We investigated the role of fire and other potential biotic and abiotic determinants for the occurrence of an obligate seeder
and active pyrophyte, Cistus salviifolius, a vulnerable species in the Swiss Red List. Populations of this species are threatened by the change of fire regime that
has occurred during the last ten years. Fires and burnt areas are today less frequent and it is not clear whether other factors
can compensate for such fire-free periods should this trend continue. We used a stratified random design to collect data on
the cover of three growth stages, i.e., juvenile, chamaephytic and nanophanerophytic (oldest), of C. salviifolius, on plant communities, and on environmental factors. A tree-based discriminant analysis showed that the time elapsed since
last fire was the best predictor of Cistus occurrence, but plants could survive on rocky and sunny sites if no fire had occurred for more than 32 years. Contrary to
our expectations, the number of fires and time elapsed since the last event was correlated with the oldest stages only, and
not with that of seedlings or juveniles. Rank correlations showed that bare soil was a good predictor of young stages, whereas
rocky outcrops and shallow soils were important for the chamaephytic stage. Our results confirm the role of disturbances and
competition-free habitats as determinants of the survival of this vulnerable pyrophyte during long periods without fires and
the existence of ontogenetic niche segregation of this species. In order to maintain viable C. salviifolius populations in the Alps, managers should undertake conservation actions according to the requirements of the different growth

Peroxisome proliferator-activated receptor-gamma (PPAR-gamma) plays a central role in whole body metabolism by regulating adipocyte differentiation and energy storage. Recently, however, PPAR-gamma has also been demonstrated to affect proliferation, differentiation, and apoptosis of different cell types. As we have previously shown that BAY 11-7085-induced synovial fibroblast apoptosis is prevented by PPAR-gamma agonist 15d-PGJ2; the expression of PPAR-gamma in these cells was studied. Both PPAR-gamma1 and PPAR-gamma2 isoforms were cloned from synovial fibroblast RNA, but only PPAR-gamma1 was detected by Western blot, showing constitutive nuclear expression. Within minutes of BAY 11-7085 treatment, a PPAR-gamma1-specific band was shifted into a form of higher mobility, suggesting dephosphorylation, as confirmed by phosphatase treatment of cell extracts. Of interest, BAY 11-7085-induced PPAR-gamma1 dephosphorylation was followed by PARP and caspase-8 cleavage as well as by PPAR-gamma1 protein degradation. PPAR-gamma1 dephosphorylation was followed by the loss of PPAR-DNA binding activity ubiquitously present in synovial fibroblast nuclear extracts. Unlike the phosphorylated form, dephosphorylated PPAR-gamma1 was found in insoluble membrane cell fraction and was not ubiquitinated before degradation. PPAR-gamma1 dephosphorylation coincided with ERK1/2 phosphorylation that accompanies BAY 11-7085-induced synovial fibroblasts apoptosis. 15d-PGJ2, PGD2, and partially UO126, down-regulated ERK1/2 phosphorylation, protected cells from BAY 11-7085-induced apoptosis, and reversed both PPAR-gamma dephosphorylation and degradation. Furthermore, PPAR-gamma antagonist BADGE induced PPAR-gamma1 degradation, ERK1/2 phosphorylation, and synovial fibroblasts apoptosis. The results presented suggest an anti-apoptotic role for PPAR-gamma1 in synovial fibroblasts. Since apoptotic marker PARP is cleaved after PPAR-gamma1 dephosphorylation but before PPAR-gamma1 degradation, dephosphorylation event might be enough to mediate BAY 11-7085-induced apoptosis in synovial fibroblasts. 相似文献

The degree and the origins of quantitative variability of most human plasma proteins are largely unknown. Because the twin study design provides a natural opportunity to estimate the relative contribution of heritability and environment to different traits in human population, we applied here the highly accurate and reproducible SWATH mass spectrometry technique to quantify 1,904 peptides defining 342 unique plasma proteins in 232 plasma samples collected longitudinally from pairs of monozygotic and dizygotic twins at intervals of 2–7 years, and proportioned the observed total quantitative variability to its root causes, genes, and environmental and longitudinal factors. The data indicate that different proteins show vastly different patterns of abundance variability among humans and that genetic control and longitudinal variation affect protein levels and biological processes to different degrees. The data further strongly suggest that the plasma concentrations of clinical biomarkers need to be calibrated against genetic and temporal factors. Moreover, we identified 13 cis‐SNPs significantly influencing the level of specific plasma proteins. These results therefore have immediate implications for the effective design of blood‐based biomarker studies. 相似文献

