Carbamoylphosphate synthetase (CPS) catalyzes the first committed step in
pyrimidine biosynthesis, arginine biosynthesis, or the urea cycle.
Organisms may contain either one generalized or two specific CPS enzymes,
and these enzymes may be heterodimeric (encoded by linked or unlinked
genes), monomeric, or part of a multifunctional protein. In order to help
elucidate the evolution of CPS, we have performed a comprehensive
phylogenetic analysis using the 21 available complete CPS sequences,
including a sequence from Sulfolobus solfataricus P2 which we report in
this paper. This is the first report of a complete CPS gene sequence from
an archaeon, and sequence analysis suggests that it encodes an enzyme
similar to heterodimeric CPSII. We confirm that internal similarity within
the synthetase domain of CPS is the result of an ancient gene duplication
that preceded the divergence of the Bacteria, Archaea, and Eukarya, and use
this internal duplication in phylogenetic tree construction to root the
tree of life. Our analysis indicates with high confidence that this
archaeal sequence is more closely related to those of Eukarya than to those
of Bacteria. In addition to this ancient duplication which created the
synthetase domain, our phylogenetic analysis reveals a complex history of
further gene duplications, fusions, and other events which have played an
integral part in the evolution of CPS.

Background
Adipose tissue patterning has a major influence on the risk of developing chronic disease. Environmental influences on both body fat patterning and appetite regulation are not fully understood. This study was performed to investigate the impact of resistant starch (RS) on adipose tissue deposition and central regulation of appetite in mice.Methodology and Principle Findings
Forty mice were randomised to a diet supplemented with either the high resistant starch (HRS), or the readily digestible starch (LRS). Using 1H magnetic resonance (MR) methods, whole body adiposity, intrahepatocellular lipids (IHCL) and intramyocellular lipids (IMCL) were measured. Manganese-enhanced MRI (MEMRI) was used to investigate neuronal activity in hypothalamic regions involved in appetite control when fed ad libitum. At the end of the interventional period, adipocytes were isolated from epididymal adipose tissue and fasting plasma collected for hormonal and adipokine measurement. Mice on the HRS and LRS diet had similar body weights although total body adiposity, subcutaneous and visceral fat, IHCL, plasma leptin, plasma adiponectin plasma insulin/glucose ratios was significantly greater in the latter group. Adipocytes isolated from the LRS group were significantly larger and had lower insulin-stimulated glucose uptake. MEMRI data obtained from the ventromedial and paraventricular hypothalamic nuclei suggests a satiating effect of the HRS diet despite a lower energy intake.Conclusion and Significance
Dietary RS significantly impacts on adipose tissue patterning, adipocyte morphology and metabolism, glucose and insulin metabolism, as well as affecting appetite regulation, supported by changes in neuronal activity in hypothalamic appetite regulation centres which are suggestive of satiation. 相似文献124.

Adiponectin, a collagenous hormone secreted abundantly from adipocytes, possesses potent antidiabetic and anti-inflammatory properties. Mediated by the conserved Cys39 located in the variable region of the N terminus, the trimeric (low molecular weight (LMW)) adiponectin subunit assembles into different higher order complexes, e.g. hexamers (middle molecular weight (MMW)) and 12–18-mers (high molecular weight (HMW)), the latter being mostly responsible for the insulin-sensitizing activity of adiponectin. The endoplasmic reticulum (ER) chaperone ERp44 retains adiponectin in the early secretory compartment and tightly controls the oxidative state of Cys39 and the oligomerization of adiponectin. Using cellular and in vitro assays, we show that ERp44 specifically recognizes the LMW and MMW forms but not the HMW form. Our binding assays with short peptide mimetics of adiponectin suggest that ERp44 intercepts and converts the pool of fully oxidized LMW and MMW adiponectin, but not the HMW form, into reduced trimeric precursors. These ERp44-bound precursors in the cis-Golgi may be transported back to the ER and released to enhance the population of adiponectin intermediates with appropriate oxidative state for HMW assembly, thereby underpinning the process of ERp44 quality control. 相似文献

A case of acute lymphoblastic leukaemia in a 17-year-old male is described. The patient had many distinctive features including a very high blast cell count, prominent lymphadenopathy and hepatosplenomegaly, thymic mass, and a fulminant clinical course. Immunological studies on the blast cells using a variety of techniques showed the presence of two distinct subpopulations, one having the surface characteristics of thymus-dependent (T) lymphoid cells and the other those of bursa-equivalent (B) lymphoid cells.
