The Tapora Landcare Group, operating on the Okahukura Peninsula, has the long-term goal of making this region predator fenced. The aim of this study was to obtain information on the current status of avian biodiversity and the bird community across the band of coastal wetlands on the Okahukura Peninsula. Bird counts were conducted and playback lures used to detect three cryptic wetland species: fernbirds (Bowdleria punctata); spotless crakes (Porzana tabuensis); and banded rails (Gallirallus philippensis). Fernbirds and banded rails were detected at seven of the eight wetland sites sampled whereas spotless crakes were detected at two sites. The native species with the highest relative abundance across the eight sites were silvereyes (Zosterops lateralis) and South Island pied oystercatchers (Haematopus finschi). Changes in avian biodiversity over time in the region can now be monitored, and comprehensive long-term data on the status of avian biodiversity over time obtained.  相似文献

Essential role of lysyl oxidases in notochord development   总被引:1,自引:0,他引:1  
Recent studies reveal a critical role for copper in the development of the zebrafish notochord, suggesting that specific cuproenzymes are required for the structural integrity of the notochord sheath. We now demonstrate that beta-aminopropionitrile, a known inhibitor of the copper-dependent lysyl oxidases, causes notochord distortion in the zebrafish embryo identical to that seen in copper deficiency. Characterization of the zebrafish lysyl oxidase genes reveals eight unique sequences, several of which are expressed in the developing notochord. Specific gene knockdown demonstrates that loss of loxl1 results in notochord distortion, and that loxl1 and loxl5b have overlapping roles in notochord formation. Interestingly, while notochord abnormalities are not observed following partial knockdown of loxl1 or loxl5b alone, in each case this markedly sensitizes developing embryos to notochord distortion if copper availability is diminished. Likewise, partial knockdown of the lysyl oxidase substrate col2a1 results in notochord distortion when combined with reduced copper availability or partial knockdown of loxl1 or loxl5b. These data reveal a complex interplay of gene expression and nutrient availability critical to notochord development. They also provide insight into specific genetic and nutritional factors that may play a role in the pathogenesis of structural birth defects of the axial skeleton.  相似文献

The paper presents the second part of the review on a high-sensitive technique for time-resolved imaging and measurements of the 2D intensity profiles of millimeter-wave radiation by means of Visible Continuum Radiation emitted by the positive column of a medium-pressure Cs-Xe DC Discharge (VCRD method). The first part of the review was focused on the operating principles and fundamentals of this new technique [Plasma Phys. Rep. 43, 253 (2017)]. The second part of the review focuses on experiments demonstrating application of this imaging technique to measure the parameters of radiation at the output of moderate-power millimeter-wave sources. In particular, the output waveguide mode of a moderate-power W-band gyrotron with a pulsed magnetic field was identified and the relative powers of some spurious modes at the outputs of this gyrotron and a pulsed D-band orotron were evaluated. The paper also reviews applications of the VCRD technique for real-time imaging and nondestructive testing with a frame rate of higher than 10 fps by using millimeter waves. Shadow projection images of objects opaque and transparent for millimeter waves have been obtained using pulsed watt-scale millimeter waves for object illumination. Near video frame rate millimeter-wave shadowgraphy has been demonstrated. It is shown that this technique can be used for single-shot screening (including detection of concealed objects) and time-resolved imaging of time-dependent processes.  相似文献

Oligogalacturonic acids (OGAs), derived from plant cell wall pectin, have been implicated in a number of signal transduction pathways involved in growth, development and defense responses of higher plants. This study investigates the size range of OGAs capable of inducing ethylene synthesis in tomato plants, and demonstrates that in contrast with many other effects, only short chain OGAs are active. Oligomers across a range of DP from 2-15 were separated and purified to homogeneity by QAE-Sephadex anion exchange chromatography using a novel elution system. The OGAs were applied to tomato plants and assayed for their ability to induce ethylene gas release and changes in steady state levels of mRNA encoding the ethylene forming enzyme aminocyclopropane-1-carboxylic acid oxidase (ACO). The study demonstrated that only OGAs in the size range of DP4-6 were active both in eliciting ACO expression and in the production of ethylene.   相似文献

A protocol which involves sequential ammonium sulfate precipitation and zinc chelate chromatography to purify the HTLV-I Tax1 protein expressed in E. coli is described. The final Tax1 product is greater than 90% pure and the yield is approximately 1 mg per liter of liquid culture. The purified Tax1 protein is biologically active in indirect in vitro DNA binding assays and cellular NF-kB induction experiments.  相似文献

To elucidate the mechanisms of cuproprotein biosynthesis in the secretory pathway, a polyclonal antiserum was generated against hephaestin, a multicopper oxidase essential for enteric iron absorption. Immunoblot analysis and pulse-chase metabolic labeling revealed that hephaestin is synthesized as a single-chain polypeptide modified by N-linked glycosylation to a mature 161-kDa species. Cell surface biotinylation and immunofluorescent studies of polarized, differentiated colon carcinoma cells detected hephaestin on the basolateral surface under steady-state conditions. However, a decrease in the intracellular copper concentration resulted in a marked diminution in the abundance of this protein. Metabolic studies revealed no effect of decreased intracellular copper on the rate of hephaestin synthesis but a dramatic, specific, and reproducible increase in the turnover of the mature 161-kDa protein. Surprisingly, inhibitor studies revealed that this turnover occurs exclusively in the proteasome, and consistent with this finding, in vitro studies identified polyubiquitinated hephaestin under conditions abrogating copper incorporation into this protein. Taken together, these studies demonstrate the presence of a quality control system for posttranslational protein modification occurring beyond the endoplasmic reticulum that, in the case of hephaestin, directly links the rate of enteric iron uptake to nutritional copper status.  相似文献

Most natural populations of Drosophila melanogaster are polymorphic for two major electrophoretic variants at the esterase-6 locus. The frequency of the EST 6F allozyme is greatest in populations in warmer latitudes, whereas the EST 6S allozyme is predominant in colder latitudes. Latitudinal clines in electromorph frequencies are found on three continents. Purified preparations of the allozymes have been characterized for their pH optimum, substrate specificity, organophosphate inhibition, alcohol activation, thermal stability, and kinetic parameters. These and previous analyses of the EST 6 allozymes reveal that the two variants have differences in their physical and kinetic properties that may provide a basis for the selective maintenance of the polymorphisms and an explanation of the clinal variation observed in natural populations.   相似文献

The first part of the review is presented which is dedicated to the time-resolved method of imaging and measuring the spatial distribution of the intensity of millimeter waves by using visible continuum (VC) emitted by the positive column (PC) of a dc discharge in a mixture of cesium vapor with xenon. The review focuses on the operating principles, fundamentals, and applications of this new technique. The design of the discharge tube and experimental setup used to create a wide homogeneous plasma slab with the help of the Cs–Xe discharge at a gas pressure of 45 Torr are described. The millimeter-wave effects on the plasma slab are studied experimentally. The mechanism of microwave-induced variations in the VC brightness and the causes of violation of the local relation between the VC brightness and the intensity of millimeter waves are discussed. Experiments on the imaging of the field patterns of horn antennas and quasi-optical beams demonstrate that this technique can be used for good-quality imaging of millimeter-wave beams in the entire millimeter-wavelength band. The method has a microsecond temporal resolution and a spatial resolution of about 2 mm. Energy sensitivities of about 10 μJ/cm2 in the Ka-band and about 200 μJ/cm2 in the D-band have been demonstrated.  相似文献
