Radial growth, asexual sporulation, and cleistothecia formation as well as extracellular chitinase and proteinase formation of Aspergillus nidulans were monitored in surface cultures in order to study the physiological role of extracellular hydrolase production in carbon-stressed cultures. We set up carbon-stressed and carbon-overfed experimental conditions by varying the starting glucose concentration within the range of 2.5 and 40 g/L. Glucose starvation induced radial growth and hydrolase production and enhanced the maturation of cleistothecia; meanwhile, glucose-rich conditions enhanced mycelial biomass, conidia, and cleistothecia production. Double deletion of chiB and engA (encoding an extracellular endochitinase and a β-1,3-endoglucanase, respectively) decreased conidia production under carbon-stressed conditions, suggesting that these autolytic hydrolases can support conidia formation by releasing nutrients from the cell wall polysaccharides of dead hyphae. Double deletion of prtA and pepJ (both genes encode extracellular proteases) reduced the number of cleistothecia even under carbon-rich conditions except in the presence of casamino acids, which supports the view that sexual development and amino acid metabolism are tightly connected to each other in this fungus. 相似文献

Inoculations with both Botrytis cinerea and Geotrichum candidum stimulated ethylene evolution in the pre-climacteric normal tomato fruit and the non-ripening nor mutant which did not show any rise in ethylene when uninfected. In the post-climacteric normal fruits, new peaks in ethylene production were formed. The rise in ethylene evolution in all types of infected fruits has already been detected during the incubation period of the disease. Ethylene peaks were detected earlier and were higher in fruits infected with B. cinerea than with G. candidum, coinciding with the faster rate of growth of the former. Mechanical wounding also stimulated ethylene synthesis by the non-ripening fruits, production being directly proportional to wound dimension. Considerably higher rates of ethylene were recorded for infected fruits than for mechanically-injured fruits in which wound dimensions were similar to those of lesion development. Applying aminoxyacetic acid at the site of inoculation inhibited ethylene production by 55–60 % in the normal fruits and by about 80 % in the nor mutant fruits. A similar pathway of ethylene synthesis was suggested for normally ripening tomato fruit and non-ripening infected tissues. 相似文献

The pattern of proline accumulation and the growth response were followed in several tomato ( Lycopersicon esculentum Mill.) varieties which were exposed to 7 days of drought stress followed by a 15-day period of rewatering. During dehydration, water potential and leaf elongation rates decreased more in var. 'Hosen' and 'S-5' than in 'LX-11', '1970', 'Pakmor', 'Faculty-16', 'Alcobaca' and '475'. Proline accumulation during stress was greatest in the first two varieties. In 'Hosen' and 'S-5' rewatering resulted in a decrease of proline to control levels, whereas in the other varieties accumulation of proline continued long after turgor had been regained. The extent of this continued accumulation was not correlated with the degree to which each variety was dehydrated. Upon rewatering of the plants the rate of leaf elongation was increased, but the final leaf size as well as whole shoot and root fresh weight of the recovered plants were not colated with the degree of "suffering" that each variety experienced during the drought period. Incubation of detached young tomato leaves in polyethylene glycol solution for 48 h resulted in a substantial accumulation of proline. The varietal differences observed under these conditions were reminiscent of the differential responses in proline accumulation obtained in the intact plants. It is concluded that proline accumulation at the time of dehydration signals drought stress in tomato plants but does not correlate with the overall varietal sensitivity to transient dehydration in recovered plants. 相似文献

Mutations of the Adenomatous polyposis coli (APC) gene are common and strongly associated with the development of colorectal adenomas and carcinomas. While extensively studied in modern populations, reports on visceral tumors in ancient populations are scarce. To the best of our knowledge, genetic characterization of mutations associated with colorectal cancer in ancient specimens has not yet been described. In this study we have sequenced hotspots for mutations in the APC gene isolated from 18th century naturally preserved human Hungarian mummies. While wild type APC sequences were found in two mummies, we discovered the E1317Q missense mutation, known to be a colorectal cancer predisposing mutation, in a large intestine tissue of an 18th century mummy. Our data suggests that this genetic predisposition to cancer already existed in the pre-industrialization era. This study calls for similar investigations of ancient specimens from different periods and geographical locations to be conducted and shared for the purpose of obtaining a larger scale analysis that will shed light on past cancer epidemiology and on cancer evolution. 相似文献

