We provide an overview of the physical oceanographic and geological processes that affect marine biological habitats and production in the marine waters throughout the archipelago and continental shelf of Southeast Alaska. Given the paucity of regional data, our overview summarizes work done in adjacent regions of the Gulf of Alaska shelf and basin, and draws on research carried out in similar settings elsewhere. The geological setting, which critically influences the regional meteorology and oceanography, includes a narrow continental shelf, deep channels that permeate the archipelago, fjords, glaciers and a rugged, mountainous coast. The large-scale meteorology is influenced primarily by seasonal variations in the intensity and position of the Aleutian Low. Winds, freshwater runoff, tides and cross-shelf exchange control the regional oceanography. The large-scale flow field advects mass, heat, salt, nutrients and planktonic organisms northward from British Columbia (and even further south) to the northern Gulf of Alaska along the slope, shelf, and within the channels of Southeast Alaska. The deep channels permeating the island archipelago and narrow continental shelf facilitate communication between basin and interior waters. Water properties and flow fields are subject to large annual variations in response to similarly large variations in winds and coastal freshwater discharge. The complex geological setting leads to large spatial heterogeneity in the physical processes controlling the local circulation fields and mixing, thereby creating numerous and diverse marine biological habitats. These various circulation and mixing processes modify substantially Southeast Alaska water masses and thus influence marine ecosystem processes downstream over the northern and western Gulf of Alaska shelf.  相似文献

The serine/threonine protein phosphatases are targeted to specific subcellular locations and substrates in part via interactions with a wide variety of regulatory proteins. Understanding these interactions is thus critical to understanding phosphatase function. Using an iterative affinity purification/mass spectrometry approach, we generated a high density interaction map surrounding the protein phosphatase 2A catalytic subunit. This approach recapitulated the assembly of the PP2A catalytic subunit into many different trimeric complexes but also revealed several new protein-protein interactions. Here we define a novel large multiprotein assembly, referred to as the striatin-interacting phosphatase and kinase (STRIPAK) complex. STRIPAK contains the PP2A catalytic (PP2Ac) and scaffolding (PP2A A) subunits, the striatins (PP2A regulatory B' subunits), the striatin-associated protein Mob3, the novel proteins STRIP1 and STRIP2 (formerly FAM40A and FAM40B), the cerebral cavernous malformation 3 (CCM3) protein, and members of the germinal center kinase III family of Ste20 kinases. Although the function of the CCM3 protein is unknown, the CCM3 gene is mutated in familial cerebral cavernous malformations, a condition associated with seizures and strokes. Our proteomics survey indicates that a large portion of the CCM3 protein resides within the STRIPAK complex, opening the way for further studies of CCM3 biology. The STRIPAK assembly establishes mutually exclusive interactions with either the CTTNBP2 proteins (which interact with the cytoskeletal protein cortactin) or a second subcomplex consisting of the sarcolemmal membrane-associated protein (SLMAP) and the related coiled-coil proteins suppressor of IKKepsilon (SIKE) and FGFR1OP2. We have thus identified several novel PP2A-containing protein complexes, including a large assembly linking kinases and phosphatases to a gene mutated in human disease.  相似文献

The purpose of this study was to evaluate the effect of embryonic incubation temperature on the morphology, energetic content, and individual weights of C. opilio zoeae. Ovigerous female C. opilio were collected by trawl from the eastern Bering Sea in the summer of 2002 and were held in the laboratory at five temperature treatments (− 1, 0, 1, 3, and 6 °C) from embryo stage five to hatching. The mean duration of incubation ranged from 240 d at 6 °C to 353 d at − 1 °C. No relationship between larval weight and caloric content with incubation temperature was detected, indicating that increased development time at cooler temperatures may not have an energetic cost. However, several morphological features varied with temperature. Most larval characters including rostro-dorsal length, rostral spine length, lateral spine length, and carapace width were smaller at the warmest incubation temperature. Protopodite length did not differ with temperature. The length of the 3rd abdominal somite consistently increased with increased temperature and may serve as a suitable indicator that could be used in field populations to infer temperature of incubation. Variation in morphology with temperature could confound identification of C. opilio zoeae from those of a congener, Chionoecetes bairdi because identification of C. bairdi is based on the length of the 3rd abdominal somite.  相似文献

