Twisted fibrous extracellular matrices observed in section often show alternating clear and dark bands. Three different methods of observation (high voltage electron microscopy, shadowing of thin sections and stereoscopic views) show the presence of ruffling effects and relief at the surface of crab cuticle sections. These effects appear uneven on both sides of the sections. As shown in a series of diagrams, the localization of the microtomy artefact is a function of the orientation of the cuticle laminae relative to the knife direction, and this creates variations in the position and the extent of the microtomy effect over each lamina. Confirmation of this analysis is obtained in a particular geometrical situation which appears in sections of tubercles in the crab cuticle where the twisted plywood stratification is deformed into a dome. By shadowing thin sections, perpendicular to the tubercle axis, nested crescents are visualized on the surface of the samples. All observations demonstrate that the clear and dark lamellae are due to a microtomy artefact which is a three-dimensional process, and not, as usually considered, due to chemical or physical variations in the structure.  相似文献

The Pilbara Craton of Western Australia and the Barberton Greenstone Belt of the Kaapvaal Craton, South Africa, contain some of the oldest and best preserved Archaean rocks and microfossils in the world. Two stratigraphic horizons in the Pilbara Craton were drilled as part of a collaborative effort between France and Australia (the Pilbara Drilling Project) during August 2004, including the 3481 Ma Dresser Formation (Warrawoona Group) and 2724 Ma Tumbiana Formation (Fortescue Group). A new diamond drill hole was cored in August 2008 through part of the ~3250 Ma Fig Tree Group in the Barberton Greenstone Belt as part of a joint project between France and South Africa. These pristine diamond drill cores present a unique opportunity to constrain the chemistry of the earliest ocean, the composition of the atmosphere, and the settings and types of microbial ecosystems spanning the Archean Eon. These drill core samples can also provide new clues on the earliest metabolic pathways.  相似文献

Microsomal prostaglandin E synthase (mPGES)-1 is a newly identified inducible enzyme of the arachidonic acid cascade with a key function in prostaglandin (PG)E2 synthesis. We investigated the kinetics of inducible cyclo-oxygenase (COX)-2 and mPGES-1 expression with respect to the production of 6-keto-PGF and PGE2 in rat chondrocytes stimulated with 10 ng/ml IL-1β, and compared their modulation by peroxisome-proliferator-activated receptor (PPAR)γ agonists. Real-time PCR analysis showed that IL-1β induced COX-2 expression maximally (37-fold) at 12 hours and mPGES-1 expression maximally (68-fold) at 24 hours. Levels of 6-keto-PGF and PGE2 peaked 24 hours after stimulation with IL-1β; the induction of PGE2 was greater (11-fold versus 70-fold, respectively). The cyclopentenone 15-deoxy-Δ12,14prostaglandin J2 (15d-PGJ2) decreased prostaglandin synthesis in a dose-dependent manner (0.1 to 10 μM), with more potency on PGE2 level than on 6-keto-PGF level (-90% versus -66% at 10 μM). A high dose of 15d-PGJ2 partly decreased COX-2 expression but decreased mPGES-1 expression almost completely at both the mRNA and protein levels. Rosiglitazone was poorly effective on these parameters even at 10 μM. Inhibitory effects of 10 μM 15d-PGJ2 were neither reduced by PPARγ blockade with GW-9662 nor enhanced by PPARγ overexpression, supporting a PPARγ-independent mechanism. EMSA and TransAM? analyses demonstrated that mutated IκBα almost completely suppressed the stimulating effect of IL-1β on mPGES-1 expression and PGE2 production, whereas 15d-PGJ2 inhibited NF-κB transactivation. These data demonstrate the following in IL-1-stimulated rat chondrocytes: first, mPGES-1 is rate limiting for PGE2 synthesis; second, activation of the prostaglandin cascade requires NF-κB activation; third, 15d-PGJ2 strongly inhibits the synthesis of prostaglandins, in contrast with rosiglitazone; fourth, inhibition by 15d-PGJ2 occurs independently of PPARγ through inhibition of the NF-κB pathway; fifth, mPGES-1 is the main target of 15d-PGJ2.  相似文献

Records were collected in an experimental herd over an 11-year period from purebred Charolais heifers (n = 351), cows (n = 615) and young entire bulls (n = 383). The objective of the study was to estimate the genetic relationship between the components of female ovarian activity (age at puberty and postpartum anoestrus length), their growth rate and body condition score and beef traits measured on related bulls. Two methods were used to estimate age at puberty and postpartum anoestrus length: the detection of oestrous behaviour and a test of cyclicity based on plasmatic progesterone assay. This study shows the existence of significant heritability estimates for the different cyclicity traits (h2 between 0.11 and 0.38). Most of the genetic correlation coefficients between ovarian activity and growth rate of females and males are negative and favourable (rg between -0.43 and 0.06). Cyclicity is also favourably related with body condition score in young or adult females (rg between -0.65 and -0.22). The genetic relationship between female ovarian activity and proportion of adipose tissue in the male carcass is, however, close to zero. These results show that an antagonism between male beef traits measured in this study and female ovarian activity is unlikely to be a cause for concern in the short term.  相似文献

A new protocol was developed to produce dense organized collagen matrices hierarchically ordered on a large scale. It consists of a two stage process: (1) the organization of a collagen solution and (2) the stabilization of the organizations by a sol-gel transition that leads to the formation of collagen fibrils. This new protocol relies on the continuous injection of an acid-soluble collagen solution into glass microchambers. It leads to extended concentration gradients of collagen, ranging from 5 to 1000 mg/ml. The self-organization of collagen solutions into a wide array of spatial organizations was investigated. The final matrices obtained by this procedure varied in concentration, structure and density. Changes in the liquid state of the samples were followed by polarized light microscopy, and the final stabilized gel states obtained after fibrillogenesis were analyzed by both light and electron microscopy. Typical organizations extended homogeneously by up to three centimetres in one direction and several hundreds of micrometers in other directions. Fibrillogenesis of collagen solutions of high and low concentrations led to fibrils spatially arranged as has been described in bone and derm, respectively. Moreover, a relationship was revealed between the collagen concentration and the aggregation of and rotational angles between lateral fibrils. These results constitute a strong base from which to further develop highly enriched collagen matrices that could lead to substitutes that mimic connective tissues. The matrices thus obtained may also be good candidates for the study of the three-dimensional migration of cells.  相似文献
