The structure-function relationship of the human integrin VLA-4 (alpha 4/beta 1; CD49d/CD29), has been studied in the human B-cell line Ramos by immunochemical and functional analysis. Ramos cells expressed the 150-kDa non-proteolyzed form of the alpha 4 chain, which could be digested upon mild trypsin treatment to generate the 80- and 65-kDa proteolyzed forms, as well as alpha 4 polypeptides of 55 and 50 kDa. In addition, treatment of Ramos cells with high doses of pronase predominantly yielded the 55- and 50-kDa alpha 4 peptides. The trypsin-generated 80- and 65-kDa alpha 4 polypeptides, but not the 55- and 50-kDa fragments, were able to associate with the beta 1 chain. Distinct anti-VLA-4 mAb against four different alpha 4 epitopes, referred to as epitopes A, B1, B2, and C, recognized the 150-kDa alpha 4 chain both associated or non-associated with the beta 1 chain. The alpha 4 proteolytic forms of 80, 65 and 50 kDa were precipitated by the anti-alpha 4 mAb directed against the four different alpha 4 epitopes. On the other hand, the 55-kDa alpha 4 peptide was present in precipitates from anti-alpha 4 mAb specific for epitopes A, B1 and C, but absent in precipitates from the anti-alpha 4 mAb specific for epitope B2. The different adhesive capacities of the VLA-4 integrin, namely the interaction with a 38-kDa fibronectin fragment containing the CS-1 region of plasma fibronectin (Fn-38), the binding to the vascular cell adhesion molecule-1 (VCAM-1), or the ability to mediate the anti-alpha 4-induced cell aggregation, were not altered on VLA-4 from cells upon mild trypsin treatment, when compared to non-treated cells. However, the 55- and 50-kDa alpha 4 forms generated by high-dose pronase cell treatment, failed to mediate cell interaction with Fn-38 or VCAM-1 ligands, and cell aggregation could not be triggered through VLA-4 under these conditions.  相似文献

Three recently isolated catalase-negative mutants ofHansenula polymorpha lost the ability to grow on methanol but grew in media containing glucose, ethanol or glycerol. Their incubation in a medium with methanol resulted in an accumulation of hydrogen peroxide and cell death. During growth of a catalase-negative mutant in chemostat on a mixture of methanol and glucose, neither H2O2 accumulation nor cell death were observed up to the molar ratio of 10:1 of the two substrates. Cytochrome-c peroxidase and NADH-peroxidase activities were detected in the cells. In methylotrophic yeasts, catalase seems to be an enzyme characteristic of the metabolism of methanol but not needed for the metabolism of multicarbon substrates. The hydrogen peroxide produced during growth of the mutants on mixed substrates is detoxified by cytochrome-c peroxidase and other peroxidases. Translated by Č. Novotny  相似文献

No significant differences in the acetylene-reducing activity and evolution of H2 and CO2 nodulated roots ofPisum sativum inoculated with soilRhizobium populations from two soils with different acidities (Ruzyně soil 7.6; Lukavec soil 4.9) were observed.Rhizobium population from Lukavec soil formed nodules, exhibiting a higher H2 evolution. The experiments reported here are aimed at determining whether mutations deleting the function of the Sex-lethal (Sxl) gene are able to suppress the lethality of X0 clones, induced in females after the time when the state of activity of Sxl is irreversibly fixed by the ratio of the number of X chromosomes to sets of autosomes (X:A). This analysis was carried out by comparing the frequency of induced male clones (X0 constitution) in SxlfLS/+ and Sxl+/Sxl+ females, following irradiation at blastoderm and larval stages. The genotype used in these experiments, however, could also give rise to 2X; 2A cells homozygous for SxlfLS, and such cells would also differentiate male structures. To minimize this possibility, we have constructed a genotype made up of a ring and a rod X chromosome. In such ring-rod females the production of 2X; 2A clones homozygous for SxlfLS is a rather rare event, if possible at all. X0 male clones were produced in both types of females following irradiation at blastoderm stage, while X0 male clones were only observed in SxlfLS/+ females when irradiation took place at larval stage. In this latter case, the only X0 male clones were those that contained the SxlfLS mutation. These results support the idea of Sánchez & N?thiger (1983) that the X:A signal irreversibly sets the state of activity of Sxl at blastoderm stage, and in addition show that X0 clones generated after that time are viable if they contain a Sxl- mutation. These results are compatible with the idea of Sxl being the only gene that responds to the X:A signal.  相似文献

A collection of 216 well-characterized strains ofStaphylococcus, Micrococcus andStomatococcus was examined by a commercially available STAPHYtest system (Lachema, Brno, Czechoslovakia). The results of STAPHYtest agreed with those of conventional tests. The STAPHYtest permitted a clear-cut separation ofStaphylococcus fromMicrococcus andStomatococcus strains and correctly identified 104 of 145 (72%)Staphylococcus strains after 24 h of incubation. However, it allowed the identification only of 19 of 29 validly publishedStaphylococcus species. The STAPHYtest proved to be a simple and rapid system for the separation of staphylococci from micrococci and for the identification of most frequent clinically significant staphylococci.  相似文献

A new type of flow bioreactor designed to remove nitrate from water was developed. Denitrification activity of native Paracoccus denitrificans cells was used, the cells being separated from the refined medium by a semipermeable membrane. Relationships between the degree of nitrate conversion and the denitrification rate, on the one hand, and the volume flow rate and the amount of biomass, on the other, together with the results concerning denitrification during closed-circuit recirculation of the medium are discussed.  相似文献

