Rhodnius prolixus, a vector of Chagas disease, is a hematophagous insect that feeds exclusively on blood. Each blood meal is digested within the first fourteen days after feeding, providing substrates for lipid synthesis for storage and egg production. These events are precisely regulated and emerging evidence points to a key function of insulin-like peptides (ILPs) in this control. Here we investigated the role of insulin receptor in the regulation of nutrient metabolism in fed adult females. The expression of insulin receptor (RhoprIR) gene was determined in adult organs, and it was highest in ovaries and previtellogenic follicles. We generated insects with RNAi-mediated knockdown of RhoprIR to address the physiological role of this receptor. RhoprIR deficiency improved longevity and reduced triacylglycerol storage in the fat body, whereas blood digestion remained unchanged for seven days after blood meal. The lower lipid content was attributable to decreased de novo lipogenesis as well as reduced incorporation of hemolymph-derived fatty acids into newly synthesized lipids within this organ. Consistent with that, fat bodies from RhoprIR-deficient insects exhibited decreased gene expression levels of lipophorin receptor (RhoprLpR), glycerol-3-phosphate acyltransferase 1 and 4 (RhoprGpat1 and RhoprGpat4), and carnitine palmitoyltransferase 1 (RhoprCpt1). Although hemolymph lipid profile was not affected by RhoprIR disruption, the concentration of circulating vitellogenin was increased. In line with these changes, RhoprIR-deficient females exhibited smaller ovaries and a marked reduction in oviposition. Taken together, these findings support a key role of insulin receptor in nutrient homeostasis, lipid synthesis and egg production following a blood meal.  相似文献

Ecto-3′-nucleotidase/nuclease (3′NT/NU) is a membrane-bound enzyme that plays a key role in the nutrition of Leishmania sp. protozoan parasites. This enzyme generates nucleosides via hydrolyzes of 3′mononucleotides and nucleic acids, which enter the cell by specific transporters. In this work, we identify and characterize Leishmania amazonensis ecto-3′-nucleotidase activity (La3′-nucleotidase), report ammonium tetrathiomolybdate (TTM) as a novel La3′-nucleotidase inhibitor and approach the possible involvement of ecto-3′-nucleotidase in cellular adhesion. La3′-nucleotidase presented characteristics similar to those reported for the class I single-strand nuclease family; a molecular weight of approximately 40 kDa and optimum activity in an alkaline pH range were observed. Although it is conserved among the genus, La3′-nucleotidase displays different kinetic properties; it can be inhibited by vanadate, molybdate and Cu2+ ions. Interestingly, ecto-3′-nucleotidase activity is 60-fold higher than that of ecto-5′-nucleotidase in L. amazonensis. Additionally, ecto-3′-nucleotidase activity is two-fold higher in virulent L. amazonensis cells than in avirulent ones. Notably, macrophage–parasite attachment/invasion was increased by 400% in the presence of adenosine 3′-monophosphate (3′AMP); however, this effect was reverted by TTM treatment. We believe that La3′-nucleotidase may play a significant role in the generation of adenosine, which may contribute to mammalian host immune response impairment and establishment of infection.  相似文献

