Endothelial dysfunction is one of the earliest events in atherogenesis. A consequence of endothelial damage is a lower availability of nitric oxide (NO), the most potent endogenous vasodilator. NO inhibits platelet aggregation, smooth muscle cell proliferation and adhesion of monocytes to endothelial cells. Endothelial dysfunction is present in patients with cardiovascular disease and/or coronary risk factors, such as hypertension, dyslipidemia, diabetes, smoking or hyperhomocysteinemia. At present, soluble markers and high resolution ultrasound of the brachial artery, have provided simple tools for the study of endothelial function and the effects of several interventions. It has been demonstrated that dietary factors may induce significant changes on vascular reactivity. Nutrients, such as fish oil, antioxidants, L-arginine, folic acid and soy protein have shown an improvement in endothelial function that can mediate, at least partially, the cardioprotective effects of these substances. Attention has been focused on dietary patterns in populations with lower prevalence of cardiovascular disease. There is some evidence suggesting that Mediterranean diet characterized by high consumption of vegetables, fish, olive oil and moderate wine consumption may have a positive effect on endothelial function. These results give us evidence on the significant role of diet on endothelial function and its impact on the pathogenesis of atherosclerosis. 相似文献

Abstract: In the present study, the question of whether immunoreactive α-melanotropin (α-MSH1), corticotropin (ACTH1), and β-melanotropin (β-MSH1) are co-sequestered in hypothalamic granules of adult male rats was addressed. When a 900 ×g supernatant fluid prepared from a hypothalamic homogenate was fractionated on continuous sucrose density gradients under non-equilibrium Conditions, two populations of particles containing α-MSH1, ACTH1, or β-MSH1 Were observed. However, when fractionated under equilibrium conditions, the two populations of particles containing α-MSH1 ACTH1, or β-MSH1 were recovered as a single band. This sedimentation characteristic indicates that the particles containing a given peptide differ in size but are similar in density. In their sedimentation, the small particles containing α-MSH1, ACTH1, and β-MSH1 are indistinguishable from granules containing α-MSH1, whereas the large particles containing α-MSH1 (ACTH1, and β-MSH1 are indistinguishable from synaptosomes containing α-MSH1, β-MSH1 had an apparent molecular weight (M.W.) of about 5,000, which is similar to that of γ-lipotropin. ACTH1 was comprised of three species of molecules: big (M.W. ≥ 10,000), 5.7K (M.W. ≌ 5,700), and 4.5K (M.W. ≌ 4,500). Big ACTH was the predominant and 5.7K ACTH the minor component of ACTH1 present in granules as well as in synaptosomes. These results are suggestive that α-MSH, ACTH and its precursors, and γ-lipotropin are co-sequestered in hypothalamic granules. 相似文献

Although most primates live in groups, experiments on reciprocity usually test individuals in dyads. This could hide the processes emerging in richer social settings, reducing the ecological validity of the results. We run an experiment on reciprocal food transfers testing capuchin monkeys (Cebus apella) in triads, so that subjects could choose to allow access to their food to either of their two partners. We tested the hypothesis that partner choice was related to a comparison of long-term social bonds with the two partners, more than to a comparison of recent food transfer events from the two partners. The results confirmed this hypothesis, thus supporting the notion that reciprocal partner preferences are based on long-term accounts of benefits that have been exchanged. 相似文献

Voltage-gated and ligand-gated ion channels are used in eukaryotic organisms for the purpose of electrochemical signaling. There are prokaryotic homologues to major eukaryotic channels of these sorts, including voltage-gated sodium, potassium, and calcium channels, Ach-receptor and glutamate-receptor channels. The prokaryotic homologues have been less well characterized functionally than their eukaryotic counterparts. In this study we identify likely prokaryotic functional counterparts of eukaryotic glutamate receptor channels by comprehensive analysis of the prokaryotic sequences in the context of known functional domains present in the eukaryotic members of this family. In particular, we searched the nonredundant protein database for all proteins containing the following motif: the two sections of the extracellular glutamate binding domain flanking two transmembrane helices. We discovered 100 prokaryotic sequences containing this motif, with a wide variety of functional annotations. Two groups within this family have the same topology as eukaryotic glutamate receptor channels. Group 1 has a potassium-like selectivity filter. Group 2 is most closely related to eukaryotic glutamate receptor channels. We present analysis of the functional domain architecture for the group of 100, a putative phylogenetic tree, comparison of the protein phylogeny with the corresponding species phylogeny, consideration of the distribution of these proteins among classes of prokaryotes, and orthologous relationships between prokaryotic and human glutamate receptor channels. We introduce a construct called the Evolutionary Domain Network, which represents a putative pathway of domain rearrangements underlying the domain composition of present channels. We believe that scientists interested in ion channels in general, and ligand-gated ion channels in particular, will be interested in this work. The work should also be of interest to bioinformatics researchers who are interested in the use of functional domain-based analysis in evolutionary and functional discovery. 相似文献

