Hemicellulose from aspen (Populus tremula) was isolated by an alkali extraction method, which was followed by hydrogen peroxide treatment, ultrafiltration and recovery by spray drying. The sugar composition and lignin content were monitored with HPLC at each step of the separation procedure. Size-exclusion chromatography showed a polymeric hemicellulose of relatively high molar mass. The product was characterized by 1H and 13C NMR spectroscopy and was found to be composed of a linear (1→4)-β-linked -xylose main chain with a 4-O-methyl-- -glucuronic acid substituting the 2-position of approximately every eighth xylose unit. Lignin and O-acetyl groups had largely been removed in the separation process. The xylan was soluble in hot water, and the film forming properties were examined at various mixtures of the hemicellulose and chitosan. These films formed hydrogels with a high swelling capacity at certain compositions. The morphologies of the films were examined with wide angle X-ray spectroscopy, and a pure xylan film was found to be crystalline, which was suggested to be a consequence of the lack of O-acetyl groups. The crystallinity of the films was found to decrease with an increasing amount of chitosan, and the film of chitosan alone showed no crystallinity. The cohesive forces of the hydrogels are suggested to be the result of the crystalline arrangement of the polymers and of electrostatic interactions between acidic groups in the hemicellulose and amine groups in the chitosan.  相似文献

Diagnosis of bacterial sepsis in preterm neonates can be difficult when using serum markers that rely on physiological changes because these changes may not necessarily be the result of bacterial infections alone. This retrospective investigation explores the potential use of the DNA methylation pattern of CpG sites in the promoter region of the calcitonin-related polypeptide α (CALCA) gene as an epigenetic biomarker for bacterial sepsis in preterm newborns. Four novel changes in the DNA methylation of eight CpG sites were detected in this gene and are present only in neonates with bacterial sepsis: (1) partial methylation at -769 CpG in gram-negative or gram-positive early onset sepsis (EOS) and late onset sepsis (LOS) episodes; (2) demethylation of 8 CpGs in gram-negative EOS followed by LOS (ELS) and in gram-negative EOS; (3) demethylation of 7 CpGs in gram-positive ELS and gram-positive EOS; (4) -771 C:G > T:A; 5′ de novo -778 CpG mutation on both alleles in EOS. These changes were not detected in birth weight and gestational age matched controls or in newborns with isolated infections. Our results indicate that the DNA methylation pattern of the promoter region of the CALCA gene varies in different types of bacterial preterm sepsis, thus suggesting a potential use as an epigenetic biomarker. A prospective confirmation of these results is essential.  相似文献

Infections of the eye can rapidly damage important functional structures and lead to permanent vision loss or blindness. Broad-spectrum antibiotics should be administered to the appropriate site of infection as soon as a diagnosis is made. Topical drops are preferred for corneal and conjunctival infections. Intravitreal antibiotics, and possibly subconjunctival and parenteral antibiotics, are preferred for endophthalmitis. Parenteral antibiotics are recommended for infection in deep adnexal structures. Adherent-invasive Escherichia coli (AIEC) bacteria isolated from Crohn's disease patients are able to extensively replicate within macrophages in large vacuoles. The mechanism by which AIEC bacteria survive within phagocytic cells is unknown. This report describes the maturation of AIEC LF82-containing phagosomes within J774 macrophages. LF82-containing phagosomes traffic through the endocytic pathway as shown by the sequential acquisition and loss of EEA1 and Rab7 and by accumulation of Lamp-1, Lamp-2 and cathepsin D. We demonstrated that AIEC LF82-containing phagosomes mature into active phagolysosomes where bacteria are exposed to low pH and to the degradative activity of cathepsin D. Finally, we showed that an acidic environment is necessary for replication of AIEC LF82 bacteria within J774 macrophages. Thus, evidence is provided that AIEC LF82 bacteria do not escape from the endocytic pathway but undergo normal interaction with host endomembrane organelles and replicate within acidic and cathepsin D-positive vacuolar phagolysosomes.  相似文献

Due to various analytical challenges, so far no proteome has been sequenced completely.