The red palm mite Raoiella indica Hirst (Tenuipalpidae) was first reported in the New World in 2004, dispersing quickly and widely while adopting new plant species as hosts. Since then, it has caused severe damage in this region, especially to coconut (Cocos nucifera L.). It was first found in Brazil in 2009, in the northern Amazonian state of Roraima. In the present study, native and introduced plants were sampled between March 2010 and February 2011 in sites of the 15 Roraima municipalities, to estimate its distribution and the associated mite fauna. In addition, monthly samples were taken from a coconut plantation in Mucajaí throughout the same period, for an initial appraisal of the levels R. indica could reach. It was found in 10 municipalities, on 19 plant species of four families. Six species are reported for the first time as hosts. Among the associated predators, 89.1% were Phytoseiidae, most commonly Amblyseius largoensis (Muma), Iphiseiodes zuluagai Denmark & Muma and Euseius concordis (Chant). The highest densities of R. indica, 1.5 and 0.35 mites/cm2 of leaflet (approx total of 331 and 77 mites/leaflet), were reached respectively in March 2010 and February 2011. The highest density of phytoseiids on coconut (0.009 mites/cm2 or about 2 mites/leaflet) was reached in November 2010. The average densities of R. indica recorded for Roraima were comparable to those reported for countries in which the mite is reportedly economically damaging. The dispersal of R. indica through the Amazon forest may result in damage to cultivated and native palms, and plants of other families, if the projected increase in both the frequency and the severity of drought events occurs. Parts of the Amazon have undergone periods of low rainfall, a condition that appears to favour the biology of this mite. Its eventual arrival to northeastern Brazil may result in heavy economic and ecological losses.  相似文献

The non-target effects of acaricides, used against Aceria guerreronis (Acari: Eriophyidae) the coconut mite, on its natural enemies are not known. Therefore we assessed the susceptibility of A. guerreronis and its predator Neoseiulus baraki (Acari: Phytoseiidae) to selected acaricides, their impact on N. baraki rate of increase, and the synergism of fenpyroximate towards this predator. Toxicity bioassays and synergism of fenpyroximate (with piperonyl butoxide, triphenyl phosphate and diethyl maleate) were performed by spraying the mites under a Potter tower. The instantaneous rate of increase (r i ) was calculated ten days after spraying the predator. Chlorfenapyr and fenpyroximate were selective (both LC50 and LC90 were higher for N. baraki than for A. guerreronis) and did not affect the predator r i . Only piperonyl butoxide significantly synergized fenpyroximate suggesting the involvement of cytochrome P450 monooxygenase in the N. baraki tolerance. Fenpyroximate and chlorfenapyr are promising agents for managing A. guerreronis in combination with N. baraki because both are selective and do not affect its predator r i .  相似文献

Insect eggs must contain the necessary nutrients for embryonic growth. In this article, we investigated the accumulation of triacylglycerol (TAG) in growing oocytes and its utilization during embryonic development. TAG makes up about 60% of the neutral lipids in oocytes and accumulates as oocytes grow, from 2.2 ± 0.1 µg in follicles containing 1.0 mm length oocytes to 10.2 ± 0.8 µg in 2.0 mm length oocytes. Lipophorin (Lp), the hemolymphatic lipoprotein, radioactively labeled in free fatty acid (FFA) or diacylglycerol (DAG), was used to follow the transport of these lipids to the ovary. Radioactivity from both lipid classes accumulated in the oocytes, which was abolished at 4°C. The capacity of the ovary to receive FFA or DAG from Lp varied according to time after a blood meal and reached a maximum around the second day. 3H‐DAG supplied by Lp to the ovaries was used in the synthesis of TAG as, 48 hr after injection, most of the radioactivity was found in TAG (85.7% of labeling in neutral lipids). During embryogenesis, lipid stores were mobilized, and the TAG content decreased from 16.4 ± 2.1 µg/egg on the first day to 10.0 ± 1.3 µg on day 15, just before hatching. Of these, 7.4 ± 0.9 µg were found in the newly emerged nymphs. In unfertilized eggs, the TAG content did not change. Although the TAG content decreased during embryogenesis, the relative lipid composition of the egg did not change. This study was undertaken to elucidate the effect of the essential oil from Alpinia speciosa (EOAs) on cardiac contractility and the underlying mechanisms. The essential oil was obtained from Alpinia speciosa leaves and flowers and the oil was analyzed by GC-MS method. Chemical analysis revealed the presence of at least 18 components. Terpinen-4-ol and 1,8-cineole corresponded to 38% and 18% of the crude oil, respectively. The experiments were conducted on spontaneously-beating right atria and on electrically stimulated left atria isolated from adult rats. The effect of EOAs on the isometric contractions and cardiac frequency in vitro was examined. EOAs decreased rat left atrial force of contraction with an EC50 of 292.2 ± 75.7 μg/ml. Nifedipine, a well known L-type Ca2+ blocker, inhibited in a concentration-dependent manner left atrial force of contraction with an EC50 of 12.1 ± 3.5 μg/ml. Sinus rhythm was diminished by EOAs with an EC50 of 595.4 ± 56.2 μg/ml. Whole-cell L-type Ca2+ currents were recorded by using the patch-clamp technique. EOAs at 25 μg/ml decreased ICa,L by 32.6 ± 9.2% and at 250 μg/ml it decreased by 89.3 ± 7.4%. Thus, inhibition of L-type Ca2+ channels is involved in the cardiodepressive effect elicited by the essential oil of Alpinia speciosa in rat heart.  相似文献

