Short courses of nitrofurantoin and ampicillin produced an immediate cure in 80% of adult non-pregnant bacteriuric women. Of the subjects so treated, 55% remained cured at the end of one year. Over the same follow-up period 36% of untreated bacteriuric women developed a spontaneous remission of bacteriuria. Treatment failed to prevent the development of symptomatic infection, and the reinfections which followed successful treatment were more commonly associated with the development of symptoms than the persistent or relapsing infections in untreated or unsuccessfully treated subjects.It is concluded that a search for bacteriuria in non-pregnant women is unlikely to be of value as a preventive measure, since in many instances it fails to detect urinary tract infection at an early stage and since treatment by methods suitable for large-scale use is ineffective.  相似文献

Differences in the metabolic fate of antithyroid drugs influence the optimal frequency of administration and their therapeutic efficacy. 35S propylthiouracil differed from the 35S imidazoles (carbimazole and methimazole) in the more rapid absorption and excretion and the shorter biological half-life in the plasma of the former. Renal function may have a more important influence on the biological half-life of the drugs than thyroid status. Further work is required to determine the optimal frequency of administration for each compound.  相似文献

West Indian spiny lobsters, Panulirus argus, synthesized a hemolymph bactericidin after being injected with killed suspensions of gram-negative bacillus EMB-1 isolated from the normal gut of this lobster. To study differences between the primary response and secondary response, animals were given a primary antigen injection of EMB-1 followed by a second injection of the same antigen 22 to 51 days later. As a rule, secondary bactericidal responses were enhanced over the primary in a manner reminiscent of specific anamnesis in mammalian immunoglobulin synthesis. Immunological memory was also suggested when tertiary responses were compared to secondary and by the persistence of residual titers for many days or weeks without additional antigenic stimulation.  相似文献

This investigation revealed that the ribonucleotide reductases in extracts of Rhizobium leguminosarum, R. trifolii, R. phaseoli, R. japonicum, and R. meliloti 3DOal (ineffective in nitrogen fixation) are dependent upon B(12) coenzyme for activity. Rhizobium and certain Lactobacillus species are the only two groups of organisms known to contain B(12) coenzyme-dependent ribonucleotide reductases. Extracts of cobalt-deficient R. meliloti cells assayed in the presence of optimum B(12) coenzyme showed a 5- to 10-fold greater ribonucleotide reductase activity than comparable extracts from cells grown on a complete medium. Furthermore, cobalt-deficient cells were abnormally elongated and contained reduced contents of deoxyribonucleic acid. The addition of purified deoxyribonucleosides to cobalt-deficient cultures of R. meliloti failed to alleviate deficiency symptoms.  相似文献

1. There is a well-recognized genetic polymorphism for the N-acetylation of isoniazid and sulphamethazine by human livers. 2. Serotonin was found to be acetylated by human liver enzyme preparations and the N-acetylserotonin formed was identified and determined quantitatively. 3. In 13 livers examined there was a wide variability in the capacity to acetylate serotonin that did not correlate with the capacity of the same livers to acetylate isoniazid and sulphamethazine. The results suggest that serotonin is not a natural substrate for the polymorphic N-acetyltransferase and that it may be acetylated by a different enzyme.  相似文献

Membrane ghost preparations of Escherichia coli K-12 obtained by osmotic lysis of lysozyme-induced spheroplasts were found to possess both Mg(++)- and Ca(++)-activated adenosine 5'-triphosphatase (ATPase, EC 3.6.1.3) activities. Maximal activities of 1.0 to 1.5 mumoles of orthophosphate released per min per mg of protein were obtained at pH 9.0 with a molar Mg(++) to adenosine 5'triphosphate (ATP) ratio of 2:5 and at pH 9.9 with a molar Ca(++) to ATP ratio of 1:5. These ATPase activities were not altered by ouabain, fluoride, N-ethylmaleimide, 2,4-dinitrophenol, cyanide, or dithionite, but were inhibited by low concentrations of azide, p-chloromercuribenzoate, and pentachlorophenol. Mg(++) ATPase was more susceptible to inhibition by azide than was Ca(++) ATPase. Fifty per cent inactivation of both activities was observed when membrane ghost preparations were preincubated at 66 C for 10 min. The Mg(++) and Ca(++) ATPase activities of these preparations were not additive, but did respond independently to inhibition by monovalent cations. Ca(++) ATPase was found to be very sensitive to inhibition by K(+), Na(+), Li(+), Rb(+), and Cs(+); Mg(++) ATPase was relatively insensitive to these ions. One possible interpretation of the results presented in this paper is that the membrane of E. coli possesses an ATPase which is activated by either Mg(++) or Ca(++) and that activation by Ca(++) increases the susceptibility of this enzyme to inhibition by monovalent cations. Increased susceptibility of E. coli membrane ATPase to inhibition by monovalent cations such as Na(+) and K(+) as a consequence of Ca(++) activation could represent a regulatory mechanism.  相似文献
