Attempts were made to develop a simplified procedure for long-term cryopreservation of intestinal smooth muscle cells (ISMC). ISMC were collected from the ileum of Sprague-Dawley neonatal rats through cellular dissociation in trypsin. Cryopreservation method comprised of a rapid 1-step (protocol 1) and a slow 3-step (protocol 2) freezing of ISMC for 1 week. Preparations were thawed and single ISMC were assessed via the comet assay and damaged DNA was quantified through comet tail moment. The control unfrozen ISMC exhibited DNA damage of 2.34 ± 0.35 compared to ISMC cooled via protocol 2 (2.62 ± 0.36) and protocol 1 (10.15 ± 0.72). Thereafter, protocol 2 freezing method was adopted and ISMC were cryopreserved for 1-week, 1-month, and 4-months to analyse the temporal and long-term cryopreservation of ISMC. This revealed a DNA damage of 2.62 ± 0.36 (1-week), 3.81 ± 0.72 (1-month), and 5.1 ± 0.9 (4-months). Gradual cooling is suitable for continuing storage of ISMC and although fluctuation in cryoinjury is observed with time this is considered to reflect cell-to-cell variability.  相似文献

CD4+CD25+FoxP3+ regulatory T cells (Tregs) are key players for maintaining immune tolerance and for reducing the inflammation‐mediated tissue damage following infection. However, Tregs also suppress protective immune responses to pathogens (including virus, bacteria, parasites, and fungi) and vaccines and enhance pathogen persistence by inhibiting the activation and functions of both innate and adaptive immune cells such as dendritic cells, macrophages, and T and B lymphocytes and by promoting immunosuppressive environment. Therefore, equilibrium in the Treg number and function is important to ensure pathogen clearance and protection from infection‐associated immunopathologies. Recent advances in understanding of Treg influence on the outcome of infection opened new avenues to target them. Various small molecules, pharmacological inhibitors, monoclonal antibodies that target Tregs provided proof of concept in experimental models. The field also benefits from advances in other subjects, particularly oncology and autoimmunity, where Treg‐targeted therapies are exploited in the clinic to a greater extent. The future research should aim at translating this preclinical success to human application.  相似文献

The silkworm B. mori is a multicellular organism revealing genetic resources which makes an ideal model for lepidoptera for the present investigation. With the objective of targeting distinctive markers for utilization in future breeding programmes, Bivoltine and Polyvoltine silkworm strains were used by inter-simple sequence repeats (ISSR) and random amplified polymorphic-DNA (RAPD) fingerprinting to detect their genetic versatility and volatility. Six ISSR primers generated 99 markers, of which 76.76% were found to be polymorphic with an average number of observed alleles (N a) (1.86 ± 0.40), an effective number of alleles (N e) (1.43 ± 0.30) as well as six RAPD primers that produced a total of 95 bands, developing 61.05% polymorphism with N a (1.93 ± 0.51) and N e (1.18 ± 0.30). The dendrogram produced by UPGMA analysis, based on Dice’s coefficient, clustered four races into two major groups which accurately segregated them according to their inheritance of voltinism. In this research, the ISSR markers were more accurate than the RAPD markers and ISSR also displayed better polymorphism. The outcome showed that the bivoltine strains exhibited higher allelic expressions with ISSR primers when compared to the polyvoltine strains. Despite exhibiting their unique race by certain DNA markers, most of the primers represented voltinism-specificity. Hence molecular marker amplification is a beneficial approach to reveal genetic divergence among closely related strains, and molecular characterization of phylogenetic relationships in addressing evolutionary evidences of individuals.  相似文献

The availability of complete, annotated genomic sequence information in model organisms is a rich resource that can be extended to understudied orphan crops through comparative genomic approaches. We report here a software tool (cisprimertool) for the identification of conserved intron scanning regions using expressed sequence tag alignments to a completely sequenced model crop genome. The method used is based on earlier studies reporting the assessment of conserved intron scanning primers (called CISP) within relatively conserved exons located near exon-intron boundaries from onion, banana, sorghum and pearl millet alignments with rice. An unintended consequence of global industrialization and associated societal rearrangements is new interactions of microbes and potential hosts (especially mammals and plants), providing an opportunity for the rapid emergence of host-microbe adaptation and eventual establishment of new microbe-related diseases. We describe a new model system comprising the model plant Arabidopsis thaliana and several microbes, each representing different modes of interaction, to study such “maladaptations”. The model microbes include human and agricultural pathogens and microbes that are commonly considered innocuous. The system has a large knowledge base corresponding to each component organism and is amenable to high-throughput automation assisted perturbation screens for identifying components that modulate host-pathogen interactions. This would aid in the study of emergence and progression of host-microbe maladaptations in a controlled environment.  相似文献

Nanofibrous glucose electrodes were fabricated by the immobilization of glucose oxidase (GOx) into an electrospun composite membrane consisting of polymethylmethacrylate (PMMA) dispersed with multiwall carbon nanotubes (MWCNTs) wrapped by a cationic polymer (poly(diallyldimethylammonium chloride) (PDDA)) and this nanofibrous electrode (NFE) is abbreviated as PMMA-MWCNT(PDDA)/GOx-NFE. The NFE was characterized for morphology and electroactivity by using electron microscopy and cyclic voltammetry, respectively. Field emission transmission electron microscopy (FETEM) image reveals the dispersion of MWCNT(PDDA) within the matrix of PMMA. Cyclic voltammetry informs that NFE is suitable for performing surface-confined electrochemical reactions. PMMA-MWCNT(PDDA)/GOx-NFE exhibits excellent electrocatalytic activity towards hydrogen peroxide (H(2)O(2)) with a pronounced oxidation current at +100 mV. Glucose is amperometrically detected at +100 mV (vs. Ag/AgCl) in 0.1M phosphate buffer solution (PBS, pH 7). The linear response for glucose detection is in the range of 20 microM to 15 mM with a detection limit of 1 microM and a shorter response time of approximately 4 s. The superior performance of PMMA-MWCNT(PDDA)/GOx-NFE is due to the wrapping of PDDA over MWCNTs that binds GOx through electrostatic interactions. As a result, an effective electron mediation is achieved. A layer of nafion is made over PMMA-MWCNT(PDDA)/GOx-NFE that significantly suppressed the electrochemical interference from ascorbic acid or uric acid. In all, PMMA-MWCNT(PDDA)/GOx-nafion-NFE has exhibited excellent properties for the sensitive determination of glucose like high selectivity, good reproducibility, remarkable stability and without interference from other co-existing electroactive species.  相似文献