The presence of the red palm mite, Raoiella indica Hirst, is reported for the first time in Brazil. This invasive mite was found in July 2009 infesting coconut palms and bananas in urban areas of Boa Vista, State of Roraima, in northern Brazil. Comments on the possible pathways of R. indica into the country, present and potential impact of its introduction and mitigating measures to prevent or to delay the mite spread in Brazil are presented.  相似文献

We provide the first annotated checklist of ophiuroids from the continental shelf of the State of Paraíba, northeastern Brazil. Identification keys and taxonomic diagnoses for 23 species, belonging to 14 genera and 8 families, are provided. The material is deposited in the Invertebrate Collection Paulo Young, at the Federal University of Paraíba. Ophiopsila hartmeyeri represents the first record for the northeastern region of Brazil, while Ophiolepis impressa, Ophiolepis paucispina, Amphiura stimpsoni, Amphiodia riisei, Ophiactis quinqueradia, Ophiocoma wendtii and Ophionereis olivaceae are new records for the State of Paraíba. The number of species known for the state was increased from 16 to 23, representing approximately 17% of the species known for Brazil and 54% of the species known for northeastern Brazil. The recorded fauna has a large geographical and bathymetrical distribution.  相似文献

The purpose of this study was to investigate whether sporulated Neospora caninum oocysts, which had been stored for 46 mo in a 2% sulfuric acid solution at 4 degrees C, remain morphologically viable and infective to gerbils (Meriones unguiculatus). Six gerbils were orally inoculated with doses of 400 or 1,200 oocysts. Two mo after inoculation, the animals did not show any clinical signs, had no histological lesions, and were seronegative for N. caninum at 1: 50 in an immunofluorescent antibody test. PCR using the brain from each gerbil did not reveal N. caninum specific DNA. We conclude that oocysts preserved for 46 mo are not infective, despite being morphologically intact.  相似文献

The aim of this study was to investigate the impact of moderate aerobic training on functional, anthropometric, biochemical, and health-related quality of life (HRQOL) parameters on women with metabolic syndrome (MS). Fifteen untrained women with MS performed moderate aerobic training for 15 weeks, without modifications of dietary behaviours. Functional, anthropometric, biochemical, control diet record and HRQOL parameters were assessed before and after the training. Despite body weight maintenance, the patients presented decreases in waist circumference (P = 0.001), number of MS components (P = 0.014), total cholesterol (P = 0.049), HDL cholesterol (P = 0.004), LDL cholesterol (P = 0.027), myeloperoxidase activity (P = 0.002) and thiobarbituric acid-reactive substances levels (P = 0.006). There were no differences in total energy, carbohydrate, protein and lipid intake pre- and post-training. Furthermore, improvements in the HRQOL subscales of physical functioning (P = 0.03), role-physical (P = 0.039), bodily pain (P = 0.048), general health (P = 0.046) and social functioning scoring (P = 0.011) were reported. Despite the absence of weight loss, aerobic training induced beneficial effects on functional, anthropometric, biochemical and HRQOL parameters in women with MS.  相似文献
