Background and aims

Our aim was to investigate the effect of TNFα on muscle resting potential (RP) and then in muscle excitability and to demonstrate another mechanism implicated in intensive care units (ICU) acquired polyneuromyopathy.

Methods

Experiments were carried out on adult female Wistar rats. After isolation of muscle fibres from peroneus longus, influence of TNFα was tested on RP by using intracellular microelectrodes. Digoxin and chelerythrin were used to determine the mechanism of TNFα action.

Results

First, we found that TNFα induced a concentration dependent increase of muscle RP and that this mechanism, which was blocked by digoxin, was due to an effect on the Na/K ATPase. As it was also blocked by chelerythrin it was concluded that this effect was mediated by PKC activation of the Na/K ATPase.

Conclusions

We demonstrated that TNFα leads to a PKC mediated increase in muscle RP. Depolarization needed to reach the threshold voltage for muscle action potential should then be higher and this could be involved in the decrease in muscle excitability observed in acquired polyneuromyopathy.  相似文献

Eurasia was home to a great radiation of hominoid primates during the Miocene. All were extinct by the end of the Miocene in Western Eurasia. Here, we investigate the hypothesis of climate and vegetation changes at a local scale when the cercopithecoid Mesopithecus replaced the hominoid Ouranopithecus along the Axios River, Greece. Because they are herbivorous and were much more abundant than primates, bovids are preferred to primates to study climate change in the Balkans as a cause of hominoid extinction. By measuring carbon stable isotope ratios of bovid enamel, we conclude that Ouranopithecus and Mesopithecus both evolved in pure C3 environments. However, the large range of δ13C values of apatite carbonate from bovids combined with their molar microwear and mesowear patterns preclude the presence of dense forested landscapes in northern Greece. Instead, these bovids evolved in rather open landscapes with abundant grasses in the herbaceous layer. Coldest monthly estimated temperatures were below 10 °C and warmest monthly temperatures rose close to or above 20 °C for the two time intervals. Oxygen isotope compositions of phosphate from bulk samples did not show significant differences between sites but did show between-species variation within each site. Different factors influence oxygen isotope composition in this context, including water provenience, feeding ecology, body mass, and rate of amelogenesis. We discuss this latter factor in regard to the high intra-tooth variations in δ18Op reflecting important amplitudes of seasonal variations in temperature. These estimations fit with paleobotanical data and differ slightly from estimations based on climate models. This study found no significant change in climate before and after the extinction of Ouranopithecus along the Axios River. However, strong seasonal variations with relatively cold winters were indicated, conditions quite usual for extant monkeys but unusual for great apes distributed today in inter-tropical regions.  相似文献

Abstract

Camels are exceptionally rare in the Plio-Pleistocene fossil record of Africa, hindering attempts to understand the evolution of this family on the continent. Here we describe recently collected camel specimens from the Shungura Formation, Lower Omo Valley, Ethiopia, and attribute these remains to Camelus grattardi. The new specimens date to the late Pliocene (~3 to 2.6 Ma) and consist of three upper molars, one upper premolar, and two proximal metatarsals. The dental specimens confirm this species’ small P4 relative to its molars, a trait that differs significantly from all extant and fossil Old World camels. The metatarsals indicate that C. grattardi was similar in size to the living Bactrian camel, C. bactrianus. Phylogenetically, we find no suitable ancestor, sister, or descendant of the eastern African fossil camel, which suggests greater lineage diversity in Plio-Pleistocene Camelus than previously recognised. Microwear analyses suggest that C. grattardi was likely a mixed-feeder preferring browse, which is consistent with carbon isotopes of enamel from the Turkana Basin. A review of the fossil record of African camels suggests no clear paleoenvironmental association, as fossil camels occur in a range of environments from dry savannas with no permanent water bodies to closed woodlands along the paleo-Omo River.  相似文献

Worldwide, cattle production is struggling to face the negative impacts caused by ticks and Rhipicephalus (Boophilus) microplus is one of the most harmful ticks for livestock. Most of the people in West Africa depend on cattle farming and subsistence agriculture. The presence of ticks on cattle is a major problem faced by smallholder farmers who fight for their livelihood. National and regional tick control programs could assist these rural communities in protecting their livelihoods against ticks and tick-borne diseases, but only if they take into account the targeted herders and their perception on cattle management and tick control. This paper aims to provide a better insight in the socio-economic characteristics of Beninese cattle farmers, and their perception on tick burden, as well as to document common tick control strategies. Different tick species and their seasonality are well understood by cattle herders. For tick control, many still use manual tick removal, especially in the north of the country. The high cost of acaricides, the lack of financial means of African farmers, and of the local stockbreeders in particular, limits the use of acaricides in livestock breeding in Benin. While aiming to increase the meat or milk production of their animals, stockbreeders who can afford it sometimes turn to an abusive use of acaricides, which might in time lead to an increase in tick resistance. This study remains one of the rare studies to report extensively on the perceptions of West African cattle herders.  相似文献

