Switchgrass (Panicum virgatum L.) may have value as forage and a bioenergy feedstock. Our objective was to evaluate how harvest system and N fertilizer rates affected biomass yield and nutrient composition of young stands of switchgrass (cv. Alamo) in the southern Great Plains, USA. Nitrogen fertilization increased biomass yields from 10.4, 10.8, and 12.2 Mg ha?1 at 0 kg N?ha?1 to 13.7, 14.6, and 21.0 Mg ha?1 at 225 kg N?ha?1 when harvested after seed set (October), after frost (December), and twice per year after boot stage (July) and frost, respectively. Nutrient concentrations and removal were generally twice as great when biomass was harvested twice versus once per year. Precipitation strongly affected biomass yields across the two years of these experiments. When late-summer precipitation is available to support regrowth in this environment, harvesting switchgrass twice per year will result in greater biomass yields. Harvesting twice per year, however, will increase fertilization requirements and reduce feedstock biomass quality. Switchgrass harvested during mid-summer after boot stage was of poor forage quality. To have value as a dual-purpose forage and bioenergy feedstock, switchgrass would need to be utilized during spring to early summer while in a vegetative stage.  相似文献

Target of rapamycin (TOR) is a central regulator of cell growth, cell death, nutrition, starvation, hormone, and stress responses in diverse eukaryotes. However, very little is known about TOR signaling and the associated functional domains in plants. We have taken a genetic approach to dissect TOR functions in Arabidopsis (Arabidopsis thaliana) and report here that the kinase domain is essential for the role of TOR in embryogenesis and 45S rRNA expression. Twelve new T-DNA insertion mutants, spanning 14.2 kb of TOR-encoding genomic region, have been characterized. Nine of these share expression of defective kinase domain and embryo arrest at 16 to 32 cell stage. However, three T-DNA insertion lines affecting FATC domain displayed normal embryo development, indicating that FATC domain was dispensable in Arabidopsis. Genetic complementation showed that the TOR kinase domain alone in tor-10/tor-10 mutant background can rescue early embryo lethality and restore normal development. Overexpression of full-length TOR or kinase domain in Arabidopsis displayed developmental abnormalities in meristem, leaf, root, stem, flowering time, and senescence. We further show that TOR, especially the kinase domain, plays a role in ribosome biogenesis by activating 45S rRNA production. Of the six putative nuclear localization sequences in the kinase domain, nuclear localization sequence 6 was identified to confer TOR nuclear targeting in transient expression assays. Chromatin immunoprecipitation studies revealed that the HEAT repeat domain binds to 45S rRNA promoter and the 5' external transcribed spacer elements motif. Together, these results show that TOR controls the embryogenesis, postembryonic development, and 45S rRNA production through its kinase domain in Arabidopsis.  相似文献

Knowing threshold changes in brain lipids and lipid enzymes during dietary n-3 polyunsaturated fatty acid deprivation may elucidate dietary regulation of brain lipid metabolism. To determine thresholds, rats were fed for 15 weeks DHA-free diets having graded reductions of α-linolenic acid (α-LNA). Compared with control diet (4.6% α-LNA), plasma DHA fell significantly at 1.7% dietary α-LNA while brain DHA remained unchanged down to 0.8% α-LNA, when plasma and brain docosapentaenoic acid (DPAn-6) were increased and DHA-selective iPLA2 and COX-1 activities were downregulated. Brain AA was unchanged by deprivation, but AA selective-cPLA2, sPLA2 and COX-2 activities were increased at or below 0.8% dietary α-LNA, possibly in response to elevated brain DPAn-6. In summary, homeostatic mechanisms appear to maintain a control brain DHA concentration down to 0.8% dietary DHA despite reduced plasma DHA, when DPAn-6 replaces DHA. At extreme deprivation, decreased brain iPLA2 and COX-1 activities may reduce brain DHA loss.  相似文献

