Knowledge of the latitudinal patterns in biotic interactions, and especially in herbivory, is crucial for understanding the mechanisms that govern ecosystem functioning and for predicting their responses to climate change. We used sap‐feeding insects as a model group to test the hypotheses that the strength of plant–herbivore interactions in boreal forests decreases with latitude and that this latitudinal pattern is driven primarily by midsummer temperatures. We used a replicated sampling design and quantitatively collected and identified all sap‐feeding insects from four species of forest trees along five latitudinal gradients (750–1300 km in length, ten sites in each gradient) in northern Europe (59 to 70°N and 10 to 60°E) during 2008–2011. Similar decreases in diversity of sap‐feeding insects with latitude were observed in all gradients during all study years. The sap‐feeder load (i.e. insect biomass per unit of foliar biomass) decreased with latitude in typical summers, but increased in an exceptionally hot summer and was independent of latitude during a warm summer. Analysis of combined data from all sites and years revealed dome‐shaped relationships between the loads of sap‐feeders and midsummer temperatures, peaking at 17 °C in Picea abies, at 19.5 °C in Pinus sylvestris and Betula pubescens and at 22 °C in B. pendula. From these relationships, we predict that the losses of forest trees to sap‐feeders will increase by 0–45% of the current level in southern boreal forests and by 65–210% in subarctic forests with a 1 °C increase in summer temperatures. The observed relationships between temperatures and the loads of sap‐feeders differ between the coniferous and deciduous tree species. We conclude that climate warming will not only increase plant losses to sap‐feeding insects, especially in subarctic forests, but can also alter plant‐plant interactions, thereby affecting both the productivity and the structure of future forest ecosystems.  相似文献

Breeding outcome in birds can be influenced by both intrinsic and extrinsic factors, which may contribute differentially to each stage of the breeding cycle. Island raptors are relatively threatened species that rely on simplified food webs and usually co-occur with invasive species. Here, we studied a population of Striated Caracara Phalcoboenus australis in Isla de los Estados (Argentina) to understand the relative contribution of food availability, topographical features and territoriality to breeding parameters. Caracaras in the area depend on the short seasonal (i.e. pulsed) availability of seabirds for feeding their chicks. On the other hand, the recent introduction of non-native ungulates may have led to a suboptimal breeding environment by reducing Caracara nest-site availability within the seabird colonies via disturbance of the grassland. Using data from seven breeding seasons we analyse the abundance, the proportion of breeding individuals, the spatial structure of breeding events and the population-level breeding performance, including chick sex ratio, to study whether the breeding ecology of this species is still shaped by marine food web pulses or could be associated with invasive herbivore disturbance. The breeding adults represented only 6–15% of the population. The spatial pattern of nest-sites did not differ from random arrangement after considering the heterogeneous distribution of food availability. Territory identity accounted for most of the variation in clutch size and productivity, and penguin availability explained part of the variability in territory occupancy and hatching rate. The chick sex ratio was biased towards females and varied according to brood size, with the more productive territories producing 91% of males. The breeding cycle of this population is still mainly dependent upon the Rockhopper Penguin Eudyptes chrysocome food pulse and presents mixed evidence of being disrupted by invasive herbivores. The studied factors contributed differentially to the output of each reproductive stage, with territory identity accounting for most of the variation. Our results suggest that individual-level, long-term data could be relevant to understanding the dynamics of this species in the Fuegian archipelago, and thus a broad-scale research programme is warranted to assess the conservation status and propose future management scenarios for this species.  相似文献

The LEXE motif, conserved in eukaryotic type DNA polymerases, is placed close to the polymerization active site. Previous studies suggested that the second Glu was involved in binding a third noncatalytic ion in bacteriophage RB69 DNA polymerase. In the protein-primed DNA polymerase subgroup, the LEXE motif lacks the first Glu in most cases, but it has a conserved Phe/Trp and a Gly preceding that position. To ascertain the role of those residues, we have analyzed the behavior of mutants at the corresponding φ29 DNA polymerase residues Gly-481, Trp-483, Ala-484, and Glu-486. We show that mutations at Gly-481 and Trp-483 hamper insertion of the incoming dNTP in the presence of Mg2+ ions, a reaction highly improved when Mn2+ was used as metal activator. These results, together with previous crystallographic resolution of φ29 DNA polymerase ternary complex, allow us to infer that Gly-481 and Trp-483 could form a pocket that orients Val-250 to interact with the dNTP. Mutants at Glu-486 are also defective in polymerization and, as mutants at Gly-481 and Trp-483, in the pyrophosphorolytic activity with Mg2+. Recovery of both reactions with Mn2+ supports a role for Glu-486 in the interaction with the pyrophosphate moiety of the dNTP.  相似文献

