Agrobiodiversity plays a fundamental role in guaranteeing food security. However, still little is known about the diversity within crop and livestock species: the genetic diversity. In this paper we present a set of indicators of crop accession and breed diversity for different farm types at farm-level, which may potentially supply a useful tool to assess and monitor farming system agrobiodiversity in a feasible and relatively affordable way. A generic questionnaire was developed to capture the information on crops and livestock in 12 European case study regions and in Uganda by 203 on-farm interviews. Through a participatory approach, which involved a number of stakeholders, eight potential indicators were selected and tested. Five of them are recommended as potentially useful indicators for agrobiodiversity monitoring per farm: (1) crop-species richness (up to 16 crop species), (2) crop-cultivar diversity (up to 15 crop cultivars, 1–2 on average), (3) type of crop accessions (landraces accounted for 3 % of all crop cultivars in Europe, 31 % in Uganda), (4) livestock-species diversity (up to 5 livestock species), and (5) breed diversity (up to five cattle and eight sheep breeds, on average 1–2).We demonstrated that the selected indicators are able to detect differences between farms, regions and dominant farm types. Given the present rate of agrobiodiversity loss and the dramatic effects that this may have on food production and food security, extensive monitoring is urgent. A consistent survey of crop cultivars and livestock breeds on-farm will detect losses and help to improve strategies for the management and conservation of on-farm genetic resources. 相似文献
Zinc (Zn2+) has been recently recognized as a crucial element for male gamete function in many species although its detailed mechanism of action is poorly understood. In sea urchin spermatozoa, Zn2+ was reported as an essential trace ion for efficient sperm motility initiation and the acrosome reaction by modulating intracellular pH (pHi). In this study we found that submicromolar concentrations of free Zn2+ change membrane potential (Em) and increase the concentration of intracellular Ca2+ ([Ca2+]i) and cAMP in Lytechinus pictus sperm. Our results indicate that the Zn2+ response in sperm of this species mainly involves an Em hyperpolarization caused by K+ channel activation. The pharmacological profile of the Zn2+-induced hyperpolarization indicates that the cGMP-gated K+ selective channel (tetraKCNG/CNGK), which is crucial for speract signaling, is likely a main target for Zn2+. Considering that Zn2+ also induces [Ca2+]i fluctuations, our observations suggest that Zn2+ activates the signaling cascade of speract, except for an increase in cGMP, and facilitates sperm motility initiation upon spawning. The WWC1 gene has been genetically associated with human episodic memory performance, and its product KIdney/BRAin protein (KIBRA) has been shown to interact with the atypical protein kinase protein kinase M ζ (PKMζ). Although recently challenged, PKMζ remains a candidate postsynaptic regulator of memory maintenance. Here, we show that PKMζ is subject to rapid proteasomal degradation and that KIBRA is both necessary and sufficient to counteract this process, thus stabilizing the kinase and maintaining its function for a prolonged time. We define the binding sequence on KIBRA, a short amino acid motif near the C‐terminus. Both hippocampal knock‐down of KIBRA in rats and KIBRA knock‐out in mice result in decreased learning and memory performance in spatial memory tasks supporting the notion that KIBRA is a player in episodic memory. Interestingly, decreased memory performance is accompanied by decreased PKMζ protein levels. We speculate that the stabilization of synaptic PKMζ protein levels by KIBRA may be one mechanism by which KIBRA acts in memory maintenance.
Phenazine 5,10-dioxides are prodrugs for antitumor therapy that undergo hypoxic-selective bioreduction to form cytotoxic species. Here we investigate the expanded system benzo[a]phenazine 7,12-dioxides as selective hypoxic cytotoxin-scaffold. The clonogenic survival of V79 cells on aerobic and anaerobic conditions, conduct us to study antiproliferative activity on Caco-2 tumoral cells in normoxia. Electrochemical, DNA-interaction and DNA-damage studies were performed to establish the mode of action. The results demonstrated the potential biological properties of the studied scaffold being derivatives 6–10 structural hits for further chemical-modifications to become into therapeutics for solid tumors. Compounds 6 and 8 with cytotoxicity against V79 cells in both conditions (aerobia and anaerobia) were also cytotoxic against Caco-2 tumoral cells in aerobiosis. 相似文献
Human and animal bones from the Pre-Pottery Neolithic B site of Nevali Cori (southeast Anatolia) were analyzed with regard to stable carbon and nitrogen isotopes in bone collagen, and stable carbon and oxygen isotopes in bone carbonate. The reconstruction of the vertebrate food web at this site revealed that humans may have faced difficulties with meat procurement, since their stable-isotope ratios reflect a largely herbivorous diet. This is in contrast with the preceding Pre-Pottery Neolithic A contexts and late Neolithic sites in the Fertile Crescent, where humans are located at the top of the food chain. Conceivably, Nevali Cori represents a community in the transition from a hunting and gathering subsistence to an economy with agriculture and animal husbandry, since domesticated einkorn and sheep, pigs, and probably also goats are in evidence at the site. In the second half of the 9th millennium calibrated (cal.) BC, however, the contribution of stock on the hoof to the human diet still seems modest. Animals kept under cultural control obviously had a dietary spectrum different from their free-ranging relatives. We conclude that these animals had been deliberately nourished by their owners, whereby the overall low delta(15)N-signatures in both humans and livestock might result from the consumption of protein-rich pulses. 相似文献
In the last two decades, the analysis of stable isotopes of carbonate and collagen from archaeological bone finds became a useful tool in the reconstruction of ancient food webs. Nevertheless there is still only little information available about aquatic food webs, in particular concerning brackish water ecosystems. The Schlei Fjord in Schleswig-Holstein, Germany, is such an ecosystem and was investigated to determine stable isotopic values for nitrogen, carbon and oxygen from bone collagen and carbonate in archaeological bone finds. Wild birds, mammals and fish bones from the Viking settlement Haithabu and the medieval town of Schleswig have been analyzed in this study to determine stable isotope values for marine and limnic species and to investigate possible isotopic gradients for mixing fresh water and salt water ecosystems. 相似文献
During normal developmental tissue growth and in a number of diseases of the cardiopulmonary system, adventitial and interstitial fibroblasts are subjected to increased mechanical strain. This leads to fibroblast activation and enhanced collagen synthesis, but the underlying mechanisms involved remain poorly understood. In this study, we have begun to identify and characterize mechanical strain-responsive elements in the rat procollagen alpha 1(I) (COL1A1) gene and show that the activity of COL1A1 promoter constructs, transiently transfected into cardiac fibroblasts, was increased between 2- and 4-fold by continuous cyclic mechanical strain. This was accompanied by an approximately 3-fold increase in the levels of total active transforming growth factor-beta (TGF-beta) released into the medium. Inclusion of a pan-specific TGF-beta neutralizing antibody inhibited strain-induced COL1A1 promoter activation. Deletion analysis revealed the presence of two potential strain response regions within the proximal promoter, one of which contains an inverted CCAAT-box overlapping a GC-rich element. Both mechanical strain and exogenously added TGF-beta1 enhanced the binding activity of CCAAT-binding factor, CBF/NF-Y, at this site. Moreover, this element was sufficient to confer strain-responsiveness to an otherwise unresponsive SV40 promoter. In summary, this study demonstrates that strain-induced COL1A1 promoter activation in cardiac fibroblasts is TGF-beta-dependent and involves increased binding of CCAAT-binding factor at the proximal promoter. Furthermore, these findings suggest a novel and potentially important TGF-beta response element in the rat COL1A1 gene. 相似文献
