Inbred corn varieties differing in their tolerance to the maize dwarf mosaic virus infection also differ in varietal character and host response to simazine. Oh 7B, a tolerant cv., has less total soluble and globulin proteins than the sensitive variety, H 55, but more Kjeldahl nitrogen, albumin protein and a doubling of catalase activity. Virus inoculation in Oh 7B causes a reduction in all three protein fractions, Kjeldahl nitrogen and catalase activity. Deleterious virus-simazine interaction does not occur with either variety and the observed varietal and host response differences in protein may account for the conflicting reports of other workers. 相似文献

Litigation for personal injury following accidental trauma is an expensive and confused process involving three protagonists: patient, doctor and lawyer. Although post-traumatic conditions can be elaborately classified, the intrinsic validity of such classifications is often questionable. Current methods of evaluating psychological sequelae of accidental injury are inaccurate and unsatisfactory, partly because of the protagonists'' conceptual, motivational and semantic differences. In addition, there is no really satisfactory method of (a) determining and quantifying minor but significant degrees of brain damage, (b) distinguishing these from “post-traumatic neurosis”, or (c) determining the relationship between the trauma and subsequent disturbance of function. Increasingly “expert” advice is solicited but owing to the nature of the data and conditions of examination, such advice does little to clarify the underlying problems. Furthermore, doctors are often unable to communicate effectively to the judiciary just how the trauma has affected the patient. The calculation of rates of entry of material into an open system of multiple pools in the steady state from the specific activities of end products, which may be derived from several pools, is described. This analysis may be applied to estimate the rates of secretion of steroid hormones from the specific activities of urinary metabolites which may have various hormones as common precursors. In a previous publication (Gurpideet al., 1963) formulae have been presented by which secretory rates could be calculated after a single injection of the tracers assuming that each of the urinary metabolites was uniquely derived from one of the pools in the system. In the present article similar formulae were derived without this assumption. Consequently, it is shown that, under certain circumstances, non-uniquely derived metabolites can be used to estimate secretory rates, and that it may be unnecessary to consider the pathways of conversion of the hormones to the metabolites or the sites where these conversion occur. 相似文献

Regulation of collagenase and collagenase mRNA production in early- and late-passage human diploid fibroblasts 总被引:6,自引:0,他引:6
The levels of collagenase and collagenase mRNA produced by early-passage (less than 40% of lifespan completed) and late-passage (greater than 80% of lifespan completed) cultures of human fibroblasts were analyzed. The constitutive levels of collagenase and collagenase mRNA produced by the late-passage cultures were 10-30 x greater than the levels observed in similarly treated early-passage cultures. Immunofluorescence analysis established that the percentage of collagenase-positive cells was also greater (77% vs. 4%) in the late-passage cultures. To determine whether the difference in collagenase production resulted from cell-derived regulatory factors, collagenase production was examined in cultures plated onto substrates coated with fibroblast extracellular matrix (ECM). Collagenase and collagenase mRNA production was enhanced in both types of cultures, although amounts produced by ECM-induced early-passage cultures was significantly less than that produced by similarly treated late-passage cultures. In Drosophila, the specific morphological characteristics of each segment are determined by the homeotic genes that regulate the expression of downstream target genes. We used a subtractive hybridization procedure to isolate activated target genes of the homeotic gene Ultrabithorax (Ubx). In addition, we constructed a set of mutant genotypes that measures the regulatory contribution of individual homeotic genes to a complex target gene expression pattern. Using these mutants, we demonstrate that homeotic genes can regulate target gene expression at the start of gastrulation, suggesting a previously unknown role for the homeotic genes at this early stage. We also show that, in abdominal segments, the levels of expression for two target genes increase in response to high levels of Ubx, demonstrating that the normal down-regulation of Ubx in these segments is functional. Finally, the DNA sequence of cDNAs for one of these genes predicts a protein that is similar to a human proto-oncogene involved in acute myeloid leukemias. These results illustrate potentially general rules about the homeotic control of target gene expression and suggest that subtractive hybridization can be used to isolate interesting homeotic target genes. 相似文献

HLA antigens have been purified to homogeneity after detergent solubilization from RPMI 4265, a human lymphoblastoid line. The inhibition of cytotoxicity assay for HLA antigen was modified, using preincubation with bovine serum albumin of antigen samples containing detergent to prevent lysis of target cells by detergent. Solubilization was tested with many types of detergents. A polyethyleneglycol oleyl ether nonionic detergent mixture, Brij 99:Brij 97 (2:1) was selected for solubilization, since it selectively solubilized HLA antigens, had a low absorbance at 280 nm and was uncharded. HLA antigens were then purified by Lens culinaris lectin affinity chromatography and Bio-Gel A-5m filtration. The antigen specifity HLA-A2 was separated from specificities HLA-B7,12 by isoelectric focusing. Purified HLA antigens contained a subunit of Mr=44,000 with NH2-terminal glycine, and a subunit of Mr=12,000, beta2-microglobulin, with NH2-terminal isoleucine. 相似文献

A simplified mathematical model of the origin of the left-handed asymmetry of proteins in living matter is presented. The model is based on the hypothesis of Vester and Ulbricht that the chirality of (lefthanded) electrons from naturally beta-active elements, e.g.,14C,40K, etc., was the specific source of the asymmetry; it requires for its application data on the interaction of electrons having non-zero chirality with racemic mixtures of amino acids. This interaction is here treated theoretically in an order-of-magnitude calculation. Our analysis yields a very approximate value of the induced steady-state asymmetry in the amino acids at the beginning of protein synthesis and indicates that this asymmetry, though small, may have been suffcient to account for the dominant left-handedness of proteins now observed. 相似文献
