AIMS: To determine whether Ha-AP10, a member of the plant lipid transfer proteins (LTPs) family produces a direct cytotoxic effect on fungal cells mediated by membrane permeabilization. LTPs can inhibit fungal growth and are considered members of the ubiquitous class of antimicrobial peptides. However, the way they exert their effects on target cells is not yet understood. METHODS AND RESULTS: Viability assays demonstrate that Ha-AP10 acts as a fungicidal compound but no harmful effect is observed on plant cells. Liposome leakage assays show that the protein induces a moderate release of fluorescent probes encapsulated in model membranes, indicating its ability to interact with phospholipids. Using a fluorescent indicator of damage at the membrane level, we demonstrate that Ha-AP10 is able to induce the permeabilization of intact fungal spores in a dose-dependent manner. CONCLUSION: The results presented here demonstrate the permeabilization of fungal spores caused by Ha-AP10. SIGNIFICANCE AND IMPACT OF THE STUDY: To our knowledge, this is the first demonstration of fungal membrane damage by an LTP, giving a clue to elucidate the basis of its antimicrobial properties. 相似文献

A morphologically structured model is proposed to describe trends in biomass growth, substrate consumption, and antitumoral retamycin production during batch and fed-batch cultivations of Streptomyces olindensis. Filamentous biomass is structured into three morphological compartments (apical, subapical, and hyphal), and the production of retamycin, a secondary metabolite, is assumed to take place in the subapical cell compartment. Model accounts for the effect of glucose as well as complex nitrogen source on both the biomass growth and retamycin production. Laboratory data from bench-scale batch and fed-batch fermentations were used to estimate some model parameters by nonlinear regression. The predictive capability of the model was then tested for additional fed-batch and continuous experiments not used in the previous fitting procedure. The model predictions show fair agreement to the experimental data. The proposed model can be useful for further studies on process optimization and control. 相似文献

Lysosomal storage diseases result from defects in the activity of the lysosomal enzymes that break down macromolecules in the cell. These enzyme defects contribute to over 30 separate storage diseases that result in nearomuscular and intellectual impairment and, in some cases, early childhood death. This report describes a new method for identifying defects in the lysosomal enzymes and in the metabolic pathway that functions in the degradation of complex carbohydrates. The method involves identifying abnormal carbohydrates in the urine of affected patients using fluorescent carbohydrate tags and polyacrylamide gel electrophoresis. Currently, the method can be used as a simple screen for the identification of at least 12 different lysosomal storage diseases using a single electrophoretic procedure. Both oligosaccharidoses and mucopolysaccharidoses (MPS) can be identified, and in many cases the MPS subtype can be determined. In addition, the method can be used to confirm enzymatically the results of the initial screening test. We believe that this method will become extremely useful not only in the diagnosis of these diseases but in the management of patients on therapy. 相似文献

The study reports the functional affinity of an amidino derivative of pirenzepine, guanylpirenzepine, for muscarinic receptors mediating relaxation of rat duodenum, inhibition of rabbit vas deferens twitch contraction (both receptors previously classified as M1), guinea pig negative inotropism (M2) and ileal contraction (M3). Unlike pirenzepine, guanylpirenzepine discriminated between duodenum and vas deferens receptors, with a 30-fold greater affinity for the former subtype. The effects of chronic exposure to 5 g·1–1 cadmium or copper on the crustacean Isopod Asellus aquaticus (L.) were studied by analyzing survival and body growth in the first stages of the life-cycle and by determining fecundity and survival of embryo-bearing females. Juveniles survival is differently affected by the two metals in that embryonic development is more sensitive to cadmium while juvenile development is more sensitive to copper. Juvenile body growth is stimulated by cadmium and depressed by copper. Embryo-bearing female survival and fecundity are significantly reduced by cadmium but are not affected by copper. The consequence of environmental contamination by a sublethal cadmium or copper concentration is discussed. 相似文献

Light-induced betalain synthesis has been studied in three species of Amaranthaceae. The results obtained suggest that the response to short-term irradiation with white light is mediated by phytochrome, while the effects of prolonged illumination are controlled by the photosynthetic system. This article aims to contribute to the study of migration and nation-building in stateless nations. It explores the media portrayal of Italian immigration in Wales after the 1997 devolution. It assesses why Italian immigrant narratives have figured prominently in the Welsh media after devolution. It shows that, being seen as Wales's ‘model minority’, Italians have ideally served a reinforcement of the surrounding inclusive rhetoric, one that draws on an old-standing Welsh tolerance-myth. Thus, this article argues that stateless nations aiming to gain increasing autonomy can use their immigration history to construct and disseminate a tolerant and inclusive national image, one that aims to foster both indigenous people and migrants' sense of national belonging. 相似文献

