尽管几乎所有的农业物种在育种过程中都经历了从多物种环境向密集单种农田的过渡,但对非农业植物物种在单一栽培条件下的表现知之甚少。鉴于最近的研究成果突出杂草物种拟南芥为作物育种提供新视角的可能性,本研究的目的是了解播种密度对拟南芥在不同光周期环境下群体和个体表现的影响。我们研究了拟南芥Cvi-0生态型的表现。Cvi-0的选择基于一项初步实验,发现Cvi-0生态型植物表现出强烈的竞争性能。播种密度处理设置为每株17.6、8.8、4.4、2.2和1.1 cm2,光周期环境为日照12小时或16小时。结果表明,植物在所有密度处理下均获得了恒定的总种子产量。播种密度和日照时间处理会有一些相互作用:在高播种密度和长日照时间的处理下,种子重量更大,而日照时间短的处理下种子重量与密度呈负相关。这些结果阐明了一年生植物在面对激烈的种内竞争时能够采取不同的策略,并有助于为提高群体表现的作物育种提供新的前景。  相似文献

In view of the projected climatic changes and the global decrease in plant species diversity, it is critical to understand the effects of elevated air temperature (Tair) and species richness (S) on physiological processes in plant communities. Therefore, an experiment of artificially assembled grassland ecosystems, with different S (one, three or nine species), growing in sunlit climate-controlled chambers at ambient Tair and ambient Tair + 3°C was established. We investigated whether grassland species would be more affected by midday high-temperature stress during summer in a warmer climate scenario. The effect of elevated Tair was expected to differ with S. This was tested in the second and third experimental years by means of chlorophyll a fluorescence. Because acclimation to elevated Tair would affect the plant's stress response, the hypothesis of photosynthetic acclimation to elevated Tair was tested in the third year by gas exchange measurements in the monocultures. Plants in the elevated Tair chambers suffered more from midday stress on warm summer days than those in ambient chambers. In absence of severe drought, the quantum yield of PSII was not affected by elevated Tair. Our results further indicate that species had not photosynthetically acclimated to a temperature increase of 3°C after 3 years exposure to a warmer climate. Although effects of S and Tair × S interactions were mostly not significant in our study, we expect that combined effects of Tair and S would be important in conditions of severe drought events.  相似文献

The impingement of a submerged, liquid jet onto a cell-covered surface allows assessing cell attachment on surfaces in a straightforward and quantitative manner and in real time, yielding valuable information on cell adhesion. However, this approach is insufficiently characterized for reliable and routine use. In this work, we both model and measure the shear stress exerted by the jet on the impingement surface in the micrometer-domain, and subsequently correlate this to jet-induced cell detachment. The measured and numerically calculated shear stress data are in good agreement with each other, and with previously published values. Real-time monitoring of the cell detachment reveals the creation of a circular cell-free area upon jet impingement, with two successive detachment regimes: 1), a dynamic regime, during which the cell-free area grows as a function of both the maximum shear stress exerted by the jet and the jet diameter; followed by 2), a stationary regime, with no further evolution of the cell-free area. For the latter regime, which is relevant for cell adhesion strength assessment, a relationship between the jet Reynolds number, the cell-free area, and the cell adhesion strength is proposed. To illustrate the capability of the technique, the adhesion strength of HeLa cervical cancer cells is determined ((34 ± 14) N/m2). Real-time visualization of cell detachment in the dynamic regime shows that cells detach either cell-by-cell or by collectively (for which intact parts of the monolayer detach as cell sheets). This process is dictated by the cell monolayer density, with a typical threshold of (1.8 ± 0.2) × 109 cells/m2, above which the collective behavior is mostly observed. The jet impingement method presents great promises for the field of tissue engineering, as the influence of both the shear stress and the surface characteristics on cell adhesion can be systematically studied.  相似文献

Background

Supernumerary sex chromosome aneuploidies (sSCA) are characterized by the presence of one or more additional sex chromosomes in an individual’s karyotype; they affect around 1 in 400 individuals. Although there is high variability, each sSCA subtype has a characteristic set of cognitive and physical phenotypes. Here, we investigated the differences in the morphometry of the human corpus callosum (CC) between sex-matched controls 46,XY (N =99), 46,XX (N =93), and six unique sSCA karyotypes: 47,XYY (N =29), 47,XXY (N =58), 48,XXYY (N =20), 47,XXX (N =30), 48,XXXY (N =5), and 49,XXXXY (N =6).