Two pathways serve for assimilation of ammonia inParacoccus denitrificans. Glutamate dehydrogenase (NADP+) catalyzes the assimilation at a high NH4 + concentration. If nitrate serves as the nitrogen source, glutamate is synthesized by glutamate-ammonia ligase and glutamate synthase (NADPH). At a very low NH4 + concentration, all three enzymes are synthesized simultaneously. No direct relationship exists between glutamate dehydrogenase (NADP+) and glutamate-ammonia ligase inP. denitrificans, while the glutamate synthase (NADPH) activity changes in parallel with that of the latter enzyme. Ammonia does not influence the induction or repression of glutamate dehydrogenase (NADP+). The inner concentration of metabolites indicates a possible repression of glutamate dehydrogenase (NADP+) by the high concentration of glutamine or its metabolic products as in the case when NH4 + is formed by assimilative nitrate reduction. No direct effect of the intermediates of nitrate assimilation on the synthesis of glutamate dehydrogenase (NADP+) was observed.  相似文献

Summary The vocal repertoire of magpie (Pica pica) chicks consists of six calls: Begging Trill (BT), Soft Whistle (SW), Begging Scream (BS), Alarm Call (AC), Distress Call (DC) and Brief Contact Note (BCN). Both BT and SW have a tonal structure and their occurrence is restricted to the nestling period. At fledging, there is a gradual change from BT into BS and a sudden appearance of harsh calls similar to those of adult birds (AC, DC, BCN), without evident transitional forms with preceding tonal calls. Both the existence and the structural design of calls seem to be adapted for providing nestlings with immediate benefits linked to the two major chapters of allocation of parental care. Emission rates of BT increase with hunger motivation under laboratory conditions. Their structure suggests that they are easily located but liable to suffer from environmental degradation. BS of fledglings may be more resistant to degradation, a trait which may facilitate the identification by parents of their own offspring. Both AC and DC attract parents to defend the nest against potential predators, and their structure make them to be easily located and detectable at long distances. BCN are given by fledglings during bouts of locomotory activity (exploration and play) and they probably help in maintaining the cohesion of the group under conditions of poor visibility. In accordance, this call may be fairly located at short distances. The function of SW was unclear. It is given during periods of nestling inactivity between begging bouts, and could be easily elicited by tactile and auditory stimuli. After laboratory experiments, it is concluded that SW serve to indicate parents that nestlings are in good condition, hence to benefit from the parental willingness to invest in a brood with high prospects of survival. Since (i) there is a widespread lack of continuity in the development of adult vocalizations starting from nestling calls, and (ii) nestling calls seem to have evolved to provide birds with benefits in the short-term, these facts argue against the prevailing idea that the main function of calls early in ontogeny is to act as precursors of adult vocalizations.
Zusammenfassung Das Lautrepertoire von Elsternestlingen besteht aus 6 Lautäußerungen: Betteltrillern (BT), Sanftes Pfeifen (SP), Bettelkreischen (BK), Alarmruf (AR), Angstschreien (AS) und kurzer Kontaktruf (KR). BT und SP sind tonal und treten nur während der Nestlingsperiode auf. Zum Zeitpunkt des Ausfliegens geht BT graduell in BK über und plötzlich treten auch ohne vorausgehende tonale Übergangsformen rauhe Rufe ähnlich denen der Altvögel auf (AR, AS, KR). Sowohl die Existenz als auch die Struktur der Lautäußerungen scheinen als Anpassungen im Zusammenhang mit einer Optimierung der Brutpflege zu interpretieren zu sein. Die Emissionsrate von BT steigt unter Laborbedingungen mit der Hungermotivation. Die Struktur legt nahe, daß BT leicht geortet werden kann, aber auch leicht von der Umgebung verschluckt wird. BS der flüggen Jungen scheint davon weniger betroffen zu sein, so daß die Eltern leichter ihre Jungen identifizieren können. AR und AS veranlassen die Altvögel, ihr Nest gegenüber potentiellen Prädatoren zu verteidigen; die Struktur der Laute begünstigt ihre Ortung und Wahrnehmung über größere Entfernungen. KR werden von den flüggen Jungen bei lebhafter lokomotorischer Aktivität (Exploration, Spiel) geäußert; sie tragen vielleicht dazu bei, die Gruppe auch unter erschwerten optischen Kontaktmöglichkeiten zusammenzuhalten. Die Funktion von SP blieb unklar. SP war während inaktiver Perioden zwischen Bettelverhalten zu hören und konnte leicht durch taktile und akustische Reize ausgelöst werden. Nach Laborexperimenten ist zu schließen, daß SP den Eltern Wohlbefinden der Jungen anzeigt und so Bereitschaft zur Investition in eine Brut mit hoher Überlebenswahrscheinlichkeit fördert. Daß (1) zwischen den Rufen der Altvögel und dem Repertoire der Nestlingen weitgehend keine kontinuierlichen Übergänge festzustellen und (2) Nestlingsrufe offensichtlich im Hinblick auf kuzfristige Gewinne zu interpretieren sind, spricht gegen die allgemein vorherrschende Ansicht, das Lautrepertoire in frühen Stadien der Ontogenie sei hauptsächlich ein Vorläufer der Adultlaute.

Summary It is apparent in the genetic code that amino acids of similar chemical nature have similar codons. I show how through successive codon captures (multiple rounds of Osawa-Jukes type reassignments), complete codon swappings in an unfavorable genetic code are evolutionarily feasible. This mechanisms could have complemented the ambiguity reduction and the vocabulary extension processes of codon-amino acid assignments. Evolution of wobble rules is implied. Transfer RNA molecules and synthetases may still carry memories of it.  相似文献