Green nanotechnology has acquired immense demand due to its cost-effective, eco-friendly and benevolent approach for the synthesis of nanoparticles. Among the biological methods, plants aid as a significant green resource for synthesizing nanoparticles that are safe and non-toxic for human use. In the present investigation, Silver nanoparticles (AgNPs) were synthesized using bulbs extract of Allium ampeloprasum under the influence of sunlight irradiation and characterized using different techniques. Distinct in-vitro assays were performed to test the antioxidant and anticandida potential of the synthesized AgNPs. Results suggested the efficient and rapid sunlight-driven synthesis of AgNPs using A. ampeloprasum extract. UV–Vis spectrum showed absorption peak at 446 nm which confirmed the formation of AgNPs. FTIR analysis suggested the presence of functional groups associated with flavonoids and sulfur compounds in A. ampeloprasum extract. The synthesized AgNPs showed Face Centred Cubic (FCC) structure with an average size of 35 nm. Spherical, quasi spherical, triangular and ellipsoidal morphology of the NPs were observed from the TEM micrograph. The synthesized AgNPs showed pronounced free radical scavenging potential for DPPH, ABTS∙+ and H2O2 radicals. The anticandida potency of the synthesized AgNPs was observed as follows: C. albicans ≥ C. tropicalis ≥ C. glabrata ≥ C. parapsilosis ≥ C. krusei. Results showed that sunlight driven nanoparticle synthesis of AgNPs is rapid, facile and exhibit enhanced antioxidant and antifungal activity. 相似文献

A large number of protected areas worldwide have been impacted by biological invasions, threatening the biodiversity they aim to protect. The wild boar (Sus scrofa) is one of the most threatening invasive species in Argentina, already occupying many ecoregions, including the central Monte Desert. However, there are no studies regarding the use that wild boars make of this invaded biome and what factors (climate or landscape) determine or contribute to the establishment of this species. The objectives of this study were to assess habitat use of the wild boar at spatial and temporal scales in the central Monte Desert, and to assess if climatic factors influence its abundance. Our results show that, at habitat-level the wild boar exhibited preferences for a particular habitat (Larrea shrubland) for feeding. At microhabitat-level, we found a positive association between herb cover and wild boar presence. In addition, we found a strong and positive association between the number of days with low temperatures and the number of wild boar signs registered. Therefore, we consider that in the central Monte Desert, habitat selection by wild boars is most likely determined by a maximization of food intake and a minimization of exposure to high temperature. 相似文献

In yeast cells, subunit a of the vacuolar proton pump (V-ATPase) is encoded by two organelle-specific isoforms, VPH1 and STV1. V-ATPases containing Vph1 and Stv1 localize predominantly to the vacuole and the Golgi apparatus/endosomes, respectively. Ratiometric measurements of vacuolar pH confirm that loss of STV1 has little effect on vacuolar pH. Loss of VPH1 results in vacuolar alkalinization that is even more rapid and pronounced than in vma mutants, which lack all V-ATPase activity. Cytosolic pH responses to glucose addition in the vph1Δ mutant are similar to those in vma mutants. The extended cytosolic acidification in these mutants arises from reduced activity of the plasma membrane proton pump, Pma1p. Pma1p is mislocalized in vma mutants but remains at the plasma membrane in both vph1Δ and stv1Δ mutants, suggesting multiple mechanisms for limiting Pma1 activity when organelle acidification is compromised. pH measurements in early prevacuolar compartments via a pHluorin fusion to the Golgi protein Gef1 demonstrate that pH responses of these compartments parallel cytosolic pH changes. Surprisingly, these compartments remain acidic even in the absence of V-ATPase function, possibly as a result of cytosolic acidification. These results emphasize that loss of a single subunit isoform may have effects far beyond the organelle where it resides. 相似文献