Résumé A la suite de recherches antérieures intéressant la vascularisation de la trompe utérine et celle de l'ovaire, la présente étude concerne la microvascularisation de la muqueuse vaginale chez la Femme et se fonde sur l'examen de dix vagins humains prélevés chez des sujets en période d'activité génitale, vagins dont le système vasculaire est injecté à l'encre de Chine gélosée.L'étude de l'angioarchitectonie de la muqueuse vaginale en général laisse apparaître un dispositif vasculaire spécifique de cette dernière et représenté au niveau du chorion par des artérioles superficielles qui s'arborisent en de multiples branches se résolvant en un réseau capillaire sous-muqueux; ce dernier est constitué d'une part, d'un dense réseau capillaire à mailles ovalaires ou polygonales, étalé sous la couche basale de l'épithélium malpighien, d'autre part, d'anses capillaires intra-papillaires, sensiblement perpendiculaires à l'épithélium malpighien et se répétant à intervalles réguliers. The angioarchitectony of the submucous network of the human vagina and its variations during the ovarian cycle
Summary The microvascularization of the vaginal mucosa has been investigated on 10 human vaginae taken from autopsies of women having been in their sexual active period, the vascular system of the vaginae being injected with solutions of China ink and gelose.The angioarchitectony of the vaginal mucosa shows a specific vascular pattern. The latter is represented at the level of the stroma by superficial arterioles which divide into multiple branches giving rise to the submucous capillary network. This network consists on one hand of a thick capillary plexus with ovale or polygonal stitches, spread under the basal layer of the malpighian epithelium, and on the other hand of intrapapillary capillary creeks arranged in regular intervals and perpendicularly oriented to the malpighian epithelium. This capillary network is drained by a wide spread venous plexus connected with big veins of irregular calibre.The variations of this angioarchitectony are especially pronounced during the second part of the menstrual period, when the calibre of the arterioles as well as that of the capillaries creeks grow in height and become spiralized.相似文献

A mini-compression jig was built to perform in situ tests on bovine trabecular bone monitored by micro-MRI. The MRI antenna provided an isotropic resolution of 78 μm that allows for a volume correlation method to be used. Three-dimensional displacement fields are then evaluated within the bone sample during the compression test. The performances of the correlation method are evaluated and discussed to validate the technique on trabecular bone. By considering correlation residuals and estimates of acquisition noise, the measured results are shown to be trustworthy. By analyzing average strain levels for different interrogation volumes along the loading direction, it is shown that the sample size is less than that of a representative volume element. This study shows the feasibility of the 3D-displacement and strain field analyses from micro-MRI images. Other biological tissues could be considered in future work. 相似文献

