Damage to crops by drought is still a serious problem in large areas of the world. Considerable research has been undertaken to discover the mechanisms of drought injury and drought resistance of plants. However, the critical features of drought injury have not yet been identified. In the past ten years a free radical hypothesis has been suggested to account for subcellular damage caused by severe environments. Superoxide (oxygen radical) is normally produced in hydrated tissues. It is controlled by free radical scavenging reactions. One such scavenger is the enzyme superoxide dismutase (SOD). Under water stress, production of excess free radicals may occur in dehydrated plant tissues and this probably damages the membranes by causing peroxidation of the lipid components. So far few studies have been done to determine if drought injury is correlated with the free radical mechanism. In the present study, the SOD activities in wheat seedlings under water stress have been investigated by measuring the photoreduction of nitro blue tetrazolium using a spectrometric method. Meanwhile, the viabilities of wheat seedlings during drying were followed by tetrazolium test. These results provided information on the relationship between SOD activity and the dehydration tolerance of the plant. Results indicated that SOD activity changed with the time after germination. The activity of SOD of 24 h seedlings was 1.9 times higher than those of 72 h seedlings based on fresh weight. SOD activity in shoot was also higher than in root. These results were consistent with the results obtained from rating of the viabilities of seedlings during drying. The 24h seedlings were more tolerant of dehydration than 72 h seedlings and root were more sensitive of drought than shoot. In addition, shoot and root tips showed the higher SOD activities than non-tip region and they also showed a higher survival ability upon dehydration. In dehydration and subsequent rehydration, SOD activity, different from many other enzymes in plants, increased rather than declined during drying. After rehydration SOD activity returned to nearly the original level. Therefore, the positive correlations were found to exist between SOD activity and dehydration tolerance. It is reasonable to suggest that SOD enzyme may play a protective role against damage caused by free radicals which may be produced excessively during dehydration in wheat seedling. 相似文献

Akt has emerged as a critical target for the development of anti-cancer therapies. It has been found to be amplified, overexpressed, or constitutively activated in numerous human malignancies with oncogenesis derived from the simultaneous promotion of cell survival and suppression of apoptosis. A valuable alternative to the more common ATP-mimetic based chemotherapies is a substrate-mimetic approach, which has the potential advantage of inherent specificity of the substrate-binding pocket. In this paper we present the development of high affinity non-peptidic, substrate-mimetic inhibitors based on the minimum GSK3β substrate sequence. Optimization of initial peptidic leads resulted in the development of several classes of small molecule inhibitors, which have comparable potency to the initial peptidomimetics, while eliminating the remaining amino acid residues. We have identified the first non-peptidic substrate-mimetic lead inhibitors of Akt 29a–b, which have affinities of 17 and 12 μM, respectively. This strategy has potential to provide a useful set of molecular probes to assist in the validation of Akt as a potential target for anti-cancer drug design. 相似文献

Abstract Indian meal moths, Plodia interpunctella Hübner (Lepidoptera: Pyralidae), have ears which are sensitive to high‐frequency calls produced by echolocating, insectivorous bats. The influence of artificially generated, high‐intensity, ultrasound signals (25 kHz, 106 dB SPL at 1 m distance) on different parameters involved in the odour‐mediated mating behaviour of this species and its potential use in population control was investigated. All moths flying towards olfactory cues in flight tunnel experiments reacted strongly to a 1 s ultrasound pulse by cessation of flight and falling out of the odour plume. The source contact proportion of both male moths orienting towards the female‐produced sex pheromone and of mated female moths orienting towards an oviposition cue was reduced by 40%, compared to unexposed moths. Calling females responded to the sound by retraction of the ovipositor or by falling to the ground. Long‐term exposure to repetitive pulses of ultrasound suppressed female calling by up to 27%. Furthermore, mating in plastic tents was disrupted by up to 58% in ultrasound‐treated tents using different sound regimens, compared to control tents. The results are discussed in relation to the potential use of ultrasound technology for the population control of pyralid stored product pests. 相似文献