While grazing as a selective factor towards hypsodont dentition on mammals has gained a lot of attention, the importance of fruits and seeds as fallback resources for many browsing ungulates has caught much less attention. Controlled‐food experiments, by reducing the dietary range, allow for a direct quantification of the effect of each type of items separately on enamel abrasion. We present the results of a dental microwear texture analysis on 40 ewes clustered into four different controlled diets: clover alone, and then three diets composed of clover together with either barley, corn, or chestnuts. Among the seed‐eating groups, only the barley one shows higher complexity than the seed‐free group. Canonical discriminant analysis is successful at correctly classifying the majority of clover‐ and seed‐fed ewes. Although this study focuses on diets which all fall within a single dietary category (browse), the groups show variations in dental microwear textures in relation with the presence and the type of seeds. More than a matter of seed size and hardness, a high amount of kernels ingested per day is found to be correlated with high complexity on enamel molar facets. This highlights the high variability of the physical properties of the foods falling under the browsing umbrella.  相似文献

DNA ends get exposed in cells upon either normal or dysfunctional cellular processes or molecular events. Telomeres need to be protected by the shelterin complex to avoid junctions occurring between chromosomes while failing topoisomerases or clustered DNA damage processing may produce double-strand breaks, thus requiring swift repair to avoid cell death. The rigorous study of the great many proteins involved in the maintenance of DNA integrity is a challenging task because of the innumerous unspecific electrostatic and/or hydrophobic DNA—protein interactions that arise due to the chemical nature of DNA. We devised a technique that discriminates the proteins recruited specifically at DNA ends from those that bind to DNA because of a generic affinity for the double helix. Our study shows that the DNA ends proteome comprises proteins of an unexpectedly wide functional spectrum, ranging from DNA repair to ribosome biogenesis and cytoskeleton, including novel proteins of undocumented function. A global mapping of the identified proteome on published DNA repair protein networks demonstrated the excellent specificity and functional coverage of our purification technique. Finally, the native nucleoproteic complexes that assembled specifically onto DNA ends were shown to be endowed with a highly efficient DNA repair activity.  相似文献

In insect parasitoids, mating strategy depends on mate availability and is influenced by the spatial and temporal emergence patterns of adults. For quasi-gregarious species, simultaneous emergence favors local mating and reduces search costs for partners while increasing the risk of inbreeding, particularly when only one female parasitizes the initial host patch. Consequently, in inbreeding sensitive species, mating on the place of adult emergence (patch mating) between siblings should be counter selected. In practice, the timing of male and female emergence and of their dispersal influences mate availability and can limit on patch mating. To test the role of these two factors, we analyzed the daily distribution of emergence and patch residence time of a cohort in the aphid parasitoid Aphidius matricariae (Hymenoptera: Braconidae). We observed that adult emergence is concentrated on the morning with males emerging on average before females with some overlaps. A more precise evaluation of emergence pattern within a brood suggests that brothers and sisters rarely emerge at the same time and rapid dispersal of males and females favors off-patch mating. The relationships between timing of emergence including differences between sex and consequences on inbreeding probability in these species are thus discussed.  相似文献

Thin film solar cells with Al/ITO/ZnO/CdS/CZTSSe/Mo‐glass structure are fabricated employing a fast and low‐cost preparation procedure using an aqueous ink deposited by nonpyrolytic spray, followed by high temperature crystallization and selenization steps. Capacitance–voltage measurements on previously reported devices with >8% efficiency under 1 sun irradiation show a charge carrier density of the order of 1017 cm?3. Moreover, admittance spectroscopy indicates the presence of mid‐bandgap defects that are tentatively attributed to a Sn deficit in the film. In order to reduce the number of these deep defects within the active layer of our solar cells, the Sn content is tuned in the precursor ink. Their morphology, elemental composition, crystal phases, capacitance–voltage profiling, admittance, photoluminescence, and photovoltaic performances are characterized. The results indicate that tuning the Sn content offers a strong leverage upon some key properties of the active layer, in particular the grain size, and the charge carrier and defect density. By employing this leverage to optimize the performance of our CZTSSe layers, the cell performances are increased to 10.0% without antireflection coating (ARC) and to 10.8% (on 0.25 cm2) with an ARC.  相似文献

Dermaseptin S9 (Drs S9), GLRSKIWLWVLLMIWQESNKFKKM, isolated from frog skin, does not resemble any of the cationic and amphipathic antimicrobial peptides identified to date, having a highly hydrophobic core sequence flanked at either side by cationic termini. Previous studies [Lequin O, Ladram A, Chabbert A, Bruston F, Convert O, Vanhoye D, Chassaing G, Nicolas P & Amiche M (2006) Biochemistry45, 468-480] demonstrated that this peptide adopted a non-amphipathic alpha-helical conformation in trifluoroethanol/water mixtures, but was highly aggregated in aqueous solutions and in the presence of sodium dodecyl sulfate micelles. Circular dichroism, FTIR and attenuated total reflectance FTIR spectroscopies, combined with a surface plasmon resonance study, show that Drs S9 forms stable and ordered beta-sheet aggregates in aqueous buffers or when bound to anionic or zwitterionic phospholipid vesicles. These structures slowly assembled into amyloid-like fibrils in aqueous environments via spherical intermediates, as revealed by electron microscopy and Congo red staining. Drs S9 induced the directional migration of neutrophils, T lymphocytes and monocytes. Interestingly, the antimicrobial and chemotactic activities of Drs S9 are modulated by its amyloid-like properties. Whereas spherical oligomers of Drs S9 exhibit antimicrobial activity, the soluble, weakly self-associated forms of Drs S9 act on human leukocytes to promote chemotaxis and/or immunological response activation in the same range of concentration as amyloidogenic peptides Abeta(1-42), the most fibrillogenic isoform of amyloid beta peptides, and the prion peptide PrP(106-126).  相似文献