L'augmentation du volume irradié diminue la rapidité du premier des deux processus de restauration et la quantité d'endoplasme qui reste altérée s'accroît.Dans la zone irradiée, le courant cytoplasmique est suspendu; il ne reprend que 4 minutes après le traitement alors que la couche chloroplastique se réorganise. Le glissement des inclusions qu'il charrie se fait un certain temps par saccades. La vitesse de la fraction endoplasmique protégée y augmente régulièrement témoignant ainsi du rétablissement de la force motrice. Le rétablissement a lieu suivant un schéma analogue à celui de Pendoplasme irradié sauf que ce phénomène y est plus rapide et que la force motrice du moins pendant les premières heures se rétablit intégralement. Summary In these experiments onNitella internodal cells, only a fraction of the cellular volume is irradiated (by 500 kR). The restoring of the endoplasm (by measurement of the cyclosis in the shielded zone) and the restoring of the ectoplasm where the motive force is generated (by measurement of the cyclosis in the irradiated zone) can be examinated separately during the recovery of the cytoplasmic streaming.In the shielded zone, the plot on a semi-logarithmic scale of a typical rate of streaming-time curve shows that the speed of the irradiated endoplasm. increases again according to a second order equation. With increasing irradiated cellular volume, there are a gradual slowing down of the first processus of restauration and an increase of the definitively altered fraction of the endoplasm.In the irradiated zone, the cytoplasmic streaming stopped completely. About 4 minutes after the treatment, when the chloroplasts layer is reorganising, it exhibits a tendency to flow again. The carried inclusions slide then in jerks during several minutes. The rate of the protected fraction of the endoplasm increases regularly, proving the recovery of the motive force. An analysis of this processus versus time shows that it can be exprimed by a second order equation as that of the irradiated endoplasm. In Arabidopsis, ETO1 (ETHYLENE-OVERPRODUCER1) is a negative regulator of ethylene evolution by interacting with AtACS5, an isoform of the rate-limiting enzyme, 1-aminocyclopropane-1-carboxylate synthases (ACC synthase or ACS), in ethylene biosynthetic pathway. ETO1 directly inhibits the enzymatic activity of AtACS5. In addition, a specific interaction between ETO1 and AtCUL3, a constituent of a new type of E3 ubiquitin ligase complex, suggests the molecular mechanism in promoting AtACS5 degradation by the proteasome-dependent pathway. We have used a paleogenetics approach to investigate the genetic landscape of coat color variation in ancient Eurasian dog and wolf populations. We amplified DNA fragments of two genes controlling coat color, Mc1r (Melanocortin 1 Receptor) and CBD103 (canine-β-defensin), in respectively 15 and 19 ancient canids (dogs and wolf morphotypes) from 14 different archeological sites, throughout Asia and Europe spanning from ca. 12 000 B.P. (end of Upper Palaeolithic) to ca. 4000 B.P. (Bronze Age). We provide evidence of a new variant (R301C) of the Melanocortin 1 receptor (Mc1r) and highlight the presence of the beta-defensin melanistic mutation (CDB103-K locus) on ancient DNA from dog-and wolf-morphotype specimens. We show that the dominant KB allele (CBD103), which causes melanism, and R301C (Mc1r), the variant that may cause light hair color, are present as early as the beginning of the Holocene, over 10 000 years ago. These results underline the genetic diversity of prehistoric dogs. This diversity may have partly stemmed not only from the wolf gene pool captured by domestication but also from mutations very likely linked to the relaxation of natural selection pressure occurring in-line with this process. 相似文献

Invasion success is favoured by the introduction of pre-adapted genotypes. In addition, novel pressures in the introduced
range may lead to phenotypic changes related to fitness or competitive ability of introduced plants. Polyploidy appears to
be over-represented in invasive plants, but differences between cytotypes in growth strategies including trade-offs among
plant traits have received little attention so far in the context of biological invasions. We grew Centaurea stoebe L. and Senecio inaequidens D.C. in a greenhouse experiment to test for differences in fitness (shoot biomass, reproductive output) and competitive ability
(vegetative size, specific leaf area, leaf dry matter content, root–shoot ratio) between diploid and polyploid cytotypes as
well as between native and introduced plants. For both species, diploid and tetraploid genotypes occur in the native range,
whereas only tetraploids are present in the introduced range. In the native range of both species, diploid and tetraploid
genotypes had different growth strategies. Tetraploid genotypes of C. stoebe and S. inaequidens had, respectively, higher specific leaf area and stem height than diploid ones. Thus, for both species, native tetraploids
appeared more competitive than native diploids, which could explain, at least partially, the invasion success of the pre-adapted
tetraploid genotypes. The comparison of native and introduced tetraploid genotypes revealed differences in traits linked to
competitive ability, which could be linked to novel selection in the new environment. In S. inaequidens, we found evidence for a competition-colonisation trade-off, whereas persistence of C. stoebe in the new range seemed to be linked to a competition-defence trade-off. 相似文献

Conditions for the maintenance of males in androdioecious populations (populations with both male and hermaphrodite individuals) have been derived for four different one-locus two-allele models of inheritance of androdioecy. The results are not in general accordance with those already known: depending on the mode of inheritance, males can be maintained irrespective of their fertilities. If males are sufficiently fertile, it may happen that they are maintained only for intermediate selfing rates of the hermaphrodites. A result already found for gynodioecy is confirmed for androdioecy, namely, that a 11 sex ratio is immediately established among zygotes if hermaphrodites appear as heterozygotes only. 相似文献
