Several image-based computational models have been used to perform mechanical analysis for atherosclerotic plaque progression and vulnerability investigations. However, differences of computational predictions from those models have not been quantified at multi-patient level. In vivo intravascular ultrasound (IVUS) coronary plaque data were acquired from seven patients. Seven 2D/3D models with/without circumferential shrink, cyclic bending and fluid–structure interactions (FSI) were constructed for the seven patients to perform model comparisons and quantify impact of 2D simplification, circumferential shrink, FSI and cyclic bending plaque wall stress/strain (PWS/PWSn) and flow shear stress (FSS) calculations. PWS/PWSn and FSS averages from seven patients (388 slices for 2D and 3D thin-layer models) were used for comparison. Compared to 2D models with shrink process, 2D models without shrink process overestimated PWS by 17.26%. PWS change at location with greatest curvature change from 3D FSI models with/without cyclic bending varied from 15.07% to 49.52% for the seven patients (average = 30.13%). Mean Max-FSS, Min-FSS and Ave-FSS from the flow-only models under maximum pressure condition were 4.02%, 11.29% and 5.45% higher than those from full FSI models with cycle bending, respectively. Mean PWS and PWSn differences between FSI and structure-only models were only 4.38% and 1.78%. Model differences had noticeable patient variations. FSI and flow-only model differences were greater for minimum FSS predictions, notable since low FSS is known to be related to plaque progression. Structure-only models could provide PWS/PWSn calculations as good approximations to FSI models for simplicity and time savings in calculation.

The phage-encoded proteins required for conservative integration of infecting bacteriophage Mu DNA were investigated. Our findings show that functional gpA, an essential component of the phage transposition system, is required for integration. The Mu B protein, which greatly enhances replicative transposition of Mu DNA, is also required. Furthermore, a truncated form of gpB lacking 18 amino acids from the carboxy terminus is blocked in replicative transposition, but not conservative integration. Our results point to a more prominent role for gpB than simply a replication enhancer in Mu DNA transposition. The ability of a truncated form of B to function in conservative integration, but not replicative transposition, also suggests a key role for the carboxy-terminal domain of the protein in the replicative reaction. The existence of a shortened form of gpB, which uncouples conservative integration from replicative transposition, should be invaluable for future dissection of Mu DNA transposition.  相似文献

Pantoea agglomerans strain Eh1087 produces the phenazine antibiotic D-alanylgriseoluteic acid. A glutamine auxotroph harboring an insertion in a putative glnA gene was obtained by transposon-mutagenesis of Eh1087 that produced less D-alanylgriseoluteic acid than the parental strain (strain Eh7.1). Cosmids encoding the Eh1087 glnA were isolated by their ability to complement the mutant for prototrophy. The role of the Eh1087 glnA locus was functionally confirmed by complementation of an Escherichia coli glnA mutant. Analysis of the nucleotide and deduced amino acid sequences of the Eh1087 glnA gene indicated a high degree of similarity to the glnA genes and glutamine synthetase enzymes of other Enterobacteriaceae. Isotopic labelling experiments with 15N-labelled ammonium sulfate demonstrated that wild-type Eh1087 incorporated 15N into griseoluteic acid more readily than the glnA mutant Eh7.1. We conclude that the 2 nitrogens in the phenazine nucleus originate from glutamine and the intracellular glutamine synthesized by Eh1087 is a source of the phenazine nucleus nitrogens even in glutamine-rich environments.  相似文献