wings apart (wap) is a recessive, semilethal gene located on the X chromosome in Drosophila melanogaster, which is required for normal wing-vein patterning. We show that the wap mutation also results in loss of the adult jump muscle. We use complementation mapping and gene-specific RNA interference to localize the wap locus to the proximal X chromosome. We identify the annotated gene CG14614 as the gene affected by the wap mutation, since one wap allele contains a non-sense mutation in CG14614, and a genomic fragment containing only CG14614 rescues the jump-muscle phenotypes of two wap mutant alleles. The wap gene lies centromere-proximal to touch-insensitive larva B and centromere-distal to CG14619, which is tentatively assigned as the gene affected in introverted mutants. In mutant wap animals, founder cell precursors for the jump muscle are specified early in development, but are later lost. Through tissue-specific knockdowns, we demonstrate that wap function is required in both the musculature and the nervous system for normal jump-muscle formation. wap/CG14614 is homologous to vertebrate wdr68, DDB1 and CUL4 associated factor 7, which also are expressed in neuromuscular tissues. Thus, our findings provide insight into mechanisms of neuromuscular development in higher animals and facilitate the understanding of neuromuscular diseases that may result from mis-expression of muscle-specific or neuron-specific genes.  相似文献

Although esterification of free cholesterol to cholesteryl ester in the liver is known to be catalyzed by the enzyme acyl-coenzyme A:cholesterol acyltransferase, ACAT, the neutral cholesteryl ester hydrolase (nCEH) that catalyzes the reverse reaction has remained elusive. Because cholesterol undergoes continuous cycling between free and esterified forms, the steady-state concentrations in the liver of the two species and their metabolic availability for pathways, such as lipoprotein assembly and bile acid synthesis, depend upon nCEH activity. On the basis of the general characteristics of the family of rat carboxylesterases, we hypothesized that one member, ES-4, was a promising candidate as a hepatic nCEH. Using under- and overexpression approaches, we provide multiple lines of evidence that establish ES-4 as a bona fide endogenous nCEH that can account for the majority of cholesteryl ester hydrolysis in transformed rat hepatic cells and primary rat hepatocytes.  相似文献

Plasma membranes of the somatodendritic and axonal domains of neurons are known to have different protein compositions, but the molecular mechanisms that determine this polarized protein distribution remain poorly understood. Herein we show that somatodendritic sorting of various transmembrane receptors in rat hippocampal neurons is mediated by recognition of signals within the cytosolic domains of the proteins by the μ1A subunit of?the adaptor protein-1 (AP-1) complex. This complex, in conjunction with clathrin, functions in the neuronal soma to exclude somatodendritic proteins from axonal transport carriers. Perturbation of this process affects dendritic spine morphology and decreases the number of synapses. These findings highlight the primary recognition event that underlies somatodendritic sorting and contribute to the evolving view of AP-1 as a global regulator of cell polarity.  相似文献

A series of nortropane analogs based on previously reported compound 1 have been synthesized and shown to bind to the nociceptin receptor with high affinity. The synthesis and structure–activity relationships around the C-3 nortropane substitution are described. From the SAR study and hPXR screening effort, compound 15 was identified to possess potent oral antitussive and anxiolytic-like activities in the guinea pig models.  相似文献

Phosphorylation of histone H2AX on Ser 139 (γH2AX) is one of the earliest events in the response to DNA double-strand breaks; however, the subsequent removal of γH2AX from chromatin is less understood, despite being a process tightly coordinated with DNA repair. Previous studies in yeast have identified the Pph3 phosphatase (the PP4C orthologue) as important for the dephosphorylation of γH2AX. By contrast, work in human cells attributed this activity to PP2A. Here, we report that PP4 contributes to the dephosphorylation of γH2AX, both at the sites of DNA damage and in undamaged chromatin in human cells, independently of a role in DNA repair. Furthermore, depletion of PP4C results in a prolonged checkpoint arrest, most likely owing to the persistence of mediator of DNA damage checkpoint 1 (MDC1) at the sites of DNA lesions. Taken together, these results indicate that PP4 is an evolutionarily conserved γH2AX phosphatase.  相似文献