Thirty-four Vibrio cholerae isolates collected from a cholera outbreak in Hyderabad, South India were found to belong to serogroup Ol biotype El Tor serotype Ogawa. The genotype of all the isolates was confirmed by PCR assays. All the isolates were found PCR positive for ctxAB, ompW, rflOl, rtxC, and tcpA genes. All the isolates but one harboured rstR El Tor allele. However, one isolate carried both rstR EL Tor as well as rstR Classical alleles. Cholera toxin (ctxB) genotyping of the isolates confirmed the presence of altered cholera toxin B of classical biotype in all the isolates. All the isolates except VCH35 harboured an RS1-CTX prophage array on the large chromosome. The isolate VCH35 contained a tandem repeat of classical CTX prophage on the small chromosome. The clonal relationship among the V. cholerae isolates as carried out by enterobacterial repetitive intergenic consensus sequences PCR, BOX PCR and randomly amplified polymorphic DNA, uniformly showed a genetic relationship among the outbreak isolates. The results of this study suggest that altered El Tor biotype V. cholerae with the classical cholera toxin gene are involved in cholera outbreaks in India.  相似文献

Mixed infections are one of the major therapeutic challenges, as the current strategies have had limited success. One of the most common and widespread conditions of mixed infection is respiratory syncytial virus-mediated pathology of the respiratory tract in children. There is a dire need for the development of novel therapeutic approaches during mixed infections. Therapeutic intravenous immunoglobulin preparations, obtained from plasma pools of healthy donors have been used in immune deficiencies. This study was thus designed to characterize the functional efficacy of RSV-specific antibodies in IVIg. To explore the functional ability of these affinity-purified RSV-specific antibodies, the antibody-dependent and complement dependent cytotoxicity was determined using peripheral cells of healthy donors. This study demonstrates the existence of highly potent RSV-specific antibodies in IVIg preparations and provides the basis for the use of IVIg as broad-spectrum protective shield to RSV-infected children during mixed infections.  相似文献

Biomass demand for energy will lead to utilization of marginal, low fertility soil. Application of fertilizer to such soil may increase switchgrass (Panicum virgatum L.) biomass production. In this three-way factorial field experiment, biomass yield response to potassium (K) fertilizer (0 and 68 kg?K?ha?1) on nitrogen (N)-sufficient and N-deficient switchgrass (0 and 135 kg?N?ha?1) was evaluated under two harvest systems. Harvest system included harvesting once per year after frost (December) and twice per year in summer (July) at boot stage and subsequent regrowth after frost. Under the one-cut system, there was no response to N or K only (13.4 Mg?ha?1) compared to no fertilizer (12.4 Mg?ha?1). Switchgrass receiving both N and K (14.6 Mg?ha?1) produced 18 % greater dry matter (DM) yield compared to no fertilizer check. Under the two-cut harvest system, N only (16.0 Mg?ha?1) or K only (14.1 Mg?ha?1) fertilizer produced similar DM to no fertilizer (15.1 Mg?ha?1). Switchgrass receiving both N and K in the two-cut system (19.2 Mg?ha?1) produced the greatest (P?<?0.05) DM yield, which was 32 % greater than switchgrass receiving both N and K in the one-cut system. Nutrient removal (biomass?×?nutrient concentration) was greatest in plots receiving both N and K, and the two-cut system had greater nutrient removal than the one-cut system. Based on these results, harvesting only once during winter months reduces nutrient removal in harvested biomass and requires less inorganic fertilizer for sustained yields from year to year compared to two-cut system.  相似文献

Paradoxical tuberculosis-associated immune reconstitution inflammatory syndrome (TB-IRIS) is an aberrant inflammatory response occurring in a subset of TB-HIV co-infected patients initiating anti-retroviral therapy (ART). Here, we examined monocyte activation by prospectively quantitating pro-inflammatory plasma markers and monocyte subsets in TB-HIV co-infected patients from a South Indian cohort at baseline and following ART initiation at the time of IRIS, or at equivalent time points in non-IRIS controls. Pro-inflammatory biomarkers of innate and myeloid cell activation were increased in plasma of IRIS patients pre-ART and at the time of IRIS; this association was confirmed in a second cohort in South Africa. Increased expression of these markers correlated with elevated antigen load as measured by higher sputum culture grade and shorter duration of anti-TB therapy. Phenotypic analysis revealed the frequency of CD14++CD16 monocytes was an independent predictor of TB-IRIS, and was closely associated with plasma levels of CRP, TNF, IL-6 and tissue factor during IRIS. In addition, production of inflammatory cytokines by monocytes was higher in IRIS patients compared to controls pre-ART. These data point to a major role of mycobacterial antigen load and myeloid cell hyperactivation in the pathogenesis of TB-IRIS, and implicate monocytes and monocyte-derived cytokines as potential targets for TB-IRIS prevention or treatment.  相似文献
