Understanding the relationship between growth and temperature will aid in the evaluation of thermal stress and threats to ectotherms in the context of anticipated climate changes. Most Pecten maximus scallops living at high latitudes in the northern hemisphere have a larger maximum body size than individuals further south, a common pattern among many ectotherms. We investigated differences in daily shell growth among scallop populations along the Northeast Atlantic coast from Spain to Norway. This study design allowed us to address precisely whether the asymptotic size observed along a latitudinal gradient, mainly defined by a temperature gradient, results from differences in annual or daily growth rates, or a difference in the length of the growing season. We found that low annual growth rates in northern populations are not due to low daily growth values, but to the smaller number of days available each year to achieve growth compared to the south. We documented a decrease in the annual number of growth days with age regardless of latitude. However, despite initially lower annual growth performances in terms of growing season length and growth rate, differences in asymptotic size as a function of latitude resulted from persistent annual growth performances in the north and sharp declines in the south. Our measurements of daily growth rates throughout life in a long-lived ectothermic species provide new insight into spatio-temporal variations in growth dynamics and growing season length that cannot be accounted for by classical growth models that only address asymptotic size and annual growth rate.  相似文献

The ALEU2 gene, encoding beta-isopropylmalate dehydrogenase, was isolated from the non-conventional yeast Arxula adeninivorans. The isolated gene harbours an open reading frame of 1086 bp, encoding a putative protein of 362 amino acids. The derived protein sequence shares a high degree of homology with other fungal beta-isopropylmalate dehydrogenases thus confirming the identity of the gene. The isolated ALEU2 gene was tested for its suitability to complement the auxotrophy of an A. adeninivorans aleu2 host. For this purpose the plasmid pAL-ALEU2m which contains the ALEU2 gene as a selection marker and the 25S rDNA for targeting was employed in transformation experiments. Transformants harboured a single copy of the heterologous DNA and were found to be mitotically stable. For assessment of heterologous gene expression, two model genes were incorporated into the vector: the GFP gene, encoding intracellular green fluorescent protein, and the HSA gene, encoding the secreted human serum albumin. For expression control, both gene sequences were fused to the constitutive A. adeninivorans-derived TEF1 promoter and the Saccharomyces cerevisiae-derived PHO5 terminator. In the respective recombinant strains the GFP was localised in the cytoplasm, whereas more than 95% of the HSA accumulated in the culture medium. In initial fermentation trials using a 200-ml shake flask, maximal HSA product levels were observed after 96 h of cultivation.  相似文献

Effects of the ionophore A23187 on isolated broken and intact chloroplasts in the pH range of 6.2 to 7.6 have been studied. In both types of chloroplasts, uncoupling of photosynthetic electron transport by A23187 (6–10 μm) was mediated either by Mg2+ or—in the absence of divalent cations (i.e., when EDTA was added to the medium)—by high concentrations of Na+, but not of K+ ions. At increased concentrations of the ionophore (above about 10 μm) and high pH (7.2 to 7.6), uncoupling in broken chloroplasts was also mediated by K+ ions. The inhibition of the energy-dependent slow decline of chlorophyll fluorescence in intact chloroplasts by the ionophore (which denotes uncoupling) is reversed by EDTA in the presence of K+, but not of Na+ ions. In 3-(3′,4′-dichlorophenyl)1,1-dimethylurea-poisoned intact chloroplasts, the yield of variable chlorophyll fluorescence is lowered by A23187 + EDTA and increased again by addition of NaCl or KCl. Chlorophyll fluorescence spectra at 77 °K of intact chloroplasts incubated with A23187 + EDTA indicated that the distribution of excitation energy had changed in favor of photosystem I, as expected from a depletion of Mg2+. This change was reversed by MgCl2+, KCl, or NaCl. From a comparison of low-temperature fluorescence spectra of broken and intact chloroplasts at different levels of Mg2+ in the medium, the concentration of free Mg2+ in the stroma of the intact chloroplasts at pH 7.6 in the dark was estimated at 1 to 4 mm. The results show that in chloroplasts the specificity of A23187 for divalent cations is limited. In the presence of EDTA, the ionophore mediates fast Na+H+ exchange across thylakoid membranes, whereas K+ is transferred much less efficiently. Both Na+ and K+ ions seem to be transported readily across the chloroplast envelope by the action of the ionophore, leading to an exchange of Mg2+ for monovalent cations at the thylakoid membrane surfaces in intact chloroplasts.  相似文献