The occurrence of outbreaks of cholera in Africa in 1970 and in Latin America in 1991, mainly in coastal communities, and the appearance of the new serotype Vibrio cholerae O139 in India and subsequently in Bangladesh have stimulated efforts to understand environmental factors influencing the growth and geographic distribution of epidemic Vibrio cholerae serotypes. Because of the severity of recent epidemics, cholera is now being considered by some infectious disease investigators as a "reemerging" disease, prompting new work on the ecology of vibrios. Epidemiological and ecological surveillance for cholera has been under way in four rural, geographically separated locations in Bangladesh for the past 4 years, during which both clinical and environmental samples were collected at biweekly intervals. The clinical epidemiology portion of the research has been published (Sack et al., J. Infect. Dis. 187:96-101, 2003). The results of environmental sampling and analysis of the environmental and clinical data have revealed significant correlations of water temperature, water depth, rainfall, conductivity, and copepod counts with the occurrence of cholera toxin-producing bacteria (presumably V. cholerae). The lag periods between increases or decreases in units of factors, such as temperature and salinity, and occurrence of cholera correlate with biological parameters, e.g., plankton population blooms. The new information on the ecology of V. cholerae is proving useful in developing environmental models for the prediction of cholera epidemics. 相似文献

Artemisinin (ART) is a sesquiterpene lactone natural product that is widely used to treat multi-drug resistant strains of malaria. Artemisinin and its derivatives are also selectively cytotoxic to cancer cells, which can be modulated by altering heme synthesis. Cytotoxicity to cancer cells is thought to involve generation of oxidative stress, although conflicting data exist. We have analyzed reactive oxygen species (ROS) generation using the fluorescent probes 2′,7′-dichlorodihydrofluorescein diacetate (DCF) and dihydroethidine (HET) upon exposure to dihydroartemisinin (DHA) in Molt-4 leukemia cells. HET fluorescence correlated with dose-dependent DHA-induced cytotoxicity, increased within 30 min of DHA exposure, and was significantly enhanced by increasing heme synthesis. Protein levels of copper,zinc-superoxide dismutase (CuZnSOD), manganese-superoxide dismutase (MnSOD), catalase, and glutathione peroxidases 1/2 were also found to increase with DHA exposure. 4-hydroxy-tempol (TEMPOL) and DF-Mn, MnSOD mimetics, could significantly inhibit ROS generation and reduce cell death. Seeds and fruit of 38 anemochorous species were dropped in still air to simulate their descent under natural conditions. Fall rate and lateral distance were recorded as indices of wind-borne dispersal capability. Differences in fall rates among plumed species were dependent on interspecific variation in diaspore weight and plume area, while fall rates of winged species were strongly differentiated by contrasting wing shapes. In Acer platanoides and Asclepias syriaca, representing wing and plume architectures, respectively, the range of diaspore weight was artificially extended by removing embryos or adding lead weights. In both of these species, rate of descent of altered diaspores was controlled by weight relative to wing or plume area. The wing morphology of A. platanoides showed lower fall rates than the plumed A. syriaca above 45 mg, while the plume morphology of A. syriaca achieved lower fall rates below this weight. Compared with wide variation in diaspore weight, members of the Compositae showed relatively low variation in plume loading (diaspore weight/plume area) and fall speed. These observations suggest functional and phyletic constraints on diaspore architecture. Such constraints may limit evolutionary change in diaspore size and performance. 相似文献

Aquatic vertebrates are reported from several facies of the Late Mississippian (Chesterian/Elviran/Serpukhovian) Buffalo Wallow Formation in western Kentucky, USA. Rhizodont bones and the partially articulated skeleton of a large anthracosaur (proterogyrinid) were found in rocks interpreted as a fluvial‐estuarine palaeochannel. Smaller, disarticulated tetrapod remains (anthracosaurs, whatcheeriids) were found in a weathered siltstone in an apparent channel margin‐sand flat facies. A putative oxbow‐abandoned channel facies contains skeletal elements of rhizodonts (cf. Rhizodus), colosteiids, Gyracanthus, Ageleodus, Cynopodius, xenacanthoids and palaeonisiciforms. Near the top of the channel fill, lungfish (cf. Tranodis) are found in carbonate‐rich nodules, which appear to be aestivation burrows. A presumed lacustrine facies contained a near‐complete colosteid. Thinning section, palaeosols, pedogenically‐altered carbonates and missing strata suggest tectonic and climatic overprints upon these depositional sequences. Multiple, incised channels in a low‐accommodation setting are interpreted to have provided local faunal traps for aquatic vertebrates. Late Mississippian palaeoclimate changes may have caused water table fluctuations, which might have aided in trapping and preserving aquatic vertebrates. 相似文献
