Stem cells have the ability for prolonged self‐renewal and differentiation into mature cells of various lineages, which makes them important cell sources for tissue engineering applications. Their remarkable ability to replenish and differentiate in vivo is regulated by both intrinsic and extrinsic cellular mechanisms. The anatomical location where the stem cells reside, known as the “stem cell niche or microenvironment,” provides signals conducive to the maintenance of definitive stem cell properties. Physiological condition including oxygen tension is an important component of the stem cell microenvironment and has been shown to play a role in regulating both embryonic and adult stem cells. This review focuses on oxygen as a signaling molecule and the way it regulates the stem cells' development into mesenchymal tissues in vitro. The physiological relevance of low oxygen tension as an environmental parameter that uniquely benefits stem cells' expansion and maintenance is described along with recent findings on the regulatory effects of oxygen on embryonic stem cells and adult mesenchymal stem cells. The relevance to tissue engineering is discussed in the context of the need to specifically regulate the oxygen content in the cellular microenvironment in order to optimize in vitro tissue development. © 2009 American Institute of Chemical Engineers Biotechnol. Recently, it has been demonstrated that loops of the crystallizable fragment of IgG1 (IgG1-Fc) can be engineered to form antigen-binding sites. In this work C-terminal structural loops in the CH3 domains of homodimeric IgG1-Fc have been functionalized to form integrin-binding sites in order to probe the effect of engineering on structural integrity and thermal stability of IgG1-Fc as well as on binding to the ligands Protein A, CD16 and FcRn, respectively. The peptide sequence GCRGDCL - a disulfide-bridged cyclic heptapeptide that confers binding to human αvβ3 integrin was introduced into AB, CD and/or EF loops and single and double mutants were heterologously expressed in Pichia pastoris. Integrin binding of engineered IgG-Fc was tested using both binding to coated αvβ3 integrin in ELISA or to αvβ3-expressing K562 cells in FACS analysis. Additionally, blocking of αvβ3-mediated cell adhesion to vitronectin was investigated. The data presented in this report demonstrate that bioactive integrin-binding peptide(s) can be grafted on the C-terminal loops of IgG-Fc without impairing binding to effector molecules. Observed differences between the investigated variants in structural stability and integrin binding are discussed with respect to the known structure of IgG-Fc and its structural loops. 相似文献

Wheat plants were grown from sowing to day 18 in 26-dm3 chambers at three different CO2 concentrations: 150 (-CO2), 350 (C, control), 800 (+CO2) mol mol-1. Afterwards, plants of the three variants were grown at the same natural CO2 concentration. Plant characteristics were measured just before the transfer (0 days after CO2 treatment, DAT), and at 5 – 8 DAT on the 1st leaf, and at 12 – 22 DAT on the 4th leaf. Decreased or increased CO2 concentrations caused acclimations which persisted after transplantation to natural CO2 concentration. At 5 – 8 DAT, stomatal density, stomatal conductance (gs), CO2 saturated net photosynthetic rate (PNsat0), radiation saturated net photosynthetic rate (PNsat1), and carboxylation efficiency () were higher in -CO2 plants and lower in +CO2 plants than in C plants. As compared with C plants, the photochemical efficiency () was lower in -CO2 and higher in -CO2 plants, however, chlorophyll (Chl) a, Chl b, Chl a–b and carotenoid contents were lower in both -CO2 and +CO2 plants. On the 4th leaf, which emerged on plant after finishing CO2 treatments, at 12 – 22 DAT, no differences in stomatal density and g, between treatments were observed. In -CO2 plants, pigment content and PNsat0 were higher, was lower, and PNsat1 and were not different from C plants. In contrast, in +CO2 plants, pigment content, PNsat1 and were lower, and PNsat0 and were unchanged. Leaf area, dry mass, and tiller development increased in +CO2 plants and decreased in -CO2 plants. In the interval between 8 and 22 DAT, lower net assimilation rate in +CO2 than in -CO2 plants was observed. 相似文献

The genus Impatiens (Balsaminaceae) includes three widespread species in the Czech Republic, central Europe: the native I. noli-tangere, and two invasive species, I. parviflora and I. glandulifera, differing in the dynamics of invasion. They all occur in similar habitats and share basic life-history characteristics, which make them a suitable model for studying species traits associated with invasiveness. In this study we investigated differences in habitat requirements of these Impatiens species, their coexistence and short-term population dynamics in the field. We established 84 1 × 1 m permanent plots in five localities where all three species co-occurred. In each plot vascular plant species were determined, their cover estimated and all individuals of Impatiens species counted. Site characteristics including tree canopy cover, soil moisture, nitrogen and carbon content, and slope were measured directly. Nutrients, light, humidity and soil reaction were estimated using Ellenberg indicator values. The presence of I. noli-tangere was strongly correlated with high soil moisture, that of I. parviflora with high tree canopy cover and low soil moisture. Impatiens glandulifera exhibited a unimodal response to tree canopy cover, avoiding both very shaded and fully open sites. The current-year abundances of all species were negatively related to those of congeneric species. These results suggest that the coexistence of Impatiens species in the same habitat is due to microsite differentiation. Further spread of I. glandulifera to new habitats, and reduction of the native I. noli-tangere niche, can be expected in areas where the latter species co-occurs with competitively strong invasive congeners. 相似文献

