This paper describes an effective method for the solubilization of microsomal HMG-CoA reductase from rat liver. Exposing the microsomes to a freeze-thaw treatment solubilized 80% of the microsomal reductase activity. Subsequently, a 25-fold purification has led to an enzyme preparation with a specific activity of 10–14 nmoles MVA per min per mg of protein and an increased stability. 相似文献

Amer. Jour. Bot. 49(8): 887–891. Illus. 1962.—Widespread throughout central U.S. is a rhizomatous form of B. curtipendula that basically is tetraploid (2n = 40). In the southwest the predominant type is a caespitose aneuploid with a high chromosome number (2n = ca. 80 to 2n = ca. 102). The present study has shown the presence of an extensive series of rhizomatous aneuploids in central Texas, with chromosome numbers ranging from 2n = 41 to 2n = 64. The distribution of these plants is centered about the region of overlap in the ranges of the 2 previously mentioned types. Available evidence indicates that the rhizomatous aneuploids have arisen through hybridization of the caespitose aneuploids and the rhizomatous tetraploids. 相似文献

Megasporogenesis and embryo sac development in the sexually reproducing taxa Bouteloua warnockii (2n = 22), B. media (2n = 20), B. uniflora Vasey var. uniflora (2n = 20), B. uniflora var. coahuilensis Gould and Kapadia (2n = 20), and B. curtipendula var. curlipendula (2n = 40) all were found to be of the Adoxa type, in which all 4 megaspores persist and divide once to form an 8-nucleate embryo sac. On the other hand, evidence indicated that plants of B. curtipendula var. caespitosa with high ancuploid chromosome numbers reproduce by pseudogamous fertilization of an aposporous embryo sac. In this taxon the megaspore mother cell did not go beyond the first anaphase of meiosis and the functional embryo sac developed from a nucellar cell. Human PRL receptor cDNA clones from hepatoma (Hep G2) and breast cancer (T-47D) libraries were isolated by using a rat PRL receptor cDNA probe. The nucleotide sequence predicts a mature protein of 598 amino acids with a much longer cytoplasmic domain than the rat liver PRL receptor. Although this extended region has additional segments of localized sequence identity with the human GH receptor, there is no identity with any consensus sequences known to be involved in hormonal signal transduction. This cDNA will be a valuable tool to better understand the role of PRL in the development and growth of human breast cancer. 相似文献

We have previously shown that the peroxisomal targeting signal in firefly luciferase consists of the COOH-terminal three amino acids of the protein, serine-lysine-leucine (Gould, S.J., G.A. Keller, N. Hosken, J. Wilkinson, and S. Subramani, 1989. J. Cell Biol. 108:1657-1664). Antibodies were raised against a synthetic peptide that contained this tripeptide at its COOH terminus. Immunofluorescence and immunocryoelectron microscopy revealed that the anti-peptide antibodies specifically detected peroxisomes in mammalian cells. Further characterization revealed that the antibodies were primarily directed against the COOH-terminal three amino acids of the peptide. In Western blot experiments, the antibodies recognized 15-20 rat liver peroxisomal proteins, but reacted with only a few proteins from other subcellular compartments. These results provide independent immunological evidence that the peroxisomal targeting signal identified in firefly luciferase is present in many peroxisomal proteins. 相似文献

FSH binding and cAMP responses to FSH in Sertoli cell-enriched testes were not affected by the vitamin A (retinol) status of the animals. These results indicate that changes in Sertoli cell functions during vitamin A deficiency are independent of FSH-Sertoli cell interactions. Concentrations of serum androgen binding protein (ABP) in vitamin A-deficient rats were consistently higher than those of control animals throughout the study period. The accumulation of testicular fluid after efferent duct ligation, an indication of Sertoli cell secretory function, was normal in vitamin A-deficient rats at least until 70 days of age, but declined thereafter. ABP concentrations in seminiferous tubular fluid of vitamin A-deficient rats increased transitorily during the 70-80-day age period but returned to normal by 90 days. The increment of ABP in seminiferous tubular fluid after efferent duct ligation, and ABP concentrations in interstitial fluid were consistently lower in vitamin A-deficient rats. The higher serum ABP in vitamin A-deficient rats therefore cannot be explained by an increase in the permeability of Sertoli-cell tight junctions or basement membrane. 相似文献

A Pseudomonas stutzeri isolate rapidly reduced both selenite and selenate ions to elemental selenium at initial concentrations of both anions of up to 48.1 mM. Optimal selenium reduction occurred under aerobic conditions between pH 7.0 and 9.0 and at temperatures of 25 to 35°C. Reduction of both selenite and selenate was unaffected by a number of anions except for sulfite, chromate, and tungstate ions, which inhibited both growth and reduction. 相似文献
