Capacities and effects of cold or warm acclimation were investigated in two zoarcid species from the North Sea (Zoarces viviparus) and the Antarctic (Pachycara brachycephalum) by investigating temperature dependent mitochondrial respiration and activities of citrate synthase (CS) and NADP+ -dependent isocitrate dehydrogenase (IDH) in the liver. Antarctic eelpout were acclimated to 5°C and 0°C (controls) for at least 10 months, whereas boreal eelpout, Z. viviparus (North Sea) were acclimated to 5°C and to 10°C (controls). Liver sizes were found to be increased in both species in the cold, with a concomitant rise in liver mitochondrial protein content. As a result, total liver state III rates were elevated in both cold-versus and warm-exposed P. brachycephalum and Z. viviparus, with the highest rates in boreal eelpout acclimated to 5°C. CS and IDH activities in the total liver were similar in Z. viviparus acclimated to 5°C and 10°C, but decreased in those warm acclimated versus control P. brachycephalum. Enzyme capacities in the total liver were higher in eelpout from Antarctica than those from the North Sea. In conclusion, cold compensation of aerobic capacities in the liver seems to be linked to an increase in organ size with unchanged specific mitochondrial protein content. Despite its life in permanently cold climate, P. brachycephalum was able to reduce liver aerobic capacities in warm climate and thus, displayed a capacity for temperature acclimation.  相似文献

The investigation of fragment length distributions of plasmid DNA gives insight into the influence of localized energy distribution on the induction of DNA damage, particularly the clustering of double-strand breaks. We present an approach that determines the fragment length distributions of plasmid DNA after heavy-ion irradiation by using the Local Effect Model. We find a good agreement of our simulations with experimental fragment distributions derived from atomic force microscopy (AFM) studies by including experimental constraints typical for AFM. Our calculations reveal that by comparing the fragmentation in terms of fluence, we can uniquely distinguish the effect of different radiation qualities. For very high-LET irradiation using nickel or uranium ions, no difference between their fragment distributions can be expected for the same dose level. However, for carbon ions with an intermediate LET, the fragmentation pattern differs from the distribution for very high-LET particles. The results of the model calculations can be used to determine the optimal experimental parameters for a demonstration of the influence of track structure on primary radiation damage. Additionally, we compare the results of our model for two different plasmid geometries.  相似文献

Res. 170, 316-326 (2008).Intracellular calcium is an important second messenger that regulates many cell functions. Recent studies have shown that calcium ions can also regulate the cellular responses to ionizing radiation. However, previous data are restricted to cells treated with low-LET radiations (X rays, gamma rays and beta particles). In this work, we investigated the calcium levels in cells exposed to heavy-ion radiation of high LET. The experiments were performed at the single ion hit facility of the GSI heavy-ion microprobe. Using a built-in online calcium imaging system, the intracellular calcium concentrations were examined in HeLa cells and human foreskin fibroblast AG1522-D cells before and after irradiation with 4.8 MeV/nucleon carbon or argon ions. Although the experiment was sensitive enough to detect the calcium response to other known stimuli, no response to heavy-ion radiation was found in these two cell types. We also found that heavy-ion radiation has no impact on calcium oscillation induced by hypoxia stress in fibroblast cells.  相似文献

Filamentous fungi were isolated from estuarine sediments collected from two contaminated sites. One site was contaminated mainly by polycyclic aromatic hydrocarbon (PAH) with a concentration around 407?µg g?1 of different PAHs, and the other by different sources of industrial pollutants; both were compared to a pristine site. All three sites were located in the Baixada Santista, São Paulo State, Brazil. The aim of the present investigation was the isolation of filamentous fungi from pristine and industrially polluted sediments in order to assess the mycobiota present in those sites and to evaluate their tolerance to phenanthrene and pyrene. Most of the isolated fungi were mitosporic ascomycetes, including Aspergillus sp., Chrysosporium sp., Cyclothyrium spp., Gliocladium sp., Penicillium spp., Phoma spp., and Trichoderma spp. There were also representatives of sexual Ascomycetes, Basidiomycetes, and Zygomycetes. The results showed that 59% of the evaluated fungi were tolerant to pyrene and 30% to phenanthrene. Representatives of Trichoderma were the most tolerant among the filamentous fungi investigated. A representative of Penicillium simplicissimum was the only isolate tested that had a better growth in the presence of pyrene than in its absence.  相似文献