The existence of cavernicolous sculpin (here allocated to Cottus carolinae, banded sculpin, and referred to as grotto sculpin), in the karst regions of Perry County, Missouri, first came to our attention in 1991. Examination of 35 caves in Missouri, 96 in Illinois, 17 in Tennessee, two in Indiana, and 11 in Arkansas revealed that banded sculpin are common in cave habitats; however, grotto sculpin are limited to two karst areas of Perry County, Missouri, where they are known from only six cave systems. These caves and their streams are extensive and apparently provide a unique habitat compared to other karst systems; this may be a critical factor in the present restricted distribution of the grotto sculpin. Grotto sculpin occupy pools and riffles of cave streams, and occur over a variety of substrates, from sediment to breakdown. Density estimates in Mystery and Running Bull caves were 0.29 and 0.63 individuals m-2, respectively. Grotto sculpin have small eyes (1–6% SL vs. 6–10% SL in epigean samples), significantly reduced pigmentation (including nearly complete loss of dorsal saddles), a reduction in pelvic fin ray number (from 4+4 elements to often 4+3 , or 3+3), and enlarged cephalic lateralis pores (e.g., mandibular pores of cavernicolous samples are 2–3 times those of epigean stream samples). Multivariate analyses of body shape revealed statistically significant separation of epigean and hypogean samples, with eye size highly variable, but smallest in the Running Bull Cave population. We interpret these results as representative of losses associated with long-term cave habitation. Caves of Perry County provide ample habitat for grotto sculpin, but because the caves are located downgradient of the city of Perryville and an intensively farmed landscape, point and non-point source pollution threaten their continued existence. Escape of farm-pond fishes through the extensive sinkhole network in Perry County has increased potential predation pressure on grotto sculpin by channel catfish, Ictalurus punctatus, and other species normally excluded from cave environments.  相似文献

Background

Parasitic infections, which are among the most common infections worldwide, disproportionately affect children; however, little is known about the impact of parasitic disease on growth in very early childhood. Our objective was to document the prevalence of parasitic infections and examine their association with growth during the first three years of life among children in coastal Kenya.

Methodology/Principal Findings

Children enrolled in a maternal-child cohort were tested for soil transmitted helminths (STHs: Ascaris, Trichuris, hookworm, Strongyloides), protozoa (malaria, Entamoeba histolytica and Giardia lamblia), filaria, and Schistosoma infection every six months from birth until age three years. Anthropometrics were measured at each visit. We used generalized estimating equation (GEE) models to examine the relationship between parasitic infections experienced in the first three years of life and growth outcomes (weight, length and head circumference). Of 545 children, STHs were the most common infection with 106 infections (19%) by age three years. Malaria followed in period prevalence with 68 infections (12%) by three years of age. Filaria and Schistosoma infection occurred in 26 (4.8%) and 16 (2.9%) children, respectively. Seven percent were infected with multiple parasites by three years of age. Each infection type (when all STHs were combined) was documented by six months of age. Decreases in growth of weight, length and head circumference during the first 36 months of life were associated with hookworm, Ascaris, E. histolytica, malaria and Schistosoma infection. In a subset analysis of 180 children who followed up at every visit through 24 months, infection with any parasite was associated with decelerations in weight, length and head circumference growth velocity. Multiple infections were associated with greater impairment of linear growth.

Conclusions/Significance

Our results demonstrate an under-recognized burden of parasitism in the first three years of childhood in rural Kenya. Parasitic infection and polyparasitism were common, and were associated with a range of significant growth impairment in terms of weight, length and/or head circumference.  相似文献