Climate warming affects the rate and timing of the development in ectothermic organisms. Short‐living, ectothermic organisms (including many insects) showing thermal plasticity in life‐cycle regulation could, for example, increase the number of generations per year under warmer conditions. However, changed phenology may challenge the way organisms in temperate climates deal with the available thermal time window at the end of summer. Although adaptive plasticity is widely assumed in multivoltine organisms, rapid environmental change could distort the quality of information given by environmental cues that organisms use to make developmental decisions. Developmental traps are scenarios in which rapid environmental change triggers organisms to pursue maladaptive developmental pathways. This occurs because organisms must rely upon current environmental cues to predict future environmental conditions and corresponds to a novel case of ecological or evolutionary traps. Examples of introduced, invasive species are congruent with this hypothesis. Based on preliminary experiments, we argue that the dramatic declines of the wall brown Lasiommata megera in northwestern Europe may be an example of a developmental trap. This formerly widespread, bivoltine (or even multivoltine) butterfly has become a conundrum to conservationist biologists. A split‐brood field experiment with L. megera indeed suggests issues with life‐cycle regulation decisions at the end of summer. In areas where the species went extinct recently, 100% of the individuals developed directly into a third generation without larval diapause, whereas only 42.5% did so in the areas where the species still occurs. Under unfavourable autumn conditions, the attempted third generation will result in high mortality and eventually a lost or ‘suicidal’ third generation in this insect with non‐overlapping, discrete generations. We discuss the idea of a developmental trap within an integrated framework for assessing the vulnerability of species to climate change.  相似文献

Temperature has a major influence on the rate of embryonic development in ectothermic organisms. While incubation experiments unambiguously show that constant high temperature accelerates development and shortens embryonic life, studies on the effect of fluctuating temperatures have generated contradictory results. Grass snakes (Natrix natrix) occur at latitudes and altitudes that are unusually cool for an oviparous reptile. In these cool climates females typically lay their eggs in heat‐generating anthropogenic microhabitats that provide either a highly fluctuating (compost piles) or a relatively constant (manure heaps) thermal nesting environment. A laboratory experiment with fluctuating and constant incubation temperatures mimicking those recorded in such nests in the field showed that this nest‐site dichotomy influences the development of the embryos, and the morphology and locomotor performance of the hatchlings. The incubation period increased at fluctuating temperatures and the fact that the rate of embryonic development showed a decelerating pattern with temperature suggests that periods of low temperature had a relatively larger influence on average development than periods of high temperature. Our study demonstrates how a dichotomy in the nesting environments available to female grass snakes in cool climates can affect variation in the duration of the incubation period and offspring phenotypes in ways that may have consequences for fitness. © 2012 The Linnean Society of London, Biological Journal of the Linnean Society, 2012, ??, ??–??.  相似文献

The extracellular acid phosphatase-encoding Arxula adeninivorans APHO1 gene was isolated using degenerated specific oligonucleotide primers in a PCR screening approach. The gene harbours an ORF of 1449 bp encoding a protein of 483 amino acids with a calculated molecular mass of 52.4 kDa. The sequence includes an N-terminal secretion sequence of 17 amino acids. The deduced amino acid sequence exhibits 54% identity to phytases from Aspergillus awamori, Asp. niger and Asp. ficuum and a more distant relationship to phytases of the yeasts Candida albicans and Debaryomyces hansenii (36–39% identity). The sequence contains the phosphohistidine signature and the conserved active site sequence of acid phosphatases. APHO1 expression is induced under conditions of phosphate limitation. Enzyme isolates from wild and recombinant strains with the APHO1 gene expressed under control of the strong A. adeninivorans-derived TEF1 promoter were characterized. For both proteins, a molecular mass of approx. 350 kDa, corresponding to a hexameric structure, a pH optimum of pH 4.8 and a temperature optimum of 60°C were determined. The preferred substrates include p-nitrophenyl-phosphate, pyridoxal-5-phosphate, 3-indoxyl-phosphate, 1-naphthylphosphate, ADP, glucose-6-phosphate, sodium-pyrophosphate, and phytic acid. Thus the enzyme is a secretory acid phosphatase with phytase activity and not a phytase as suggested by strong homology to such enzymes.  相似文献

The yeast Arxula adeninivorans is considered to be a promising producer of recombinant proteins. However, growth characteristics are poorly investigated and no industrial process has been established yet. Though of vital interest for strain screening and production processes, rationally defined culture conditions remain to be developed. A cultivation system was evolved based on targeted sampling and mathematical analysis of rationally designed small-scale cultivations in shake flasks. The oxygen and carbon dioxide transfer rates were analyzed as conclusive online parameters. Oxygen limitation extended cultivation and led to ethanol formation in cultures supplied with glucose. Cultures were inhibited at pH-values below 2.8. The phosphorus demand was determined as 1.55 g phosphorus per 100 g cell dry weight. Synthetic SYN6 medium with 20 g glucose l?1 was optimized for cultivation in shake flasks by buffering at pH 6.4 with 140 mmol MES l?1. Optimized SYN6 medium and operating conditions provided non-limited cultivations without by-product formation. A maximal specific growth rate of 0.32 h?1 and short fermentations of 15 h were achieved. A pH optimum curve was derived from the oxygen transfer rates of differently buffered cultures, showing maximal growth between pH 2.8 and 6.5. Furthermore, it was shown that the applied medium and cultivation conditions were also suitable for non-limiting growth and product formation of a genetically modified A. adeninivorans strain expressing a heterologous phytase.  相似文献
