The isolation of Saumarez Reef virus, a new flavivirus, from bird ticks Ornithodoros capensis and Ixodes eudyptidis in Australia.

Strains of a new flavivirus, for which the name Saumarez Reef Virus is proposed, were isolated from seabird ticks collected from four localities. Two strains were isolated from ticks of the species Ornithodoros capensis Neumann 1901 collected from the nests of Sooty Terns, Sterna fuscata Linnaeus 1766 on coral cays off the east coast of Queensland, Australia. The other three strains were isolated from ticks of the species Ixodes eudyptidis Maskell 1885 taken from two dead Silver Gulls Larus novaehollandiae Stephens 1826 in northern Tasmania. The new virus was compared serologically with 50 other flaviviruses at the Yale Arbovirus Research Unit and was found to be most closely related to Tyuleniy virus.     

Early gene interaction during prepupal expression of Drosophila arginine kinase

Arginine kinase displays a distinctive rise and fall in specific activity and specific protein levels during the prepupal stage of Drosophila development with maximal activity occurring at morphological stage P3. This developmentally regulated peak is under the influence of ecdysone. Altered doses of the major ecdysone-inducible “early” genes at cytological regions 75B and 2B5 alter this pattern of expression while altered doses of another major “early” gene at 74EF have no effect. We hypothesize that a product of the 2B5 locus and a product of the 75B locus interact to effect this developmental pattern of expression of Drosophila arginine kinase. © 1992 Wiley-Liss, Inc.     

The Ethnography of Landscape: GIS and Remote Sensing in the Study of Forest Change in West African Guinea Savanna

In this paper we explore the application of Amazonian forest change models to the Guinea savanna environment of western Africa. In particular, we examine recent research documenting the growth of peri-village forest islands in the area of Kissidougou, Guinea, which purports to challenge the pervasive view of Guinea savanna deforestation brought about by human activity. As a more systematic approach to the analysis of human/environmental interactions, we propose an "ethnography of landscape," in which the findings of detailed, local ethnographic and ecological case studies are combined with remotely sensed information on what the wider region looks like as it changes over time. In pursuing this research, we employ aerial photography and Geographic Information Systems (GIS) techniques to examine our study area of Kilimi in northwestern Sierra Leone. We show that a Kissidougou-type model of peri-village forest island growth is not applicable to this region, where other environmental change processes are underway. In future work, we propose to employ Landsat satellite image archives to examine, monitor, and compare processes of ecological change throughout the Guinea savanna. The use of spun columns in the purification of double-stranded cDNA appropriate for tailing

A reliable estimation of the efficiency of adding homopolymeric tails to double-stranded cDNA molecules by terminal deoxynucleotidyl transferase is extremely important in the construction of cDNA libraries. Appreciable differences in transformation efficiency result when the homopolymer tails to be annealed are of inadequate length. We report here that the use of a Sephadex G-50 spun column to remove oligo(dT)12-18 frequently coprecipitating in ethanol with the cDNA results in a more dependable estimation of tail lengths.     

Large-scale production of a therapeutic protein in transgenic tobacco plants: effect of subcellular targeting on quality of a recombinant dog gastric lipase

A recombinant dog gastric lipase with therapeutic potential for the treatment of exocrine pancreatic insufficiency was expressed in transgenic tobacco plants. We targeted the protein using two different signal sequences for either vacuolar retention or secretion. In both cases, an active glycosylated recombinant protein was obtained. The recombinant enzymes and the native enzyme displayed similar properties including acid resistance and acidic optimum pH. The proteolytic maturation and the specific activity of the recombinant proteins, however, were found to be dependent on subcellular compartmentalization. Expression levels of recombinant dog gastric lipase were about 5% and 7% of acid extractable plant proteins for vacuolar retention and secretion respectively. This expression system already has allowed the production of tens of grams of purified lipase through open-field culture of transgenic tobacco plants.     

Long-term effects of hydropower installations and associated river regulation on River Shannon eel populations: mitigation and management

The Shannon, Ireland’s largest river, is used for hydroelectricity generation since 1929. Subsequently, the Electricity Supply Board assumed responsibility for management of its eel stocks, due to the impact of the hydro-dam on recruitment to the commercial fishery. In order to negate a decline in juvenile recruitment resulting from the installation of hydroelectric facilities, management was focused on stocking lakes with elvers and fingerling eels. These were trapped at the hydropower facilities and in estuarine tributaries during their up-stream migrations. Due to the decline of natural recruitment in more recent times, attempts have also been made to develop an estuarine glass eel fishery. Stock levels are then monitored through annual surveys of the population trends of juvenile (glass eel, elver), growing phase (yellow eel) and downstream migrating pre-spawners (silver eels). Survey results and fishery management programmes are reviewed in this article. In addition to the long-term effects the hydroelectric facilities have had on the stock levels, there is also an annual effect on the migratory patterns of downstream migratory silver eels. In the lower reaches of the river system flow rates are regulated by the hydroelectric stations. We review previous work that had highlighted the importance of flow in determining the timing of the silver eels migrations, and assess the relationship between flow and migration in more detail through the use of hydroacoustic and telemetric studies. Current research on seaward migrating silver eel populations, suggests that spawner escapement rates can most effectively be increased by trapping migrating eels at fishing weirs located up-stream of the power station and transporting them towards the estuary.