Methods

We investigated CC morphometry using local and global area, local curvature of the CC boundary, and between-landmark distance analysis (BLDA). We hypothesized that CC morphometry would vary differentially along a proposed spectrum of Y:X chromosome ratio with supernumerary Y karyotypes having the largest CC areas and supernumerary X karyotypes having significantly smaller CC areas. To investigate this, we defined an sSCA spectrum based on a descending Y:X karyotype ratio: 47,XYY, 46,XY, 48,XXYY, 47,XXY, 48,XXXY, 49,XXXXY, 46,XX, 47,XXX. We similarly explored the effects of both X and Y chromosome numbers within sex. Results of shape-based metrics were analyzed using permutation tests consisting of 5,000 iterations.

Results

Several subregional areas, local curvature, and BLDs differed between groups.Moderate associations were found between area and curvature in relation to the spectrum and X and Y chromosome counts. BLD was strongly associated with X chromosome count in both male and female groups.

Conclusions

Our results suggest that X- and Y-linked genes have differential effects on CC morphometry. To our knowledge, this is the first study to compare CC morphometry across these extremely rare groups.

Background

People with Parkinson's disease are twice as likely to be recurrent fallers compared to other older people. As these falls have devastating consequences, there is an urgent need to identify and test innovative interventions with the potential to reduce falls in people with Parkinson's disease. The main objective of this randomised controlled trial is to determine whether fall rates can be reduced in people with Parkinson's disease using exercise targeting three potentially remediable risk factors for falls (reduced balance, reduced leg muscle strength and freezing of gait). In addition we will establish the cost effectiveness of the exercise program from the health provider's perspective.

Methods/Design

230 community-dwelling participants with idiopathic Parkinson's disease will be recruited. Eligible participants will also have a history of falls or be identified as being at risk of falls on assessment. Participants will be randomly allocated to a usual-care control group or an intervention group which will undertake weight-bearing balance and strengthening exercises and use cueing strategies to address freezing of gait. The intervention group will choose between the home-based or support group-based mode of the program. Participants in both groups will receive standardized falls prevention advice. The primary outcome measure will be fall rates. Participants will record falls and medical interventions in a diary for the duration of the 6-month intervention period. Secondary measures include the Parkinson's Disease Falls Risk Score, maximal leg muscle strength, standing balance, the Short Physical Performance Battery, freezing of gait, health and well being, habitual physical activity and positive and negative affect schedule.

Discussion

No adequately powered studies have investigated exercise interventions aimed at reducing falls in people with Parkinson's disease. This trial will determine the effectiveness of the exercise intervention in reducing falls and its cost effectiveness. This pragmatic program, if found to be effective, has the potential to be implemented within existing community services.

Genetic alterations causing constitutive tyrosine kinase activation are observed in a broad spectrum of cancers. Thus far, these mutant kinases have been localized to the plasma membrane or cytoplasm, where they engage proliferation and survival pathways. We report that the NUP214-ABL1 fusion is unique among these because of its requisite localization to the nuclear pore complex for its transforming potential. We show that NUP214-ABL1 displays attenuated transforming capacity as compared to BCR-ABL1 and that NUP214-ABL1 preferentially transforms T cells, which is in agreement with its unique occurrence in T cell acute lymphoblastic leukemia. Furthermore, NUP214-ABL1 differs from BCR-ABL1 in subcellular localization, initiation of kinase activity, and signaling and lacks phosphorylation on its activation loop. In addition to delineating an unusual mechanism for kinase activation, this study provides new insights into the spectrum of chromosomal translocations involving nucleoporins by indicating that the nuclear pore context itself may play a central role in transformation.  相似文献

While irrigation of farm dairy effluent (FDE) to land is becoming popular in New Zealand, it can lead to increased emissions of the greenhouse gas nitrous oxide (N2O). This paper reports the results from trials on N2O emissions from irrigation of FDE to two dairy-grazed pastures on two poorly drained silt-loam soils located at Waikato and Manawatu, New Zealand. These pasture soils were periodically irrigated with FDE under contrasting soil moisture conditions with water-filled pore-space (WFPS) ranging between 26% and 94%. Nitrous oxide emissions were measured from the FDE irrigated and unirrigated sites using large numbers of static chambers (12–20). Irrigation of FDE generally increased N2O emissions compared to the control. N2O emissions varied with changes in climatic conditions and soil WFPS. Overall N2O emissions from effluent-derived N ranged between 0.01% and 4.93% depending on irrigation time and soil WFPS. Lower N2O emissions from FDE were attributable to very low soil WFPS conditions during the dry seasons. Higher N2O emissions were measured from application of FDE to a recently grazed pasture on wet soil. Our results suggest strategic application of FDE during dry summer and autumn seasons can reduce N2O emissions from application of FDE. Delaying effluent-irrigation after grazing events could further reduce N2O emissions by reducing the levels of surplus mineral-N.  相似文献
