Background  

The development and implementation of innovative vector control strategies for malaria control in Africa requires in-depth ecological studies in contained semi-field environments. This particularly applies to the development and release of genetically-engineered vectors that are refractory to Plasmodium infection. Here we describe a modified greenhouse, designed to simulate a natural Anopheles gambiae Giles ecosystem, and the first successful trials to complete the life-cycle of this mosquito vector therein.  相似文献

Lipid class composition of Dunaliella viridis Teodoresco was analysed using thin layer chromatography coupled with flame ionisation detection (TLC/FID technique). D. viridis was cultured under four different photon fluence rates and in darkness, and under two different conditions of CO2 supply (atmospheric and 1%) with and without nitrogen sufficiency. Nine lipid classes were identified and quantified. Total lipids per cell and acetone-mobile polar lipids decreased with light, while the percentage of sterols and triglycerides increased with increasing irradiance. Total phospholipids increase was related with growth rate while hydrocarbons, wax esters and sterol esters accumulated in darkness. There were almost no changes in total lipids per cell because of nitrogen limitation; however, nitrogen limitation led to higher changes in lipid class composition under 1% CO2 than under atmospheric CO2 levels. The main reserve lipid, triglycerides, accumulated in high amounts under 1% CO2 and nitrogen limitation, increasing from 1% to 22% of total lipids. The ratio sterols/acetone-mobile polar lipids could be an index of the 'light status' independently of nitrogen limitation, while the ratio triglycerides/total phospholipids could indicate any physiological stress uncoupling C and N metabolism and affecting the growth rate. A novel method, single-molecule anisotropy imaging, has been employed to simultaneously study lateral and rotational diffusion of fluorescence-labeled lipids on supported phospholipid membranes. In a fluid membrane composed of 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine, in which the rotational diffusion time is on the order of the excited-state lifetime of the fluorophore rhodamine, a rotational diffusion constant, D(rot) = 7 x 10(7) rad(2)/s, was determined. The lateral diffusion constant, measured by direct analysis of single-molecule trajectories, was D(lat) = 3.5 x 10(-8) cm(2)/s. As predicted from the free-volume model for diffusion, the results exhibit a significantly enhanced mobility on the nanosecond time scale. For membranes of DPPC lipids in the L(beta) gel phase, the slow rotational mobility permitted the direct observation of the rotation of individual molecules characterized by D(rot) = 1.2 rad(2)/s. The latter data were evaluated by a mean square angular displacement analysis. The technique developed here should prove itself profitable for imaging of conformational motions of individual proteins on the time scale of milliseconds to seconds.  相似文献

Trait‐based approaches are increasingly used as a proxy for understanding the relationship between biodiversity and ecosystem functioning. Macrobenthic fauna are considered one of the major providers of ecosystem functions in marine soft sediments; however, several gaps persist in the knowledge of their trait classification, limiting the potential use of functional assessments. While trait databases are available for the well‐studied North Atlantic benthic fauna, no such trait classification system exists for Australia. Here, we present the South Australian Macrobenthic Traits (SAMT) database, the first comprehensive assessment of macrobenthic fauna traits in temperate Australian waters. The SAMT database includes 13 traits and 54 trait‐modalities (e.g., life history, morphology, physiology, and behavior), and is based on records of macrobenthic fauna from South Australia. We provide trait information for more than 250 macrobenthic taxa, including outcomes from a fuzzy coding procedure, as well as an R package for using and analyzing the SAMT database. The establishment of the SAMT constitutes the foundation for a comprehensive macrobenthic trait database for the wider southern Australian region that could facilitate future research on functional perspectives, such as assessments of functional diversity and changes to ecosystem functioning.  相似文献

Most of the experimental work on the effects of ocean acidification on the photosynthesis of algae has been performed in the laboratory using monospecific cultures. It is frequently assumed that the information obtained from these cultures can be used to predict the acclimation response in the natural environment. CO(2) concentration is known to regulate the expression and functioning of the CCMs in the natural communities; however, ambient CO(2) can become quite variable in the marine ecosystems even in the short- to mid-term. We propose that the degree of saturation of the photosynthesis for a given algal community should be defined in relation to the particular characteristics of its habitat, and not only in relation to its taxonomic composition. The convenience of high CO(2) experiments to infer the degree of photosynthesis saturation by CO(2) in the natural algal communities under the present ocean conditions, as well as its trend in a coming future is discussed taking into account other factors such as the availability of light and nutrients, and seasonality.  相似文献
