Background  

Trehalose synthase (TreS) which converts maltose to trehalose is considered to be a potential biocatalyst for trehalose production. This enzymatic process has the advantage of simple reaction and employs an inexpensive substrate. A retrovirus homologue gene of cellular cyclin D 1 , walleye dermal sarcoma virus rv-cyclin gene (orf A or rv-cyclin), was expressed in the livers of zebrafish under the control of liver fatty acid-binding protein (lfabp) promoter. To prevent possible fatality caused by overexpression of the oncogene, the GAL4/upstream activation sequence (GAL4/UAS) system was used to maintain the transgenic lines. Thus, both GAL4-activator [Tg(lfabp:GAL4)] and UAS-effector [Tg(UAS:rvcyclin)] lines were generated, and the rv-cyclin gene was activated in the liver after crossing these two lines. Since no obvious neoplasia phenotypes were observed in the double-transgenic line, cancer susceptibility of the transgenic fish expressing rv-cyclin was tested by carcinogen treatment. Unexpectedly, transgenic fish expressing rv-cyclin gene (rvcyclin+) were more resistant to the carcinogen than siblings not expressing this gene (rvcyclin–). Lower incidences of multiple and malignant liver tumors were observed in rvcyclin+ than in rvcyclin– fish, and the liver tumors in the rvcyclin+ group appeared later and were less malignant. These results suggest that expression of rv-cyclin protects the fish liver from carcinogen damage and delays onset of malignancy. These findings indicate that transgenic fish models are powerful systems for investigating mechanisms of inhibition and regression of liver tumors.  相似文献

Surface-exposed pneumococcal virulence proteins pneumococcal surface protein A (PspA) and pneumococcal surface protein C (PspC) play important roles in the pathogenesis of invasive pneumococcal diseases. Human neutrophils are principle antimicrobial effector cells of the innate and adaptive immune systems. In this study, we investigated the effects of PspA and PspC on the up-regulation of chemokine CXCL8 in human neutrophils, and characterized the underlying intracellular signaling pathways. Both PspA and PspC were found to induce the release of newly synthesized CXCL8. Synergistic effect was observed in the combined treatment of PspA and PspC on the release of CXCL8. Products from PspA-deficient or PspC-deficient mutant pneumococcus that did not express PspA or PspC induced significantly less release of CXCL8 than wild type pneumococcus. Both PspA and PspC could activate p38 MAPK and NF-κB pathways in neutrophils, while inhibition of NF-κB and p38 MAPK could suppress the release of CXCL8 from neutrophils induced by PspA and PspC. Together, our results demonstrated that the induction of CXCL8 in human neutrophils activated by PspA and PspC was regulated by p38 MAPK and NF-κB pathways.  相似文献

A novel series of Akt/PKB inhibitors derived from a screening lead (1) has been prepared. The novel trans-3,4'-bispyridinylethylenes described herein are potent inhibitors of Akt/PKB with IC(50) values in the low double-digit nanomolar range against Akt1. Compound 2q shows excellent selectivity against distinct families of kinases such as tyrosine kinases and CAMK, and displays poor to modest selectivity against closely related kinases in the AGC and CMGC families. The cellular activities including inhibition of cell growth and phosphorylation of downstream target GSK3 are also described. The X-ray structure of compound 2q complexed with PKA in the ATP binding site was determined.  相似文献

In this article, RAPD was employed to detect the DNA polymorphism of Carchesium polypinum in three lakes: Nanhu Lake (N), Shahu Lake (S) and Donghu Lake (D). The results were used to deduce the definition of the populations: (1) The 20 primers produced 172 clear bands with an average of 8.6 fragments ranging from 100bp to 1600bp. The other 11 primers produced 59 legible bands with an average of 5.4 fragments ranging from 100bp to 1 500bp; (2) SPSS 12.0 and RAPDistance 1.04 that had similar trees were composed of two parallel main branches. Samples from Shijizhong (Z) of D, Shuiguohu (H) of D and N clustered into a branch and another sample from D — Fengguangcun (F) — gathered with S or made another main branch itself. The result that the three samples from D were separated by the samples from other lakes suggested that the individuals of C. polypinum in D belong to several populations. This discovery will promote micro-hydrobiont population ecology research. In addition, it will help in the study of the essence of the transitional zone and its dispersal and vicariance behavior. We believe that this study will contribute to the study of population differentiation and the mechanism of speciation.  相似文献

不同大气校正方法对森林叶面积指数遥感估算影响的比较   总被引:5,自引:1,他引:4  
利用TM原始图像以及经过6S模型和基于影像自身的Gilabert模型大气校正后的地面绝对反射率图像,分别计算了褒河流域阔叶林和针阔混交林2种林型的5类光谱植被指数(SR、NDVI、MNDVI、ARVI和RSR),并建立各林型森林叶面积指数与同时相的各个植被指数的相关关系。结果表明,2种大气校正模型均显著提高了各植被指数与森林叶面积指数的相关关系,除了对森林叶面积指数与植被指数SR和NDVI的相关关系影响不显著外,对森林叶面积指数与植被指数MNDVI、ARVI和RSR相关关系的影响均非常显著。说明不同大气校正模型对叶面积指数的遥感估算结果有较大影响。因此,在利用遥感数据进行定量分析、信息提取和生态遥感应用时,不仅要进行大气校正,而且还要慎重选择大气校正模型和植被指数。  相似文献

基于海南岛1∶200 000土壤 地体数字化数据库(HNSOTER),在GIS系统的支持下,对海南岛土壤有机碳储量及分布特征进行了探讨.结果表明,1)标准剖深下(0~100 cm),海南岛土壤有机碳储量为2.78×108 t.2)0~20 cm剖深土壤有机碳密度变幅在0.3~18.8 kg·m-2之间,其中1.0~5.0 kg·m-2密度区占总分布面积的81.2%,按面积加权均值为3.3 kg·m-2.0~100 cm剖深的土壤有机碳密度变幅在1.0~32.1 kg·m-2 之间,其中2.0~14.0 kg·m-2密度区面积比重占89.7%,按面积加权均值为8.4 kg·m-2.不同地形、岩性及土壤类型中,土壤有机碳密度分布有较大变异.3)从中部山地向外至沿海平原,土壤有机碳密度总体上呈递减趋势,但不同剖深下其分布格局仍有一定差异.0~20 cm剖深下土壤有机碳密度高值区(丰富度指数大于1)主要分布于山地以及北部玄武岩台地,0~100 cm剖深下,有机碳密度高值区(丰富度指数大于1)趋向集中于中东部山地、台地区,且其分布重心较前者明显的向东偏移.  相似文献