A lingering issue in the area of protein engineering is the optimal design of β motifs. In this regard, the framework provided by intestinal fatty acid binding protein (IFABP) was successfully chosen to explore the consequences on structure and function of the redesign of natural motifs. A truncated form of IFABP (Δ98Δ) served to illustrate the nonintuitive notion that the integrity of the β‐barrel can indeed be compromised with no effect on the ability to attain a native‐like fold. This is most likely the outcome of the key role played by the preservation of essential core residues. In the search for the minimal structural determinants of this fold, Δ98Δ offered room for further intervention. A dissection of this protein leads to a new abridged variant, Δ78Δ, containing 60% of the amino acids of IFABP. Spectroscopic analyses indicate that Δ78Δ retains substantial β‐sheet content and preserves tertiary interactions, displaying cooperative unfolding and binding activity. Most strikingly, this construct adopts a remarkably stable dimeric structure in solution. This phenomenon takes advantage of the inherent structural plasticity of this motif, likely profitting from edge‐to‐edge interactions between β‐sheets, whereas avoiding the most commonly occurring outcome represented by aggregation.  相似文献

The need for climate change mitigation and to meet increasing energy demands has led to a rise in the land area under bioenergy crops in many countries. There are concerns that such large-scale land conversion will conflict with food production and impact on the environment. Perennial biomass crops could be grown on more marginal agricultural land. However, for sustainable solutions, biomass yields will need to be sufficient and the wider implications of land-use changes considered. Here, focusing on Miscanthus in England as an example, we combined an empirical model with GIS to produce a yield map and estimated regional energy generation potentials after masking out areas covered by environmental and socio-economic factors which could preclude the planting of energy crops. Agricultural land quality and the distributions of currently grown food crops were then taken into account. Results showed that: (i) regional contrasts occur in the importance of different factors affecting biomass planting; (ii) areas with the highest biomass yields co-locate with food producing areas on high grade land, and; (iii) when such high grade land and unsuitable areas are excluded, a policy-related scenario for increased planting on 350,000 ha utilised 4–28% (depending on the region) of lower grade land and would not necessarily greatly impact on UK food security. We conclude that the GIS-based yield and suitability mapping described here can help identify important issues in bioenergy generation potentials and land use implications at regional or finer spatial scales that would be missed in analyses at the national level.  相似文献

Bioreactor stresses, including nutrient deprivation, shear stress, and byproduct accumulation can cause apoptosis, leading to lower recombinant protein yields and increased costs in downstream processing. Although cell engineering strategies utilizing the overexpression of antiapoptotic Bcl‐2 family proteins such as Bcl‐2 and Bcl‐xL potently inhibit apoptosis, no studies have examined the use of the Bcl‐2 family protein, Mcl‐1, in commercial mammalian cell culture processes. Here, we overexpress both the wild type Mcl‐1 protein and a Mcl‐1 mutant protein that is not degraded by the proteasome in a serum‐free Chinese hamster ovary (CHO) cell line producing a therapeutic antibody. The expression of Mcl‐1 led to increased viabilities in fed‐batch culture, with cell lines expressing the Mcl‐1 mutant maintaining ~90% viability after 14 days when compared with 65% for control cells. In addition to enhanced culture viability, Mcl‐1‐expressing cell lines were isolated that consistently showed increases in antibody production of 20–35% when compared with control cultures. The quality of the antibody product was not affected in the Mcl‐1‐expressing cell lines, and Mcl‐1‐expressing cells exhibited 3‐fold lower caspase‐3 activation when compared with the control cell lines. Altogether, the expression of Mcl‐1 represents a promising alternative cell engineering strategy to delay apoptosis and increase recombinant protein production in CHO cells. © 2009 American Institute of Chemical Engineers Biotechnol.