The effect of sinusoidally varying magnetic fields (SVMF) on chick embryo fibroblasts (CEF) was examined by two independent methods: 1) measurement of cell proliferation at 0.06–0.7 mT (100, 60 and 50 Hz) using a colorimetric assay (MTT); 2) monitoring of specific activity of adenosine deaminase (ADA) at 0.3 and 0.7 mT, 60 Hz. Both increased cell proliferation and reduced ADA specific activity are associated with cell transformation. The MTT test showed an increase in cell proliferation of up to 64% after a 24 h exposure to SVMF at 100 Hz, 0.7 mT. Cell proliferation at constant frequency (100 Hz) depended on SVMF intensity. Cell proliferation at constant intensity (0.7 mT) increased with increasing field frequency. At 0.7 mT, 60 Hz cell proliferation increased by 31%, 28%, and 26% when measured by hemocytometry, 3H-thymidine incorporation, and the MTT assay, respectively. ADA specific activity in CEF decreased by circa 48% on exposure to SVMF at 60 Hz, 0.3 mT for 24 h; only a statistically insignificant trend was seen at 0.7 mT, 60 Hz. Our findings showed that CEF cell proliferation and ADA specific activity were modified by SVMF. Both methods, independently, qualitatively detect a magnetic field effect. In a previous work we studied the ratio between the amplitudes of the second and first phases (which we call PPR, after peak-to-peak ratio) of the single fibre action potential (SFAP) for a collection of fibrillation potentials (FPs) extracted from two pathological muscles. These FPs showed a wider PPR range than the Dimitrov–Dimitrova (D–D) convolutional model could provide. We proposed a modification of the D–D intracellular action potential (IAP) in order to obtain a range of PPRs comparable to that observed in our FPs. This paper extends that study to a large number of SFAPs extracted from the tibialis anterior muscle of normal subjects. The estimation of the average PPR range of non-diseased muscles in non-fatigued conditions is important since it can be used as a reference to establish a comparison with PPR ranges from muscles suffering some disorder or from fibres that are fatigued. Other aspects of the PPR, as its sensitivity with volume conductor parameters or to what extent changes in the SFAP PPR reflects changes in IAP spatial profile are also examined. We found that the PPR of experimental SFAPs ranges from 0.3 to 2.5 in all subjects and that all PPR histograms contain a well-defined single peak around the PPR value 1.0. 相似文献

IntroductionDopamine (DA) binds to five receptors (DAR), classified by their ability to increase (D1R-like) or decrease (D2R-like) cAMP. In humans, most DA circulates as dopamine sulfate (DA-S), which can be de-conjugated to bioactive DA by arylsulfatase A (ARSA). The objective was to examine expression of DAR and ARSA in human adipose tissue and determine whether DA regulates prolactin (PRL) and adipokine expression and release.MethodsDAR were analyzed by RT-PCR and Western blotting in explants, primary adipocytes and two human adipocyte cell lines, LS14 and SW872. ARSA expression and activity were determined by qPCR and enzymatic assay. PRL expression and release were determined by luciferase reporter and Nb2 bioassay. Analysis of cAMP, cGMP, leptin, adiponectin and interleukin 6 (IL-6) was done by ELISA. Activation of MAPK and PI3 kinase/Akt was determined by Western blotting.ResultsDAR are variably expressed at the mRNA and protein levels in adipose tissue and adipocytes during adipogenesis. ARSA activity in adipocyte increases after differentiation. DA at nM concentrations suppresses cAMP, stimulates cGMP, and activates MAPK in adipocytes. Acting via D2R-like receptors, DA and DA-S inhibit PRL gene expression and release. Acting via D1R/D5R receptors, DA suppresses leptin and stimulates adiponectin and IL-6 release.ConclusionsThis is the first report that human adipocytes express functional DAR and ARSA, suggesting a regulatory role for peripheral DA in adipose functions. We speculate that the propensity of some DAR-activating antipsychotics to increase weight and alter metabolic homeostasis is due, in part, to their direct action on adipose tissue. 相似文献
