n-Alkyltrimethylammonium bromides (CnTAB) have been synthesized with n-alkyl chain lengths of between C5 and C22 . Antimicrobial activity was assessed as growth inhibitory activity (MIC) towards seven representative strains of micro-organisms and as bacterial activity towards Staphylococcus aureus, Saccharomyces cerevisiae and Pseudomonas aeruginosa . These data indicated that whilst the level of activity was generally parabolically related to n-alkyl chain length, separate dependencies existed for short (n < 10) and long (n > 10) chain substituents, with disproportionately large increases in activity, in the order of three log cycles, occurring as substituent chain lengths increased from n = 10 to n = 12. This is interpreted in terms of distinct mechanisms of action, binding site and/or physicochemical properties for extreme members of the series. 相似文献

Synopsis Adult clearnose skates, Raja eglanteria, were captured during the winters of 1981 and 1983, and observed to mate in captivity. Mating and egg depositions take place on the central west coast of Florida from December through mid-May. During copulation the male holds the trailing edge of the female's right or left pectoral fin firmly in his mouth, swings his tail beneath hers and inserts one clasper into the distal end of her reproductive tract. Copulation may last one to four hours during which time sperm pass from the urogenital papilla of the male along the clasper groove to the female. Sperm move cranially to the upper portion of the shell gland where they are stored and remain viable for at least three months. The ovum is fertilized in the shell gland. The egg case bears a prominent projection or horn at each corner. The two posterior ones are shorter and bear tendrils which are covered with a sticky substance that insures attachment to the substrate when the egg is deposited. Fertilized eggs are laid in pairs at intervals ranging from 1 to 13 days (mean of 4.5 ± 2.2 days). As development proceeds within the egg case a plugged slit on the lateral side of each horn opens and permits seawater to wash the developing embryo. Incubation periods for eggs maintained between 20–22°C decrease in duration throughout the egg laying season, ranging from 94 days initially to 77 days for eggs laid later in the spring. At hatching, the anterior end of the egg case ruptures, and the skate emerges abruptly with its pectoral fins rolled dorsally. 相似文献

Summary Peripheral lymphocytes from 16 healthy adults, 9 pregnant women, and 3 fragile X syndrome patients were cultured in Eagle's minimum essential medium without folic acid (MEM-FA). The addition of 2mM, 4mM, or 8mM uridine 24h or 72h prior to harvest resulted in increases of chromosome gaps or breaks, especially at hot points 3p14, 16q23-24, and at fragile site Xq27. Pregnant women showed higher frequencies of 3p14 breaks and total chromosome breaks than men and non-pregnant women. The other chromosome regions, such as 6q26, 7q23, 7q35, 6p25, Xp22, 14q23 and 11p13, also frequently showed gaps or breaks. The results indicated that the unbalance of nucleotide pools was one of the causes of chromosome breakages. The higher frequencies of chromosome gaps and breaks under the condition of thymidylate stress may be due to the misincorporation of uracil instead of thymine into DNA. 相似文献

Cyclic-AMP-binding proteins in membrane and soluble fractions from rat forebrain were compared; membrane fractions included smooth and rough microsomes and a plasma membrane fraction enriched in synaptic membranes. Protein fractions were treated with 8-azido-[32P]cyclic AMP and ultraviolet irradiation to covalently tag cyclic-AMP-binding proteins. Labeled proteins were then analyzed by two-dimensional gel electrophoresis (2DGE) and fluorography. The soluble CNS proteins contained two major cyclic-AMP-binding species at 48K (48K 5.5 and 48K 5.45), differing slightly in their isoelectric points. Another protein was seen at 54K (54K 5.3) adjacent to the beta-tubulin subunits in the 2D electrophoretogram. The analysis of the smooth microsome and plasma membrane fractions differed from the soluble fraction in that there were two cyclic-AMP-binding proteins adjacent to the beta-tubulin region (54K 5.3 and 52K 5.3) differing slightly in apparent molecular weight. The membrane fractions also contained a cyclic-AMP-binding protein at 54K 5.8. The 52K 5.3 and 54K 5.8 species were unique to the membrane fractions. The rough microsomes did not contain detectable amounts of cyclic-AMP-binding proteins. Free polysomes were isolated from brain tissue, and translation products were analyzed by cyclic AMP affinity chromatography and immunopurification with antibodies to the brain specific type II regulatory subunit. The translation products that were found to bind cyclic AMP Sepharose are as follows: 48K 5.5, 48K 5.45, 52K 5.3, and 54K 5.8. 建立了8个分泌抗新城疫病毒(NDV)特异性单克隆抗体(McAb)的杂交瘤细胞株,根据它们的免疫生物学特性可以分为三种类型:(1)具有FA和ELISA特性(FN1、FN4、FN29、FN30、FN35、FNl22);(2)具有FA、ELISA和HI特性(FN7);(3)具有ELISA、HI特性和中和能力(FN106),根据FN30和FN106的ELISA试验,可将11个NDV毒株分为二种不同的抗原群,应用FN4-FITC,FN7-FITC和FN29-HRP试剂,对人工感染NDV和野外送检病例检测结果表明,单抗试剂的DFA阳性率(92.3%)高于病毒分离阳性率(87.2%),两种方法的符合率89.7%,这些单抗试剂用于临床诊断敏感性和特异性高,且方法快速、简便。 相似文献

