Bovine and human lens membrane, when reconstituted into lipid vesicles containing oxidized cytochrome C, will mediate the transmembrane passage of ascorbate into the vesicles, where the reduction of cytochrome C is measured spectrophotometrically. This channel forming activity is specifically inhibited by antiserum made against a synthetic octapeptide near the C-terminus of MIP26K. Together, these studies describe a direct and more sensitive assay system for measurement of channel-forming activity of MIP26K, and suggest that the C-terminus of this molecule may be particularly important in the regulation of channel formation.  相似文献

The relative importance of chylomicrons (Sf greater than 400) and very low density lipoproteins (Sf 20--400) in transporting lipids in lymph was investigated in surgically prepared adult sheep and pre-ruminant lambs fed low fat diets or infused intraduodenally with corn oil. The concentration of triacylglycerol in the intestinal lymph of sheep and lambs was increased from 520 and 925 mg/100 ml to 2326 and 2367 mg/100 ml respectively when corn oil was infused into the duodenum and the ratio of triacylglycerol to phospholipid changed from 3.7 and 5.5 to 9.5 and 9.7 respectively. The flow of lymph also increased. Electron microscopy and analytical and preparative ultracentrifugation showed that lymph lipoproteins from sheep and lambs fed low fat diets consisted mainly of lipoproteins 50 nm in diameter and that very low density lipoproteins (Sf 20--400) contirbuted up to 75% of the Sf greater than 20 lipoproteins. There were no lipoproteins with diameters above 150 nm. Infusion of corn oil into the duodenum of sheep and lambs increased the diameters of lymph lipoproteins. Most were 80--100 nm in diameter but substantial numbers above 150 and up to 400 mn were observed. The maximum contribution of very low density lipoproteins (Sf 20--400) to lipoproteins of Sf greater than 20 was 27--30%. The above findings demonstrate that the size of intestinal lymph lipoprotein particles increases with the amount of lipid absorbed from the small intestines and that the transport of lymph lipids, in ruminants, is similar to that previously found in rats, rabbits and man.  相似文献

The effect of different nitrogen compounds on the induction of reduced nicotinamide adenine dinucleotide phosphate-nitrate reductase was examined in Neurospora crassa. Whereas in the wild-type strain several amino acids and ammonia inhibit the formation of nitrate reductase, only glutamine, cysteine, and histidine are shown to inhibit the synthesis of nitrate reductase in a glutamine-requiring auxotroph. None of the amino acids inhibited nitrate reductase activity in vitro. The effects of cysteine and histidine are nonspecific, these amino acids being inhibitory of the growth of the organism. The effect of glutamine on the induction of nitrate reductase is not due to an inhibition of the uptake of the inducer nitrate. By the use of histidine-, pyrimidine-, and arginine-requiring auxotrophs, it was shown that glutamine appears to act per se and does not seem to be converted to another product in order to be effective in repression. The repression of nitrate reductase by ammonia appears, from the results described herein, to be indirect; ammonia has to be converted first to glutamine in order to be effective in repression.  相似文献

Mutations in the metabolic enzyme isocitrate dehydrogenase 1 (IDH1) are commonly found in gliomas. AGI-5198, a potent and selective inhibitor of the mutant IDH1 enzyme, was radiolabeled with radioiodine and fluorine-18. These radiotracers were evaluated as potential probes for imaging mutant IDH1 expression in tumors with positron emission tomography (PET). Radioiodination of AGI-5198 was achieved using a tin precursor in 79?±?6% yield (n?=?9), and 18F-labeling was accomplished by the Ugi reaction in a decay-corrected radiochemical yield of 2.6?±?1.6% (n?=?5). The inhibitory potency of the analogous nonradioactive compounds against mutant IDH1 (IDH1-R132H) was determined in enzymatic assays. Cell uptake studies using radiolabeled AGI-5198 analogues revealed somewhat higher uptake in IDH1-mutated cells than that in wild-type IDH1 cells. The radiolabeled compounds displayed favorable tissue distribution characteristics in vivo, and good initial uptake in IDH1-mutated tumor xenografts; however, tumor uptake decreased with time. Radioiodinated AGI-5198 exhibited higher tumor-to-background ratios compared with 18F-labeled AGI-5198; unfortunately, similar results were observed in wild-type IDH1 tumor xenografts as well, indicating lack of selectivity for mutant IDH1 for this tracer. These results suggest that AGI-5198 analogues are not a promising platform for radiotracer development. Nonetheless, insights gained from this study may help in design and optimization of novel chemical scaffolds for developing radiotracers for imaging the mutant IDH1 enzyme.  相似文献