Arterial regions exposed to oscillatory shear (OS) in branched arteries are lesion-prone sites of atherosclerosis, whereas those of laminar shear (LS) are relatively well protected. Here, we examined the hypothesis that OS and LS differentially regulate production of O2- from the endothelial NAD(P)H oxidase, which, in turn, is responsible for their opposite effects on a critical atherogenic event, monocyte adhesion. We used aortic endothelial cells obtained from C57BL/6 (MAE-C57) and p47phox-/- (MAE-p47-/-) mice, which lack a component of NAD(P)H oxidase. O2- production was determined by dihydroethidium staining and an electron spin resonance using an electron spin trap methoxycarbonyl-2,2,5,5-tetramethyl-pyrrolidine. Chronic exposure (18 h) to an arterial level of OS (+/- 5 dynes/cm2) increased O2- (2-fold) and monocyte adhesion (3-fold) in MAE-C57 cells, whereas chronic LS (15 dynes/cm2, 18 h) significantly decreased both monocyte adhesion and O2- compared with static conditions. In contrast, neither LS nor OS were able to induce O2- production and monocyte adhesion to MAE-p47-/-. Treating MAE-C57 with a cell-permeable superoxide dismutase compound, polyethylene glycol-superoxide dismutase, also inhibited OS-induced monocyte adhesion. In addition, over-expressing p47phox in MAE-p47-/- restored OS-induced O2- production and monocyte adhesion. These results suggest that chronic exposure of endothelial cells to OS stimulates O2- and/or its derivatives produced from p47phox-dependent NAD(P)H oxidase, which, in turn, leads to monocyte adhesion, an early and critical atherogenic event.  相似文献

BACKGROUND: Intimal hyperplastic thickening (IHT) is a frequent cause of prosthetic bypass graft failure. Induction and progression of IHT is thought to involve a number of mechanisms related to variation in the flow field, injury and the prosthetic nature of the conduit. This study was designed to examine the relative contribution of wall shear stress and injury to the induction of IHT at defined regions of experimental end-to-side prosthetic anastomoses. METHODS AND RESULTS: The distribution of IHT was determined at the distal end-to-side anastomosis of seven canine Iliofemoral PTFE grafts after 12 weeks of implantation. An upscaled transparent model was constructed using the in vivo anastomotic geometry, and wall shear stress was determined at 24 axial locations from laser Doppler anemometry measurements of the near wall velocity under conditions of pulsatile flow similar to that present in vivo. The distribution of IHT at the end-to-side PTFE graft was determined using computer assisted morphometry. IHT involving the native artery ranged from 0.0+/-0.1 mm to 0.05+/-0.03 mm. A greater amount of IHT was found on the graft hood (PTFE) and ranged from 0.09+/-0.06 to 0.24+/-0.06 mm. Nonlinear multivariable logistic analysis was used to model IHT as a function of the reciprocal of wall shear stress, distance from the suture line, and vascular conduit type (i.e. PTFE versus host artery). Vascular conduit type and distance from the suture line independently contributed to IHT. An inverse correlation between wall shear stress and IHT was found only for those regions located on the juxta-anastomotic PTFE graft. CONCLUSIONS: The data are consistent with a model of intimal thickening in which the intimal hyperplastic pannus migrating from the suture line was enhanced by reduced levels of wall shear stress at the PTFE graft/host artery interface. Such hemodynamic modulation of injury induced IHT was absent at the neighboring artery wall.  相似文献

Samples of grain from three spring barley cultivars of differing malting quality were collected at regular intervals during four weeks prior to harvest. The samples were dried, then assessed for relative grain hardness using the "Milling Energy" test. Ranking order of the cultivars for this character, which relates strongly to malting quality, was unaltered throughout. In a further experiment, it was demonstrated that selection for milling energy could be successfully practised on oven-dried grain collected six weeks after ear emergence.  相似文献

Blood flow velocity was measured in the dog aorta distal to mechanically induced constrictions of various degrees of severity employing an 8-MHz pulsed Doppler ultrasound velocimeter and a phase-lock loop frequency tracking method for extracting velocity from the Doppler quadrature signals. The data were analyzed to construct ensemble average velocity waveforms and random velocity disturbances. In any individual animal the effect of increasing the degree of stenosis beyond approximately 25 percent area reduction was to produce increasing levels of random velocity disturbance. However, variability among animals was such that the sensitivity of random behavior to the degree of stenosis was degraded to the point that it appears difficult to employ Doppler ultrasound measurements of random disturbances to discriminate among stenoses with area reductions less than approximately 75 percent. On the other hand, coherent vortex structures in velocity waveforms consistently occurred distal to mild constrictions (25-50 percent area reduction). Comparison of the phase-lock loop Doppler ultrasound data with simultaneous measurements using invasive hot-film anemometry, which possesses excellent frequency response, demonstrates that the ultrasound method can reliably detect those flow phenomena in such cases. Thus, the identification of coherent, rather than random, flow disturbances may offer improved diagnostic capability for noninvasively detecting arteriosclerotic plaques at relatively early stages of development.  相似文献

