The qualitative and quantitative composition of the essential oils obtained from wild Sicilian rosemary plants (Rosmarinus officinalis L.) has been investigated. Samples were collected in Sicily after an ideal division of the island into three large areas, namely Val Mazara (M), Val di Noto (N) and Val Demona (D), according to an ancient and historical territorial subdivision of the island. The essential oils were obtained by hydrodistillation and analyzed by GC-FID-MS, identifying 100 compounds representing more than 96% of the oils. Monoterpenes, both hydrocarbons and oxygenated, were the most highly represented components: the former with a range of 21–68% and the latter with a range of 29–79%. On the basis of the results of this study, comprising a cluster analysis, and on the commonly accepted subdivision of rosemary essential oils, namely cineoliferum, verbenoniferum and camphoriferum chemotypes, most of the wild Sicilian rosemary samples must be classified as cineoliferum chemotype. 相似文献
An extensive survey of wild Sicilian oregano was made. A total of 57 samples were collected from various sites, followed by taxonomic characterization from an agronomic perspective. Based on morphological and production characteristics obtained from the 57 samples, cluster analysis was used to divide the samples into homogeneous groups, to identify the best biotypes. All samples were analyzed for their phytochemical content, applying a cascade‐extraction protocol and hydrodistillation, to obtain the non volatile components and the essential oils, respectively. The extracts contained thirteen polyphenol derivatives, i.e., four flavanones, seven flavones, and two organic acids. Their qualitative and quantitative characterization was carried out by LC/MS analyses. The essential oils were characterized using a combination of GC‐FID and GC/MS analyses; a total of 81 components were identified. The major components of the oils were thymol, p‐cymene, and γ‐terpinene. Cluster analysis was carried out on both phytochemical profiles and resulted in the division of the oregano samples into different chemical groups. The antioxidant activity of the essential oils and extracts was investigated by the Folin? Ciocalteau (FC) colorimetric assay, by UV radiation‐induced peroxidation in liposomal membranes (UV‐IP test), and by determining the O ‐scavenging activity. 相似文献
The binding sites for xanthophylls in the CP29 antenna protein of higher plant Photosystem II have been investigated using recombinant proteins refolded in vitro. Despite the presence of three xanthophyll species CP29 binds two carotenoids per polypeptide. The localization of neoxanthin was studied producing a chimeric protein constructed by swapping the C-helix domain from CP29 to LHCII. The resulting holoprotein did not bind neoxanthin, confirming that the N1 site is not present in CP29. Neoxanthin in CP29 was, instead, bound to the L2 site, which is thus shown to have a wider specificity with respect to the homologous site L2 in LHCII. Lutein was found in the L1 site of CP29. For each site the selectivity for individual xanthophyll species was studied as well as its role in protein stabilization, energy transfer, and photoprotection. Putative xanthophyll binding sequences, identified by primary structure analysis as a stretch of hydrophobic residues including an acidic term, were analyzed by site-directed mutagenesis or, in one case, by deleting the entire sequence. The mutant proteins were unaffected in their xanthophyll composition, thus suggesting that the target motifs had little influence in determining xanthophyll binding, whereas hydrophobic sequences in the membrane-spanning helices are important. 相似文献
Postprandial hypotension occurs frequently, and current management is suboptimal. Recent studies suggest that the magnitude of the fall in postprandial blood pressure (BP) may be attenuated by gastric distension. The aim of this study was to determine the effect of gastric distension on the hypotensive response to intraduodenal (ID) glucose. Eight healthy subjects (5 males, 3 females, aged 65-76 years) received an ID infusion of either 1) 50 g glucose in 300 ml saline (ID glucose) over 60 min (t=0-60 min), 2) 50 g glucose in 300 ml saline over 60 min and intragastric (4) infusion of 500 ml water between t=7-10 min (IG water and ID glucose), or 3) ID saline (0.9%) infusion over 60 min and IG infusion of 500 ml water (IG water and ID saline) all followed by ID saline infusion for another 60 min (t=60-120 min) on three separate days. BP and heart rate (HR) were measured. Gastric emptying (GE) of the IG water was quantified by two-dimensional ultrasonography. Between t=0-60 min, systolic and diastolic BP was greater (P<0.05 for both) with IG water and ID saline compared with IG water and ID glucose, and less (P<0.05 for both) with ID glucose compared with IG water and ID glucose. These effects were evident at relatively low IG volumes (approximately 300 ml). GE was faster with IG water and ID saline when compared with IG water and ID glucose. We conclude that, in healthy older subjects, IG administration of water markedly attenuates the hypotensive response to ID glucose, presumably as a result of gastric distension. 相似文献
One of the most substantial and established environmental risk factors for neurological and psychiatric disorders is stress exposure, whose detrimental consequences hinge on several variables including time. In this regard the gestational period is known to present an intrinsic vulnerability to environmental insults and thus stressful events during pregnancy can lead to severe consequences on the offspring’s brain development with long-term repercussions throughout adulthood. On this basis, we investigated the long-lasting impact of prenatal stress exposure on the susceptibility to the experimental autoimmune encephalomyelitis (EAE), a well-established murine model of multiple sclerosis. Although stress is considered a triggering factor for this chronic, progressive, autoimmune disease, little is known about the underlying mechanisms. To this end, EAE was induced by immunization with MOG35-55/CFA and pertussis toxin administration in adult female C57BL/6 mice born from control or stressed dams exposed to restraint stress during the last days of gestation. Our results demonstrate that gestational stress induces a marked increase in the severity of EAE symptoms in adulthood. Further, we highlight an altered maturation of oligodendrocytes in the spinal cord of prenatally stressed EAE mice, as indicated by the higher levels of GPR17, a marker of immature oligodendrocyte precursor cells. These behavioral and molecular alterations are paralleled by changes in the expression and signaling of the neurotrophin BDNF, an important mediator of neural plasticity that may contribute to stress-induced impaired remyelination. Since several already marketed drugs are able to modulate BDNF levels, these results pave the way to the possibility of repositioning these drugs in multiple sclerosis.
Our study was designed to examine how components of complex mixtures can
inhibit the binding of other components to receptor sites in the olfactory
system of the spiny lobster Panulirus argus. Biochemical binding assays
were used to study how two- to six-component mixtures inhibit binding of
the radiolabeled odorants taurine, L-glutamate and
adenosine-5'-monophosphate to a tissue fraction rich in dendritic membrane
of olfactory receptor neurons. Our results indicate that binding inhibition
by mixtures can be large and is dependent on the nature of the odorant
ligand and on the concentration and composition of the mixture. The binding
inhibition by mixtures of structurally related components was generally
predicted using a competitive binding model and binding inhibition data for
the individual components. This was not the case for binding inhibition by
most mixtures of structurally unrelated odorants. The binding inhibition
for these mixtures was generally smaller than that for one or more of their
components, indicating that complex binding interactions between components
can reduce their ability to inhibit binding. The magnitude of binding
inhibition was influenced more by the mixture's precise composition than by
the number of components in it, since mixtures with few components were
sometimes more inhibitory than mixtures with more components. These
findings raise the possibility that complex binding interactions between
components of a mixture and their receptors may shape the output of
olfactory receptor neurons to complex mixtures.