The Bellingshausen Sea is one of the most remote and least surveyed seas of the Southern Ocean, so that little was known about benthic communities and those factors that determine community structuring until recently. The present work aims at characterizing the structure and spatial distribution of echinoid assemblages in the Bellingshausen Sea, as well as identifying the environmental factors that determine assemblage structuring. Echinoids were collected at 32 stations using an Agassiz trawl, at depths of 86–3,304?m, during BENTART oceanographic expeditions led in 2003 and 2006. Sediment and bottom water properties were analysed using an USNEL-type box corer and a Neil Brown Instrument System Mark III CTD, respectively. Echinoids were found at all stations, except Peter I Island. Seventeen species were identified, representing 22?% of the echinoid species present in the Southern Ocean and increasing twofold the number of species recorded in the Bellingshausen Sea so far. The echinoid fauna is dominated by the very abundant species Sterechinus antarcticus. Depth is the key factor that determines the nature of echinoid assemblages, which are mainly divided into the continental shelf, the slope and the deep-sea basin. In addition, sediment properties, namely redox values, organic matter and mud content, best match species dispersion on the shelf. Sediment properties affect echinoid distribution depending on species food range and feeding strategy. As it might be expected, sediment properties more strongly influence specialist feeders (Schizasteridae and Cidaridae) than generalists (Echinidae).  相似文献

The Anagni intra-apennine basin is especially rich in palaeontological and archaeological sites, spanning between middle Villafranchian and upper Pleistocene. The Italian Institute of Human Palaeontology has carried out researches in this region in the last 30 years. At Costa San Giacomo, middle Villafranchian yellow sands contain Mastodon arvernensis and Elephas meridionalis. At Colle Marino, a travertine layer and lime-clay contains mode 1 lithic industry, below any volcanic layers whose lower limit is 700 Ky old. Excavations have been restarted at Fontana Ranuccio, a 458 Ky Acheulian site with rich mammalian fauna (Elephas antiquus, Ursus deningeri, Bos primigenius, Dama clactoniana, Equus mosbachensis), a remarkable bone industry and four human teeth. Drillings, for about 40 m, have identified Matuyama-Brunhes limit 23 m under the main volcanic pyroclastite, in a thick limnic clay layer.  相似文献

Many repair and recombination proteins play essential roles in telomere function and chromosome stability, notwithstanding the role of telomeres in “hiding” chromosome ends from DNA repair and recombination. Among these are XPF and ERCC1, which form a structure-specific endonuclease known for its essential role in nucleotide excision repair and is the subject of considerable interest in studies of recombination. In contrast to observations in mammalian cells, we observe no enhancement of chromosomal instability in Arabidopsis plants mutated for either XPF (AtRAD1) or ERCC1 (AtERCC1) orthologs, which develop normally and show wild-type telomere length. However, in the absence of telomerase, mutation of either of these two genes induces a significantly earlier onset of chromosomal instability. This early appearance of telomere instability is not due to a general acceleration of telomeric repeat loss, but is associated with the presence of dicentric chromosome bridges and cytologically visible extrachromosomal DNA fragments in mitotic anaphase. Such extrachromosomal fragments are not observed in later-generation single-telomerase mutant plants presenting similar frequencies of anaphase bridges. Extensive FISH analyses show that these DNAs are broken chromosomes and correspond to two specific chromosome arms. Analysis of the Arabidopsis genome sequence identified two extensive blocks of degenerate telomeric repeats, which lie at the bases of these two arms. Our data thus indicate a protective role of ERCC1/XPF against 3′ G-strand overhang invasion of interstitial telomeric repeats. The fact that the Atercc1 (and Atrad1) mutants dramatically potentiate levels of chromosome instability in Attert mutants, and the absence of such events in the presence of telomerase, have important implications for models of the roles of recombination at telomeres and is a striking illustration of the impact of genome structure on the outcomes of equivalent recombination processes in different organisms.  相似文献

