Aleppo pine ( Pinus halepensis Mill.) regenerates after wildfires only by germination. This post-fire germination is controlled by temperature, light and the chemical characteristics of the ash-bed. The extremely high pH caused by the ash inhibits germination of Aleppo pine as well as of many other species. Here we report that ash extracts at neutral pH stimulate P. halepensis germination. The effects of ash extracts, nitrate, ammonium (in concentrations present in the ash) and GA3 on the germination of P. halepensis seeds and on lipase activity at early germination were studied under controlled conditions. Sub-optimal concentrations of nitrate- and ammonium-stimulated germination and the activity of lipase in the seeds. It is proposed that nitrate increases seed sensitivity to GA3 while ammonium directly affects lipase activity. The balance between the inhibition of germination by the high pH of ash and its stimulation by the nitrate and ammonium present in the ash determines the overall effect of the ash on post-fire germination of Aleppo pine. The pine seedlings appear in high density among the trees but because of the effects of the ash they appear in sparse stands under the projection of the burned pine canopies. These seedlings benefit from weak inter- and intra-specific competition as well as improved mineral nutrition and have good chances to grow into large trees that will form the regenerating forest; therefore, the enhancement of P. halepensis germination by nitrate and ammonium and its inhibition by high pH can be considered as ecophysiological adaptations to post-fire regeneration.  相似文献

Measuring pollinator performance has become increasingly important with emerging needs for risk assessment in conservation and sustainable agriculture that require multi‐year and multi‐site comparisons across studies. However, comparing pollinator performance across studies is difficult because of the diversity of concepts and disparate methods in use. Our review of the literature shows many unresolved ambiguities. Two different assessment concepts predominate: the first estimates stigmatic pollen deposition and the underlying pollinator behaviour parameters, while the second estimates the pollinator's contribution to plant reproductive success, for example in terms of seed set. Both concepts include a number of parameters combined in diverse ways and named under a diversity of synonyms and homonyms. However, these concepts are overlapping because pollen deposition success is the most frequently used proxy for assessing the pollinator's contribution to plant reproductive success. We analyse the diverse concepts and methods in the context of a new proposed conceptual framework with a modular approach based on pollen deposition, visit frequency, and contribution to seed set relative to the plant's maximum female reproductive potential. A system of equations is proposed to optimize the balance between idealised theoretical concepts and practical operational methods. Our framework permits comparisons over a range of floral phenotypes, and spatial and temporal scales, because scaling up is based on the same fundamental unit of analysis, the single visit.  相似文献

The role of heat-shock in stimulating the germination of soil-stored seeds from fire-following plant species is well known. However, the effects of high pre-germination temperatures on subsequent seedling growth are less well understood. In this study, we examined the effect of pre-germination heat shock at five temperatures (60°, 75°, 90°, 105° and 120°C, each applied for 5 min) on the seedling growth of four, fire-following Fabaceae species from four Mediterranean-type ecosystems; Hippocrepis multisiliquosa (Israel), Gastrolobium villosum (Western Australia), Cyclopia pubescens (South Africa) and Lupinus succulentus (California). Following heat treatment and subsequent germination, seedlings were grown in controlled conditions before being harvested at either 10, 20- or 40 d old. A significant increase in mean dry weight biomass was found at 10 days for Hippocrepis seedlings germinated from seeds pre-heated to 90°C. However, subsequent comparison of mean dry weight biomass for seedlings of this species at 20 and 40 d old showed no significant response to heat shock pre-treatment. Similarly, an initial increase in growth of Gastrolobium seedlings germinated from seeds heated to 90° and 105°C disappeared as the plants matured. Seedling growth of Lupinus and Cyclopia was unaffected by the pre-germination heat treatment of their seeds. Since seedling competition is influenced by the size and growth rates of neighbouring plants, any changes in seedling growth rates as a consequence of the temperature environment experienced by their seeds, may therefore influence patterns of post-fire plant community recovery.  相似文献

Cripto is a multifunctional cell surface protein with important roles in vertebrate embryogenesis and the progression of human tumors. While Cripto has been shown to modulate multiple signaling pathways, its binding partners do not appear to fully explain its molecular actions. Therefore, we conducted a screen aimed at identifying novel Cripto-interacting proteins. This screen led to our identification of glucose-regulated protein 78 (GRP78), an endoplasmic reticulum (ER) chaperone that is also expressed at the surfaces of tumor cells. Here we demonstrate that Cripto and GRP78 interact at the cell surfaces of multiple cell lines and that their interaction is independent of prior association within the ER. Interestingly, short hairpin RNA knockdown of endogenous GRP78 resulted in enhanced transforming growth factor β (TGF-β) signaling, indicating that like Cripto, GRP78 inhibits this pathway. We further show that when coexpressed, GRP78 and Cripto collaborate to antagonize TGF-β responses, including Smad phosphorylation and growth inhibition of prostate cancer cells grown under anchorage-dependent or -independent conditions. Finally, we provide evidence that cells coexpressing GRP78 and Cripto grow much more rapidly in soft agar than do cells expressing either protein individually. Together, our results indicate that these proteins bind at the cell surface to enhance tumor growth via the inhibition of TGF-β signaling.  相似文献