1985年6—10月于长江口共获得白鲟当年的1龄幼鱼标本49尾(体长80—530毫米),进行了有关形态、生长以及食性的观察研究。①幼鱼形态与成鱼基本相似,其体长与体长,眼径以及体长/吻须长呈明显的正相关;③6月21日至10月12日间的幼鱼长度生长方程为Lg=79.9723e^0.0213t(式中Lg为体长,毫米;t为天数),其相对增长率以8月为最快;④长江口白鲟幼鱼摄食频度为100%,平均胃饱满指数为198.46,它们是肉食性鱼类,以底栖小型虾类和鱼类为主食,食物共有8类16个可鉴别种类;④考虑到这种珍稀鱼类的种群与资源衰退现状,建议在长江河口区制定必要韵保护措施。 相似文献

A new adsorption chromatography procedure for the purification of calmodulin from bovine brain was developed using polymeric adsorbent 3520. Calmodulin was first isolated by DEAE-Cellulose column chromatography and further purified to apparent homogeneity following elution with 50% ethanol from the adsorbent column. Polyacrylamide gel electrophoresis showed one band either in the presence of Ca2+ or EGTA. The polymeric adsorbent 3520 is a non-polar polymer lacking exchangeable groups. The selective adsorption of calmodulin is based on hydrophobic interaction within the matrix, and is Ca2+ independent. Neither high salt (0.5 M NaC1) nor EGTA (5 mM) was able to elute the CaM from the adsorption column whereas ethanol (50%) eluted it completely. This method is simple to use and it provides highly purified calmodulin with high yield. 相似文献

A new species, K. carlsbergiana , is described in what is currently considered a monotypic genus. The new species is only known from permanent streams in the Arssi and Bale regions of S Ethiopia. The delimitation and position of the genus is reconsidered in light of the additional information provided by the new species. The distinctiveness of the genus is reconfirmed whilst no new clues to possible relationships with other genera were observed. The taxonomy of the other species, K. laniflora (Forssk.) R. The release of growth hormone-releasing factor (GHRF) from rat hypothalamus was investigated in vitro. After 60 min preincubation the released GHRF from sliced rat hypothalamic fragments during 60 min incubation was detected by a highly specific and sensitive radioimmunoassay for rat GHRF. The release of GHRF was Ca2+-dependent and enhanced by high concentration of K+. Insulin-like growth factor-1 (IGF-1) significantly decreased GHRF release to 65% and 84% of the control at concentrations of 10(-8) M and 10(-7) M, respectively. These results suggest that this in vitro system is useful for the investigation of the mechanism of GHRF release from the hypothalamus and that IGF-1 is probably involved in the feedback inhibition of growth hormone secretion by attenuating GHRF release from the hypothalamus besides countering the effect of GHRF on the pituitary. 相似文献

Phorbol myristate acetate (PMA)-stimulated human monocyte-like cells (U-937) were found to synthesize the third component of complement (C3), as shown by enzyme-linked immunosorbent assay and immunoprecipitation from [35S]methionine-labelled culture supernatants. C3 synthesis occurred at a rate of about 160 ng of C3/24 h per 10(6) cells on day 7 after addition of PMA; it was blocked by cycloheximide treatment and was restored after removal of the inhibitor. SDS/polyacrylamide-gel-electrophoretic analysis of the immunoprecipitated protein showed that the size and subunit structure of the newly synthesized C3 were identical with those of plasma C3, and that a single-chain intracellular precursor was present in the cell lysates. Haemolytic assays showed that the synthesized C3 fully expressed functional activity in early culture within 4 h. After longer culture, a loss of haemolytic activity was observed. The possibility that newly secreted C3 is cleaved by U-937 cells themselves was suggested. 相似文献
