The sequences encoding the QUAD1 RNAs were initially identified as four repeats in Escherichia coli. These repeats, herein renamed SIB, are conserved in closely related bacteria, although the number of repeats varies. All five Sib RNAs in E. coli MG1655 are expressed, and no phenotype was observed for a five-sib deletion strain. However, a phenotype reminiscent of plasmid addiction was observed for overexpression of the Sib RNAs, and further examination of the SIB repeat sequences revealed conserved open reading frames encoding highly hydrophobic 18- to 19-amino-acid proteins (Ibs) opposite each sib gene. The Ibs proteins were found to be toxic when overexpressed and this toxicity could be prevented by coexpression of the corresponding Sib RNA. Two other RNAs encoded divergently in the yfhL-acpS intergenic region were similarly found to encode a small hydrophobic protein (ShoB) and an antisense RNA regulator (OhsC). Overexpression of both IbsC and ShoB led to immediate changes in membrane potential suggesting both proteins affect the cell envelope. Whole genome expression analysis showed that overexpression of IbsC and ShoB, as well as the small hydrophobic LdrD and TisB proteins, has both overlapping and unique consequences for the cell.  相似文献

Cyclists seek to maximize performance during competition, and gross efficiency is an important factor affecting performance. Gross efficiency is itself affected by pedal rate. Thus, it is important to understand factors that affect freely chosen pedal rate. Crank inertial load varies greatly during road cycling based on the selected gear ratio. Nevertheless, the possible influence of crank inertial load on freely chosen pedal rate and gross efficiency has never been investigated. This study tested the hypotheses that during cycling with sub-maximal work rates, a considerable increase in crank inertial load would cause (1) freely chosen pedal rate to increase, and as a consequence, (2) gross efficiency to decrease. Furthermore, that it would cause (3) peak crank torque to increase if a constant pedal rate was maintained. Subjects cycled on a treadmill at 150 and 250W, with low and high crank inertial load, and with preset and freely chosen pedal rate. Freely chosen pedal rate was higher at high compared with low crank inertial load. Notably, the change in crank inertial load affected the freely chosen pedal rate as much as did the 100W increase in work rate. Along with freely chosen pedal rate being higher, gross efficiency at 250W was lower during cycling with high compared with low crank inertial load. Peak crank torque was higher during cycling at 90rpm with high compared with low crank inertial load. Possibly, the subjects increased the pedal rate to compensate for the higher peak crank torque accompanying cycling with high compared with low crank inertial load.  相似文献

In many dioecious bryophyte species, population sex ratios range from all female to all male. The focal species of the present study, the liverwort Marchantia inflexa, forms patches on rock and bark surfaces, and these differ widely in sex ratio at a rainforest field site in Trinidad. This analysis – to our knowledge the first modeling study of sex-ratio dynamics in a dioecious clonal organism – addresses abundances of male and female M. inflexa through time within an individual patch. We represent the life history of this species using seven different stages (non-reproductive, asexually reproductive, sexually reproductive males, non-reproductive, asexually reproductive, unfertilized and fertilized sexual females) and express their dynamics using ordinary differential equations. Some of the stages become more abundant as thalli extend over the substrate and may overgrow each other to capture space. Our simple representation of dynamics within the patch failed to stabilize the sex ratio: females gradually eliminated males at low to moderate disturbance frequency and males eliminated females at high disturbance frequency. This pattern did not hinge on whether sexual propagules could germinate within the patch, but asexual reproduction (via gemmae dispersed within the patch) played an important role. This suggests that the maintenance of sex in these populations may hinge on metapopulation structure and dynamics. Though sexual reproduction appears to be unimportant within patches, spores provide the primary means of recolonizing patches eliminated by large-scale disturbances. We found that shortly after the patch was fully occupied, the production of these wind-dispersed spores was maximized, but spore production declined thereafter as the sex ratio became increasingly biased toward one sex or the other. Much additional modeling and empirical work is needed to link within-patch dynamics across patches and account for dynamics at the metapopulation level.  相似文献

Flustra foliacea is a cheilostome marine bryozoan which, after initial horizontal growth during the first year, changes to erect growth thereafter. Appraising this slow-growing species for biotechnological use, required the development of cultivation methods for economical biomass production. Vegetative reproduction via cuttings, as employed in the horticulture of various plants, was tested. Fronds of F. foliacea were cut into pieces (10–150 mg fresh weight) and fixed in longitudinally cut silicon tubes. Bryozoa were fed with a mixture of Isochrysis galbana, Dunaliella tertiolecta and Cryptomonas sp. yielding growth rates of 0.3% ± 0.02 day?1of the initial fresh weight. Growth of the colony occurred firstly at the original growth margin, but later also at the transections, including the side fixed in the silicon tube, opposite to the growth margin. After three months of growth, the two layers of zooids split at the growth margin, forming two monolayers. This growth feature, not described previously for Flustra foliacea,was observed under experimental and natural conditions, and is interpreted as the beginning of one type of ramification. According to our findings, vegetative reproduction of F. foliacea under laboratory conditions is possible and might be an alternative to natural resources regarding biomass of this species.  相似文献

Early cellular events during secondary embryogenesis were studiedin a cork oak recurrent embryogenic system in which embryosarise either in a multicellular budding pathway from a compactmass of proliferation or from isolated single cells in friablecallus. The compact mass of proliferation originated from theepidermal cells at the hypocotyl whose growth and convolutionwas characterized by a decrease in the nucleus/cytoplasm ratioand a marked increase in storage products. The transition fromthe compact mass to meristematic primordia occurred at the peripheryand was accompanied by cell dedifferentiation and a drasticreduction of storage products. Meristematic primordia evolvedto globular embryos by the organization of a protodermis andtwo internal centres. Microscope analysis of friable callusshowed an hypothetical sequence from single cells to aggregatesof a few cells, meristematic cell clusters and globular embryos.Single cells showed typical features of embryogenic cells suchas rich cytoplasm and a large number of starch grains and lipidbodies. A progressive cell dedifferentiation and a drastic reductionof storage products was observed when aggregates of a few cellsand meristematic cell clusters were compared. Progressive bipolarizationin large meristematic cell clusters initiated globular embryoformation. The comparison of both embryogenic pathways at theultrastructural level showed that subcellular changes followa similar sequential pattern, especially with regard to thestorage products. The possible role of plastid extrusions andmultivesicular bodies in the changing pattern of starch metabolismduring embryogenesis is discussed. The effect of the presence of a familiar social partner on the interactions of saddle back tamarins with unfamiliar conspecifics was studied. Fifteen adult male-female pairs, of which six were composed of a castrated male and an intact female, served as subjects. All subject pairs were given two social encounters during which both mates met a strange male and two encounters during which they met a strange female. In addition, all subjects were given four encounters during which they met the same strangers while their own pair mates were absent. As a group, the subjects showed higher intensities of injurious aggression and of agonistic displays when they met strangers in the presence of their own pair mates. Females and castrated males, as subgroups, showed significant increases in most agonistic responses when they met strangers in the presence of their pair mates.