The application of a selected Acetobacter pasteurianus strain for traditional balsamic vinegar production was assessed. Genomic DNA was extracted from biofilms after enrichment cultures on GYC medium (10% glucose, 1.0% yeast extract, 2.0% calcium carbonate) and used for PCR/denaturing gradient gel electrophoresis, 16S rRNA gene sequencing, and enterobacterial repetitive intergenic consensus/PCR sequencing. Results suggested that double-culture fermentation is suitable for traditional balsamic vinegar acetification.The use of selected starter cultures (SSC) in fermented food production is widely applied throughout the food industry, in particular for wine, dairy products, sausages, and a variety of vegetables (3, 11). The advantages of their use are related to the improvement of the process control, hygiene, and quality with respect to fermented foods obtained through indigenous fermentation. Vinegar is one of the fermented beverages produced without SSC inoculation, in both small- and large-scale production, mainly for the following reasons: (i) the majority of vinegars have low commercial value, and often technological innovation is not considered profitable, and (ii) there is limited knowledge of the ecophysiology of acetic acid bacteria (AAB) due to the difficulty in accessing, sampling, isolating, and preserving strains (2, 12, 15, 16, 17). Among vinegars, traditional balsamic vinegar (TBV) is an Italian aged condiment produced by “seed vinegar,” the so-called “mother of vinegar” that is an indigenous starter culture withdrawn from acetifying vinegar through back-slopping procedures. The raw material is a fermented and cooked grape must (here indicated as must) at a soluble solids content ranging from 20 to 60°Bx (10). TBV production is regulated by denomination of protected origin guidelines that specify procedures and final product features. In particular, the raw material characteristics, the production process (e.g., must cooking, alcoholic fermentation, acetic oxidation, and ageing), features of the production area (no environmental condition management is permitted), and analytical and sensorial parameters are stated as follows: acidity (not less than 4.5% [wt/wt], expressed as grams of acetic acid per 100 g of product), density at 20°C (not less than 1.240 g per liter), color, aroma, and taste. The production is performed in wood barrels, and the process is carried out by sequential refilling to acetify the must and replace the volume lost by evaporation. AAB grow on the surface of liquid by biofilm formation. No addition of any substance can be made except for the acetifying must as a starter (7). Microbial studies of TBV reported culture-dependent and -independent approaches to evaluating AAB occurrence in TBV musts (5, 10). These studies highlighted the occurrence of Gluconacetobacter europaeus as a widespread indigenous species, as well as Acetobacter pasteurianus, Acetobacter aceti, and Acetobacter malorum. However, no comprehensive studies of AAB diversity and the correlation between species occurrence and technological steps of TBV production have been published, due mainly to the difficulty of easy access to AAB microflora in vinegar matrix by both culture-dependent and -independent approaches.Regarding production technology, at least one drawback of current production procedures has been acknowledged. It concerns the difficulty of start-up acetification, which affects the minimum acidity value required for the final product. In fact, some studies showed that many variables regulate AAB growth and activity. Above all is the sugar concentration among substrates and the temperature among physical parameters. To efficiently control the acetification start-up, it is necessary to understand the function of AAB responsible for the initial colonization of musts and to investigate the microbial succession suitable to complete the acetification. Our previous researches on TBV showed that AAB strains exhibit different growing abilities. In particular, strains of Acetobacter pasteurianus grow quickly on laboratory synthetic media, wine, and cooked must. In contrast, strains belonging to G. europaeus do not grow or grow very slowly on cooked and fermented must (9, 10).The goal of this study was to implement a laboratory SSC to test it on a factory scale for TBV production purposes. In particular, we focused our attention on the effect of A. pasteurianus strain AB0220 on the acetification and dynamics of species at the end of the process. The SSC effectiveness was assessed by monitoring analytical parameters (acetic acid, ethanol, and pH), species succession, and strain persistence during three stages by the following molecular analyses: PCR/denaturing gradient gel electrophoresis (DGGE), 16S rRNA gene sequencing, and enterobacterial repetitive intergenic consensus (ERIC)/PCR sequencing using genomic DNA extracted from biofilms recovered on GYC (10% glucose, 1.0% yeast extract, 2.0% calcium carbonate) plates. 相似文献

AIMS: To identify and describe the indigenous yeast population involved in traditional balsamic vinegar (TBV) fermentation. METHODS AND RESULTS: Using the restriction analysis of the ribosomal region 5.8S (5.8S rRNA) and the internal transcribed spacers 1 and 2 (5.8S-ITS region) we were able to group 133 strains isolated from 17 cooked grape must samples into 10 different yeast species, included into 4 genera. Moreover, we sequenced the D1/D2 domains of the 26S rRNA and confirmed the reliability of each identification at species level. Most strains belonged to the genus Zygosaccharomyces. In particular, Zygosaccharomyces bailii was found in 41% of the samples, followed by Saccharomyces cerevisiae, Zygosaccharomyces pseudorouxii and Candida stellata. Strains belonging respectively to Zygosaccharomyces mellis, Zygosaccharomyces bisporus, Zygosaccharomyces rouxii, Hanseniaspora valbyensis, Hanseniaspora osmophila and Candida lactis-condensi species were also detected. Despite the great number of species recovered, the mtDNA restriction profiles showed low variability at strain level. Saccharomyces cerevisiae isolates with an higher degree of intraspecific variance were considered an exception. CONCLUSIONS: Many different indigenous yeast species were recovered and TBV yeasts population seems to be far more complex than what was reported in previous literature. SIGNIFICANCE AND IMPACT OF THE STUDY: This study has allowed us to gain a better understanding of the indigenous yeast species of TBV cooked must. 相似文献
