The aim of this study was to assess the suitability of body mass index, waist circumference, waist-to-height ratio and aerobic fitness as predictors of cardiovascular risk factor clustering in children. A cross-sectional study was conducted with 290 school boys and girls from 6 to 10 years old, randomly selected. Blood was collected after a 12-hour fasting period. Blood pressure, waist circumference (WC), height and weight were evaluated according to international standards. Aerobic fitness (AF) was assessed by the 20-metre shuttle-run test. Clustering was considered when three of these factors were present: high systolic or diastolic blood pressure, high low-density lipoprotein (LDL) cholesterol, high triglycerides, high plasma glucose, high insulin concentrations and low high-density lipoprotein (HDL) cholesterol. A ROC curve identified the cut-off points of body mass index (BMI), WC, waist-to-height ratio (WHtR) and AF as predictors of risk factor clustering. BMI, WC and WHR resulted in significant areas under the ROC curves, which was not observed for AF. The anthropometric variables were good predictors of cardiovascular risk factor clustering in both sexes, whereas aerobic fitness should not be used to identify cardiovascular risk factor clustering in these children.  相似文献

Dyskeratosis Congenita (DC) is an inherited multisystem premature aging disorder with characteristic skin and mucosal findings as well as a predisposition to cancer and bone marrow failure. DC arises due to gene mutations associated with the telomerase complex or telomere maintenance, resulting in critically shortened telomeres. The pathogenesis of DC, as well as several congenital bone marrow failure (BMF) syndromes, converges on the DNA damage response (DDR) pathway and subsequent elevation of reactive oxygen species (ROS). Historically, DC patients have had poor outcomes following bone marrow transplantation (BMT), perhaps as a consequence of an underlying DNA hypersensitivity to cytotoxic agents. Previously, we demonstrated an activated DDR and increased ROS, augmented by chemotherapy and radiation, in somatic cells isolated from DC patients with a mutation in the RNA component of telomerase, TERC. The current study was undertaken to determine whether previous findings related to ROS and DDR in TERC patients’ cells could be extended to other DC mutations. Of particular interest was whether an antioxidant approach could counter increased ROS and decrease DC pathologies. To test this, we examined lymphocytes from DC patients from different DC mutations (TERT, TINF2, and TERC) for the presence of an active DDR and increased ROS. All DC mutations led to increased steady-state p53 (2-fold to 10-fold) and ROS (1.5-fold to 2-fold). Upon exposure to ionizing radiation (XRT), DC cells increased in both DDR and ROS to a significant degree. Exposing DC cells to hydrogen peroxide also revealed that DC cells maintain a significant oxidant burden compared to controls (1.5-fold to 3-fold). DC cell culture supplemented with N-acetylcysteine, or alternatively grown in low oxygen, afforded significant proliferative benefits (proliferation: maximum 2-fold increase; NAC: 5-fold p53 decrease; low oxygen: maximum 3.5-fold p53 decrease). Together, our data supports a mechanism whereby telomerase deficiency and subsequent shortened telomeres initiate a DDR and create a pro-oxidant environment, especially in cells carrying the TINF2 mutations. Finally, the ameliorative effects of antioxidants in vitro suggest this could translate to therapeutic benefits in DC patients.  相似文献

To gain insight into the defects responsible for impaired Th1 responses in human newborns, we analyzed the production of cytokines by dendritic cells (DC) derived from cord blood monocytes. We observed that neonatal DC generated from adherent cord blood mononuclear cells cultured for 6 days in the presence of IL-4 and GM-CSF show a phenotype similar to adult DC generated from adherent PBMC, although they express lower levels of HLA-DR, CD80, and CD40. Measurement of cytokine levels produced by neonatal DC upon stimulation by LPS, CD40 ligation, or poly(I:C) indicated a selective defect in the synthesis of IL-12. Determination of IL-12(p40) and IL-12(p35) mRNA levels by real-time RT-PCR revealed that IL-12(p35) gene expression is highly repressed in stimulated neonatal DC whereas their IL-12(p40) gene expression is not altered. The addition of rIFN-gamma to LPS-stimulated newborn DC restored their expression of IL-12(p35) and their synthesis of IL-12 (p70) up to adult levels. Moreover, we observed that neonatal DC are less efficient than adult DC to induce IFN-gamma production by allogenic adult CD4(+) T cells. This defect was corrected by the addition of rIL-12. We conclude that neonatal DC are characterized by a severe defect in IL-12(p35) gene expression which is responsible for an impaired ability to elicit IFN-gamma production by T cells.  相似文献

A new instrument, based on a technique described previously, is presented for studying mechanics of micron-scale preparations of two to three myofibrils or single myofibrils from muscle. Forces in the nanonewton to micronewton range are measurable with 0.5-ms time resolution. Programmed quick (200-microseconds) steps or ramp length changes are applied to contracting myofibrils to test their mechanical properties. Individual striations can be monitored during force production and shortening. The active isometric force, force-velocity relationship, and force transients after rapid length steps were obtained from bundles of two to three myofibrils from rabbit psoas muscle. Contrary to some earlier reports on myofibrillar mechanics, these properties are generally similar to expectations from studies on intact and skinned muscle fibers. Our experiments provide strong evidence that the mechanical properties of a fiber result from a simple summation of the myofibrillar force and shortening of independently contracting sarcomeres.  相似文献

