now report that σ drugs also regulate norepinephrine release from cerebellum. In contrast to findings in the hippocampus, where regulation is via σ1 and σ2 receptors, σ-mediated regulation in cerebellum seems to be primarily via σ1 receptors. In radioligand binding studies, we find that σ receptors primarily of the σ1 type are present in the cerebellum. We further report that binding to σ receptors in cerebellum is not affected by the addition of NMDA or glycine or by the presence of NMDA antagonists, suggesting that σ receptors are not located within the NMDA-operated cation channel in this brain region.  相似文献

Chondrus crispus Stackhouse alternates between two isomorphic life history phases that differ in cell-wall phycocolloid composition. It has been long hypothesized that the gametophyte, with strong-gelling kappa-type carrageenans, is mechanically superior to the tetrasporophyte, with nongelling lambda-type carrageenans, which could contribute to the observed gametophytic dominance in many wave-swept environments. Standard mechanical tests were performed on distal tissues of C. crispus sampled from a range of environments in Narragansett Bay, Rhode Island, using a tensometer equipped with a video extensometer. Life history phase was by far the most important determinant of mechanical properties, whereas environmental factors had only modest influence (vertical distribution) or no effect (exposure); gametophytic distal tissues were 43% stronger, 21% more extensible, and 21% stiffer than tetrasporophytic distal tissues. However, the superior strength of gametophytic tissues was not evident at the stipe/holdfast junction (where breakage typically occurs), and the two phases were equally susceptible to dislodgment by a given force. The primary ecophysiological role of carrageenans in C. crispus may not be the provision of a structure to resist wave action.  相似文献

A biolistic particle delivery system was used to genetically transform embryogenic tissue of Pinus radiata. The introduced DNA contained a uidA reporter gene under the control of either the tandem CaMV 35S or the artificial Emu promoter, and the npt II selectable marker controlled by the CaMV 35S promoter. The average number of stable, geneticin-resistant lines recovered was 0.5 per 200 mg fresh weight bombarded tissue. Expression of the uidA reporter gene was detected histochemically and fluorimetrically in transformed embryogenic tissue and in derived mature somatic embryos and regenerated plants. The integration of uidA and npt II genes into the Pinus radiata genome was demonstrated using PCR amplification of the inserts and Southern hybridisation analysis. The expression of both genes in transformed tissue was confirmed by Northern hybridisation analysis. More than 150 transgenic Pinus radiata plants were produced from 20 independent transformation experiments with four different embryogenic clones. To assess the possible physiological function of chlorogenic acid (CGA, 3-O-caffeoylquinic acid) in vivo , we characterized the free radical scavenging properties of pure phenylpropanoids and leaf extracts against two free radicals, superoxide and the 2,2'-azinobis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radical cation. CGA was found to be a highly efficient scavenger of these free radicals, surpassing the activity of all other phenylpropanoids tested, as well as the 'classical' antioxidant ascorbate. Seasonal differences in the leaf content of CGA were examined in field populations of the broadleaf evergreen Mahonia repens growing in different light environments. Leaves of fully sun-exposed plants contained significantly more (74 ± 10 mmol m–2) CGA in winter than leaves from plants growing under deeply shaded conditions (17 ± 2 mmol m–2). Sun-acclimated, but not shade-acclimated, leaves also produced high levels of anthocyanins in winter, suggesting a simultaneous increase in carbon flow through the phenylpropanoid and flavonoid pathways in response to high light and seasonal low temperature stress. In summer, high light-acclimated leaves contained ≈ 2-fold less CGA than in winter, whereas CGA levels were similar between seasons in shaded leaves. Consistent with the strong scavenging capacity of CGA measured in vitro , a linear correlation was observed between CGA content and the antioxidant activity of leaf extracts in both scavenging assays. On the basis of these results, we propose that CGA is a powerful hydrogen-donating antioxidant that may play an important role in mitigating the effects of oxidative stress in plants.  相似文献