To help cells cope with protein misfolding and aggregation, Hsp70 molecular chaperones selectively bind a variety of sequences (“selective promiscuity”). Statistical analyses from substrate-derived peptide arrays reveal that DnaK, the E. coli Hsp70, binds to sequences containing three to five branched hydrophobic residues, although otherwise the specific amino acids can vary considerably. Several high-resolution structures of the substrate -binding domain (SBD) of DnaK bound to peptides reveal a highly conserved configuration of the bound substrate and further suggest that the substrate-binding cleft consists of five largely independent sites for interaction with five consecutive substrate residues. Importantly, both substrate backbone orientations (N- to C- and C- to N-) allow essentially the same backbone hydrogen-bonding and side-chain interactions with the chaperone. In order to rationalize these observations, we performed atomistic molecular dynamics simulations to sample the interactions of all 20 amino acid side chains in each of the five sites of the chaperone in the context of the conserved substrate backbone configurations. The resulting interaction energetics provide the basis set for deriving a predictive model that we call Paladin (Physics-based model of DnaK-Substrate Binding). Trained using available peptide array data, Paladin can distinguish binders and nonbinders of DnaK with accuracy comparable to existing predictors and further predicts the detailed configuration of the bound sequence. Tested using existing DnaK-peptide structures, Paladin correctly predicted the binding register in 10 out of 13 substrate sequences that bind in the N- to C- orientation, and the binding orientation in 16 out of 22 sequences. The physical basis of the Paladin model provides insight into the origins of how Hsp70s bind substrates with a balance of selectivity and promiscuity. The approach described here can be extended to other Hsp70s where extensive peptide array data is not available.  相似文献

The aim of this work was to explore for the existence of a physiological basis for the apparent exclusion of salmonid fish from the littoral zone of lakes in Patagonia in response to increasing water temperature, to the benefit of the native perca (Percichthys trucha). We conducted experiments on the thermal preferences of juvenile perca and rainbow trout collected in three sites of Northern Patagonia. We characterized the relationship between preferred and acclimation temperature (PT, AT) for perca and trout and analyzed between-sites variations in PTs. Both species responded with increasing PT to higher AT, but with differences in overall PT for perca from different sites (lower PTs for fish from cooler lake). In general, rainbow trout displayed lower PT and a narrower range of PT than perca. Our results provide the first comparative view of PTs of one important native fish and the main sport fish species of Patagonia. Native perca may be better equipped to cope with warmer waters than exotic rainbow trout, in agreement with their geographic distribution. Also, perca may have an advantage at the local level, particularly in littoral areas of lakes, through the alleviation of predation by and trophic competition with trout.  相似文献

A single intraperitoneal injection of endotoxin (40 microg/kg) significantly delayed gastric emptying of a solid nutrient meal. Blockade of nitric oxide synthase (NOS) with 30 mg/kg ip N(G)-nitro-L-arginine methyl ester or 20 mg/kg ip 7-nitroindazole [neuronal NOS (nNOS) inhibitor] significantly delayed gastric emptying in control animals but failed to modify gastric emptying in endotoxin-treated rats. Administration of 2.5, 5, and 10 mg/kg ip N(6)-iminoethyl-L-lysine [inducible NOS (iNOS) inhibitor] had no effect in either experimental group. Indomethacin (5 mg/kg sc), NS-398 (cyclooxygenase-2 inhibitor; 10 mg/kg ip), and dexamethasone (10 mg/kg sc) but not quinacrine (20 mg/kg ip) significantly prevented delay in gastric emptying induced by endotoxin but failed to modify gastric emptying in vehicle-treated animals. Ca(2+)-dependent NOS activity in the antrum pylorus of the stomach was diminished by endotoxin, whereas Ca(2+)-independent NOS activity was not changed. In addition, decreased nNOS mRNA and protein were observed in the antrum pylorus of endotoxin-treated rats. Our results suggest that downregulation of nNOS in the antrum pylorus of the stomach and synthesis of prostaglandins mediate the delay in gastric emptying of a solid nutrient meal induced by endotoxin.  相似文献