The regulatory effect of calcium added in vitro on 25-hydroxycholecalciferol metabolism was studied in kidney mitochondria and in renal tubules from vitamin D-deficient chicks. The addition of calcium (0.05 – 0.2 mm) to mitochondrial suspensions prepared with calcium-chelating agents caused a marked and dose-related stimulation of 1-hydroxylation. A sharp decline in the activity was induced by higher concentrations of calcium (0.3 – 0.7 mm). A similar but less striking biphasic effect of calcium on 1-hydroxylation was observed in mitochondria prepared in the absence of calcium chelating agents. The effect of calcium was not a consequence of accelerated mitochondrial translocation of either exogenous NADP or Mg2+ but was related to mitochondrial calcium content. The addition of inhibitors of the calcium uptake, i.e., LaCl3 or ruthenium red, or a calcium ionophore (A 23187) significantly inhibited the calcium-induced stimulation of the 1-hydroxylation reaction. Similar calcium effects were also observed in renal tubules isolated from intact, but not from parathyroidectomized, vitamin D-deficient chicks. These data strongly suggest that mitochondrial calcium plays an important role in the regulation of 1-hydroxylase activity in kidney.  相似文献

The Turkish hamster is a long-day breeder that hibernates for 4-5 mo if exposed to a short-day, cold environment. The objective of this study was to assess the uterine responsiveness of the hibernating animal to ovarian steroids. Our approach was 1) to characterize and determine uterine estrogen (E) and progesterone (P) receptors (R) during hibernation as compared to the levels observed in cycling females that had terminated hibernation, and 2) to assess the responsiveness of the uterus to E during hibernation by its ability to induce uterine P receptor. Females were exposed to short days (10L:14D) for 2 mo and then were placed in a cold-room (10L: 14D, 6 +/- 1 degrees C). After 2 or 4 mo in the cold, hibernating animals were killed and uterine steroid receptors were determined by 3H-steroid binding assay. Uterine receptors were also determined in cycling Turkish hamsters on each morning of the estrous cycle. Values for uterine receptors (pmol/g tissue, n = 4-6) during the estrous cycle (estrus, diestrus I, diestrus II, proestrus) were: 4.3 +/- 0.78, 3.9 +/- 0.19, 4.1 +/- 0.25, 3.7 +/- 0.5 for cytosolic ER; 36.6 +/- 5.8, 32.2 +/- 6.8, 36.3 +/- 1.5, 54.4 +/- 1.9 for cytosolic PR; 0.59 +/- 0.11, 0.54 +/- 0.07, 1.06 +/- 0.05, 1.42 +/- 0.17 for nuclear ER. Hibernating (torpid) animals sampled after 2 mo in the cold showed a significant (p less than 0.05) depression of cytosolic ER (2.6 +/- 0.12, n = 5) and cytosolic PR (19.0 +/- 2.6, n = 8) as compared to any day of the estrous cycle.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Early leak current, i.e. for times similar to the time to peak of the transient current was measured in Myxicola giant axons in the presence of tetrodotoxin. The leak current-voltage relation rectifies, showing more current for strong depolarizing pulses than expected from symmetry around the holding potential. A satisfactory practical approximation for most leak corrections is constant resting conductance. The leak current-voltage curve rectifies less than expected from the constant field equation. These curves cannot be reconstructed by summing the constant field currents for sodium and potassium using a PNa/PK ratio obtained in the usual way, from zero current constant field fits to resting membrane potential data. Nor can they be reconstructed by summing the constant field current for potassium with that for any other single ion. They can be reconstructed, however, by summing the constant field current for potassium with a constant conductance component. It is concluded that the leak current and the resting membrane potential, therefore, are determined by multiple ionic components, at least three and possibly many. Arguments are presented suggesting that ion permeability ratios obtained in the usual way, by fitting the constant field equation to resting membrane potential data should be viewed with skepticism.  相似文献

Phylogenomic approaches to the resolution of inter-species relationships have become well established in recent years. Often these involve concatenation of many orthologous genes found in the respective genomes followed by analysis using standard phylogenetic models. Genome-scale data promise increased resolution by minimising sampling error, yet are associated with well-known but often inappropriately addressed caveats arising through data heterogeneity and model violation. These can lead to the reconstruction of highly-supported but incorrect topologies. With the aim of obtaining a species tree for 18 species within the ascomycetous yeasts, we have investigated the use of appropriate evolutionary models to address inter-gene heterogeneities and the scalability and validity of supermatrix analysis as the phylogenetic problem becomes more difficult and the number of genes analysed approaches truly phylogenomic dimensions. We have extended a widely-known early phylogenomic study of yeasts by adding additional species to increase diversity and augmenting the number of genes under analysis. We have investigated sophisticated maximum likelihood analyses, considering not only a concatenated version of the data but also partitioned models where each gene constitutes a partition and parameters are free to vary between the different partitions (thereby accounting for variation in the evolutionary processes at different loci). We find considerable increases in likelihood using these complex models, arguing for the need for appropriate models when analyzing phylogenomic data. Using these methods, we were able to reconstruct a well-supported tree for 18 ascomycetous yeasts spanning about 250 million years of evolution.  相似文献