Responses to daily injections of bovine growth hormone (GH, 0.15 mg kg-1 liveweight), beginning on day 10 of lactation, were measured in lactating ewes. Milk yields of GH-treated ewes increased soon after commencement of injections and continued to increase for some 25 days before reaching plateau levels. By comparison, yields of ewes injected with excipient (controls) decreased over the experiment. There was a tendency for contents of milk fat to be higher and milk protein to be lower for GH-treated than for control ewes during the first 15-20 days after injections were started. At the beginning and over the first 15-20 days of the experiment feed intakes of both groups of ewes were similar, but thereafter intakes of GH-treated ewes gradually increased to reach plateau levels some 200-300 g day-1 higher than for control ewes by about day 35. Liveweights of both groups of ewes decreased during the first 2 weeks of treatment then increased, with GH-treated ewes losing, then gaining, more weight than control ewes. The efficiency of food utilization for milk production was higher for GH-treated than control ewes throughout the experiment but digestibility of food organic matter was not different during the eighth week of the experiment. At the end of the experiment, body composition, assessed by dilution of tritiated water, was similar for both groups of ewes. Differences in milk production were not sustained after withdrawal of GH injections. Measurements of tissue uptake of key metabolites were made on days 3 and 45 of GH treatment. On day 3, GH lowered uptake of glucose and non-esterified fatty acids by leg muscle tissue and increased mammary uptake of non-esterified fatty acids. By day 45 there were no apparent differences of tissue uptake of key metabolites. The results indicate that there is a biphasic response to exogenous GH in the lactating ruminant. It appears that initially GH affects nutrient partition thereby increasing supplies to the mammary gland of key nutrients for milk synthesis. In the longer term, GH increases feed intake, which provides sufficient nutrients to sustain increased milk production and also liveweight gain.  相似文献

Invasive plants significantly threaten native plant biodiversity, yet the mechanisms by which they drive species losses and maintain their own dominance are poorly known. We examined the effects of alien grass invasion (Stenotaphrum secundatum) on (1) abundance and frequency of occurrence, (2) reproductive effort (flowering) and output (fruit production) and (3) soil seed banks for three focal native plants that are characteristic of endangered coastal forest of south-eastern Australia. First, we sampled and compared the foliage cover abundance and frequency (proportion of sites occupied) of the focal natives across invaded and non-invaded (reference) sites (n = 20). We then intensively sampled reproductive effort and output (range of 5–9 sites per species), and density of propagules within the soil (using a standard glasshouse ‘emergence’ method; n = 26) for each species. Invasion was associated with reduced population sizes of all species within the standing vegetation but did not affect population frequency (i.e. proportion of sites where each species was present). Reproductive effort and output were about 75 % lower at invaded than native sites for all species. However, invasion had no effect on propagule densities of the focal natives within the seed bank, despite the substantial reduction in their reproduction. This indicates that the ultimate driver of population declines across invaded landscapes is post-settlement recruitment limitation from the seed bank (e.g. low rates of germination and seedling survival) rather than a reduction in the arrival and storage of propagules at invaded sites. Removal of Stenotaphrum alone might thus be sufficient to stimulate the recovery of native populations from the seed bank.  相似文献

Purpose  Ovarian cancer patients with intra-tumoral CD3+ T-lymphocytes in primary tumor tissue have a better prognosis. This study aims to analyze the presence and relative influence of three important T-lymphocyte subsets, tumor-infiltrating CD8+ cytotoxic T-lymphocytes (CTL), CD45R0+ memory T-lymphocytes, and FoxP3+ regulatory T-lymphocytes (Treg), in primary tumor tissue and omental metastases of patients with ovarian cancer. Experimental design  The number of CD8+, CD45R0+, and FoxP3+ T-lymphocytes was determined by immunohistochemistry on a tissue micro array containing ovarian tumor tissue and/or omental metastases obtained at primary debulking surgery from 306 FIGO stage I–IV ovarian cancer patients. Immunohistochemistry data were correlated to clinicopathological parameters and survival data. Results  High number of CD8+ CTL and a high CD8+/FoxP3+ ratio in ovarian-derived tumor tissue were associated with increased disease-specific survival and proved to be independent prognostic factors in multivariate analyses. In advanced stage patients, the presence of CD8+ CTL, CD45R0+ memory T-lymphocytes, FoxP3+ Treg or a high CD8+/FoxP3+ ratio in ovarian-derived tumor tissue was associated with an increased disease specific survival in univariate analysis, as was the presence of CD45R0+ memory T-lymphocytes and FoxP3+ Treg in omental metastases. Furthermore, in advanced stage patients CD8+ cytotoxic and FoxP3+ regulatory T-lymphocytes infiltrating ovarian-derived tumor tissue were independent predictors of increased prognosis. Conclusions  T-lymphocytes infiltrating primary and metastatic ovarian cancer sites are associated with improved prognosis. These associations are especially distinct in advanced stage patients, underlining the potential for immunotherapy as a broadly applicable therapeutic strategy. Ninke Leffers and Marloes J. M. The Hedgehog signaling pathway plays an essential role in embryo development and adult tissue homeostasis, in regulating stem cells and is abnormally activated in many cancers. Given the importance of this signaling pathway, we developed a novel and versatile high-throughput, cell-based screening platform using confocal imaging, based on the role of β-arrestin in Hedgehog signal transduction, that can identify agonists or antagonist of the pathway by a simple change to the screening protocol. Here we report the use of this assay in the antagonist mode to identify novel antagonists of Smoothened, including a compound (A8) with low nanomolar activity against wild-type Smo also capable of binding the Smo point mutant D473H associated with clinical resistance in medulloblastoma. Our data validate this novel screening approach in the further development of A8 and related congeners to treat hedgehog related diseases, including the treatment of basal cell carcinoma and medulloblastoma.  相似文献