In plants, ammonium released during photorespiration exceeds primary nitrogen assimilation by as much as 10-fold. Analysis of photorespiratory mutants indicates that photorespiratory ammonium released in mitochondria is reassimilated in the chloroplast by a chloroplastic isoenzyme of glutamine synthetase (GS2), the predominant GS isoform in leaves of Solanaceous species including tobacco (Nicotiana tabacum). By contrast, cytosolic GS1 is expressed in the vasculature of several species including tobacco. Here, we report the effects on growth and photorespiration of overexpressing a cytosolic GS1 isoenzyme in leaf mesophyll cells of tobacco. The plants, which ectopically overexpress cytosolic GS1 in leaves, display a light-dependent improved growth phenotype under nitrogen-limiting and nitrogen-non-limiting conditions. Improved growth was evidenced by increases in fresh weight, dry weight, and leaf soluble protein. Because the improved growth phenotype was dependent on light, this suggested that the ectopic expression of cytosolic GS1 in leaves may act via photosynthetic/photorespiratory process. The ectopic overexpression of cytosolic GS1 in tobacco leaves resulted in a 6- to 7-fold decrease in levels of free ammonium in leaves. Thus, the overexpression of cytosolic GS1 in leaf mesophyll cells seems to provide an alternate route to chloroplastic GS2 for the assimilation of photorespiratory ammonium. The cytosolic GS1 transgenic plants also exhibit an increase in the CO(2) photorespiratory burst and an increase in levels of photorespiratory intermediates, suggesting changes in photorespiration. Because the GS1 transgenic plants have an unaltered CO(2) compensation point, this may reflect an accompanying increase in photosynthetic capacity. Together, these results provide new insights into the possible mechanisms responsible for the improved growth phenotype of cytosolic GS1 overexpressing plants. Our studies provide further support for the notion that the ectopic overexpression of genes for cytosolic GS1 can potentially be used to affect increases in nitrogen use efficiency in transgenic crop plants. 相似文献

Adsorption/desorption processes (sorption isotherms) of Naproxen in a sandy aquifer matrix sediment were investigated using batch tests to compare Naproxen sorption behavior at 15°C and 25°C. Both temperatures are representative of the aquifer media and environmental conditions. Adsorption was well described by linear isotherms with low sorption affinity to aquifer material (Kd of 0.4 μg kg?1) at both temperatures (15°C and 25°C). Desorption isotherm coefficients at 15°C and 25°C were 5.0 and 4.9, respectively. Naproxen hysteresis indices were between 9.98 and 10.8, indicating that a Naproxen fraction may be irreversibly fixed in the aquifer media, being higher at 25°C (10.88) compared to 15°C, showing a decreasing trend with increasing compound concentration at 15°C. The low sorption of Naproxen leads to potential leaching to groundwater if present in irrigation water, and its prevalence in an aquifer media when directly injected in wells for groundwater recharge. 相似文献

Preclinical studies in mice and non-human primates showed that AAV serotype 5 provides efficient liver transduction and as such seems a promising vector for liver directed gene therapy. An advantage of AAV5 compared to serotype 8 already shown to provide efficient correction in a phase 1 trial in patients suffering from hemophilia B, is its lower seroprevalence in the general population. Our goal is liver directed gene therapy for Crigler-Najjar syndrome type I, inherited severe unconjugated hyperbilirubinemia caused by UGT1A1 deficiency. In a relevant animal model, the Gunn rat, we compared the efficacy of AAV 5 and 8 to that of AAV1 previously shown to be effective. Ferrying a construct driving hepatocyte specific expression of UGT1A1, both AAV8 and AAV1 provided an efficient correction of hyperbilirubinemia. In contrast to these two and to other animal models AAV5 failed to provide any correction. To clarify whether this unexpected finding was due to the rat model used or due to a problem with AAV5, the efficacy of this serotype was compared in a mouse and two additional rat strains. Administration of an AAV5 vector expressing luciferase under the control of a liver specific promoter confirmed that this serotype poorly performed in rat liver, rendering it not suitable for proof of concept studies in this species. 相似文献