Although the evolutionary drivers of genome size change are known, the general patterns and mechanisms of plant genome size evolution are yet to be established. Here we aim to assess the relative importance of proliferation of repetitive DNA, chromosomal variation (including polyploidy), and the type of endoreplication for genome size evolution of the Pleurothallidinae, the most species-rich orchid lineage. Phylogenetic relationships between 341 Pleurothallidinae representatives were refined using a target enrichment hybrid capture combined with high-throughput sequencing approach. Genome size and the type of endoreplication were assessed using flow cytometry supplemented with karyological analysis and low-coverage Illumina sequencing for repeatome analysis on a subset of samples. Data were analyzed using phylogeny-based models. Genome size diversity (0.2–5.1 Gbp) was mostly independent of profound chromosome count variation (2n = 12–90) but tightly linked with the overall content of repetitive DNA elements. Species with partial endoreplication (PE) had significantly greater genome sizes, and genomic repeat content was tightly correlated with the size of the non-endoreplicated part of the genome. In PE species, repetitive DNA is preferentially accumulated in the non-endoreplicated parts of their genomes. Our results demonstrate that proliferation of repetitive DNA elements and PE together shape the patterns of genome size diversity in orchids. 相似文献

Herein we report an assessment of 24 1,2,3,4-tetrahydroisoquinoline derivatives for potential DNase I (deoxyribonuclease I) inhibitory properties in vitro. Four of them inhibited DNase I with IC50 values below 200 μM. The most potent was 1-(6,7-dimethoxy-1,2,3,4-tetrahydroisoquinolin-1-yl)propan-2-one ( 2 ) (IC50=134.35±11.38 μM) exhibiting slightly better IC50 value compared to three other active compounds, 2-[2-(4-fluorophenyl)-1,2,3,4-tetrahydroisoquinolin-1-yl]-1-phenylethan-1-one ( 15 ) (IC50=147.51±14.87 μM), 2-[2-(4-fluorophenyl)-1,2,3,4-tetrahydroisoquinolin-1-yl]cyclohexan-1-one ( 18 ) (IC50=149.07±2.98 μM) and 2-[6,7-dimethoxy-2-(p-tolyl)-1,2,3,4-tetrahydroisoquinolin-1-yl]cyclohexan-1-one ( 22 ) (IC50=148.31±2.96 μM). Cytotoxicity assessment of the active DNase I inhibitors revealed a lack of toxic effects on the healthy cell lines MRC-5. Molecular docking and molecular dynamics simulations suggest that interactions with Glu 39, His 134, Asn 170, Tyr 211, Asp 251 and His 252 are an important factor for inhibitors affinity toward the DNase I. Observed interactions would be beneficial for the discovery of new active 1,2,3,4-tetrahydroisoquinoline-based inhibitors of DNase I, but might also encourage researchers to further explore and utilize potential therapeutic application of DNase I inhibitors, based on a versatile role of DNase I during apoptotic cell death. 相似文献

We characterized the photoautotrophic growth of glucose‐tolerant Synechocystis sp. PCC 6803 in a flat‐panel photobioreactor running on a semicontinuous regime under various lights, temperatures, and influx carbon dioxide concentrations. The maximum reached growth rate was 0.135 h?1, which corresponds to a doubling time of 5.13 h—a growth speed never reported for Synechocystis before. Saturating red light intensity for the strain was 220–360 μmol(photons) m?2 s?1, and we did not observe any photoinhibition up to 660 μmol(photons) m?2 s?1. Synechocystis was able to grow under red light only; however, photons of wavelengths 405–585 and 670–700 nm further improved its growth. Optimal growth temperature was 35°C. Below 32°C, the growth rates decreased linearly with temperature coefficient (Q10) 1.70. Semicontinuous cultivation is known to be efficient for growth characterization and optimization. However, the assumption of correct growth rates calculation—culture exponential growth—is often not fulfilled. The semicontinuous setup in this study was operated as a turbidostat. Accurate online OD measurements with high time‐resolution allowed fast and reliable growth rates determination. Repeating diluting frequencies (up to 18 dilutions per day) were essential for rapid growth stability evaluation. The presented setup provides improvement to previously published semicontinuous characterization strategies by decreasing experimental time requirements and maintaining the culture in exponential growth phase throughout the entire characterization procedure. 相似文献