Cripto plays critical roles during embryogenesis and has been implicated in promoting the growth and spread of tumors. Cripto is required for signaling by certain transforming growth factor-beta superfamily members, such as Nodal, but also antagonizes others, such as activin. The opposing effects of Cripto on Nodal and activin signaling seem contradictory, however, because these closely related ligands utilize the same type I (ALK4) and type II (ActRII/IIB) receptors. Here, we have addressed this apparent paradox by demonstrating that Cripto forms analogous receptor complexes with Nodal and activin and functions as a noncompetitive activin antagonist. Our results show that activin-A and Nodal elicit similar maximal signaling responses in the presence of Cripto that are substantially lower than that of activin-A in the absence of Cripto. In addition, we provide biochemical evidence for complexes containing activin-A, Cripto, and both receptor types and show that the assembly of such complexes is competitively inhibited by Nodal. We further demonstrate that Nodal and activin-A share the same binding site on ActRII and that ALK4 has distinct and separable binding sites for activin-A and Cripto. Finally, we show that ALK4 mutants with disrupted activin-A binding retain Cripto binding and prevent the effects of Cripto on both activin-A and Nodal signaling. Together, our data indicate that Cripto facilitates Nodal signaling and inhibits activin signaling by forming receptor complexes with these ligands that are structurally and functionally similar.  相似文献

This study describes the seasonal patterns of seed and cone abortion in natural Pinus halepensis trees and assesses the effects of pollen quantity, tree size and cone location on seed and cone abortion. The cone-set ratio was similar to the expected value, based on literature on woody perennials. The seed-ovule ratio, however, was almost twice as high as its expected value. The investment in protective cone elements was high and did not vary with the seed-ovule ratio of a cone. Pollination factors influenced cone abortion, as indicated by the high abortion rate at the end of the pollination period. Furthermore, abortive cones had lower pollination rates and lower pollen loads than well developed cones. Effects of resource availability were assessed as effects of tree size and cone position on twigs. Small trees aborted more cones than big trees and cone abortion was higher in apical cones than in basal cones. Abortion in P. halepensis is selective, mediated by both pollen quantity and resource availability. The relative importance of pollen and resources is suggested to be flexible, probably varying between trees and years. The high seed-ovule ratio of P. halepensis is the result of high pollination rate and selective abortion. The selective abortion might be due to the high allocation to protective cone elements relative to the allocation to seeds.  相似文献

The death-associated protein (DAP) kinase family includes three protein kinases, DAP kinase, DAP kinase-related protein 1, and ZIP kinase, which display 80% amino acid identity within their catalytic domains and are functionally linked to common subcellular changes occurring during cell death, such as the process of membrane blebbing. Here we show physical and functional cross talk between DAP kinase and ZIP kinase. The two kinases display strong synergistic effects on cell death when coexpressed and physically bind each other via their catalytic domains. Furthermore, DAP kinase phosphorylates ZIP kinase at six specific sites within its extracatalytic C-terminal domain. ZIP kinase localizes to both the nucleus and the cytoplasm and fractionates as monomeric and trimeric forms. Significantly, modification of the DAP kinase phosphorylation sites influences both the localization and oligomerization status of ZIP kinase. A mutant ZIP kinase construct, in which the six serine/threonine residues were mutated to aspartic acid to mimic the phosphorylated state, was found predominantly in the cytoplasm as a trimer and possessed greater cell death-inducing potency. This suggests that DAP kinase and ZIP kinase function in a biochemical pathway in which DAP kinase activates the cellular function of ZIP kinase through phosphorylation, leading to amplification of death-promoting signals.  相似文献

Both male and female solitary bees visit flowers for rewards. Sex related differences in foraging efficiency may also affect their probability to act as pollinators. In some major genera of solitary bees, males can be identified from a distance enabling a comparative foraging-behavior study. We have simultaneously examined nectar foraging of males and females of three bee species on five plant species in northern Israel. Males and females harvested equal nectar amounts but males spent less time in each flower increasing their foraging efficiency at this scale. The overall average visit frequencies of females and males was 27.2 and 21.6 visits per flower per minute respectively. Females flew shorter distances increasing their visit frequency, relative foraging efficiency and their probability to pollinate. The proportion of conspecific pollen was higher on females, indicating higher floral constancy and pollination probability. The longer flights of males increase their probability to cross-pollinate. Our results indicate that female solitary bees are more efficient foragers; females seem also to be more efficient pollinators but males contribute more to long-distance pollen flow.  相似文献

The influence of several spatial parameters on the maximal detection distance of a target by approaching foraging honeybees was examined. The roles of target diameter, color and luminance contrasts have been already demonstrated in earlier studies. The present study used, for the first time, dissected flower like targets that differed in addition to diameter (D) and area (pi(D/2)(2) = Acir) also in the length of contour line (C), the area of the colored "petals" (Acol) and the degree of dissectedness as expressed mainly by the ratio Acol2/C. The color and luminance contrasts were identical for all targets. Our results confirm the importance of size. However, we demonstrate for the first time, that full circular shapes have the greatest maximal detection distance among targets of equal diameters, and even more than dissected targets with equal Acol and double D. The parameter Acol2/C was found as the best predictor of maximal detection distance of vertically presented targets with varying diameter and degree of dissection for honeybee workers. We propose that an increase in the colored area and decrease in contour line is advantageous due to the fact that it increases the amount of contrast that the target as a whole produces against its background.  相似文献