Sixty 5 to 8 week old Sprague-Dawley (Crl:CD(SD)BR) rats were inoculated intranasally with 2000 egg infectious doses of egg-propagated Sendai virus. Virus was recovered from the upper respiratory tract and lungs on days 1 through 8 post-inoculation (PI). Serum antibody responses were measured for 12 rats over a 9 month period PI. Antibody was first detected at 7 days, peaked at 21 days, and was detected in 5 of the 12 rats at 9 months. A cell-mediated response, as measured by lymphocyte blastogenesis, also was detected at 7 days and peaked at 21 days, but was not detected at 6 months PI. Lung and serum interferon (IFN) was first detected at 3 hours and peaked at 6 hours, but was not detected by 160 hours. Lung IFN levels were 4 to 10 times those in the serum. These studies indicate that pathogenesis of Sendai virus infection in the rat is similar to that reported in the mouse, but that there are differences in the kinetics of both viral replication and morphologic changes, as described in the companion paper.  相似文献

Abstract Two chronosequences of unsaturated, buried loess sediments, ranging in age from <10,000 years to >1 million years, were investigated to reconstruct patterns of microbial ecological succession that have occurred since sediment burial. The relative importance of microbial transport and survival to succession was inferred from sediment ages, porewater ages, patterns of abundance (measured by direct counts, counts of culturable cells, and total phospholipid fatty acids), activities (measured by radiotracer and enzyme assays), and community composition (measured by phospholipid fatty acid patterns and Biolog substrate usage). Core samples were collected at two sites 40 km apart in the Palouse region of eastern Washington State, near the towns of Washtucna and Winona. The Washtucna site was flooded multiple times during the Pleistocene by glacial outburst floods; the Winona site elevation is above flood stage. Sediments at the Washtucna site were collected from near surface to 14.9 m depth, where the sediment age was approximately 250 ka and the porewater age was 3700 years; sample intervals at the Winona site ranged from near surface to 38 m (sediment age: approximately 1 Ma; porewater age: 1200 years). Microbial abundance and activities declined with depth at both sites; however, even the deepest, oldest sediments showed evidence of viable microorganisms. Same-age sediments had equal quantities of microorganisms, but different community types. Differences in community makeup between the two sites can be attributed to differences in groundwater recharge and paleoflooding. Estimates of the microbial community age can be constrained by porewater and sediment ages. In the shallower sediments (<9 m at Washtucna, <12 m at Winona), the microbial communities are likely similar in age to the groundwater; thus, microbial succession has been influenced by recent transport of microorganisms from the surface. In the deeper sediments, the populations may be considerably older than the porewater ages, since microbial transport is severely restricted in unsaturated sediments. This is particularly true at the Winona site, which was never flooded.  相似文献

Summary Alfalfa plants,Medicago sativa L., were selected from the Florida 66 cultivar for vigor in an acid (pH 4.4, Al≥.4 meq/100 g) and a limed, fertilized (pH 6.5, Al=0 meq/100 g, P and K added) Cecil topsoil. The selected plants were intermated by selection condition to achieve two germplasms, acid selected (A-1) and limed, fertile selected (L-1). ARhizobium meliloti strain (79-4s) was isolated from a high acetylene reducing nodule from a plant in a similar acid soil. The germplasms and the Rhizobium strain were then tested in greenhouse pots for agronomic performance under a variety of soil pH and fertility conditions. The 79-4s inoculum, as well as commercially prepared inoculum, gave better plant yield and acetylene reduction (N2-fixation) at all harvests when compared to a sterile peat control, but the commercial inoculum was the best inoculum treatment. The A-1 germplasm produced higher shoot dry weight at the final harvest than did the L-1 germplasm at all soil pH’s when P and K were applied at the highest rates. The A-1 germplasm also had better root weight (mainly fibrous roots) and acetylene reduction in these soil conditions. The two germplasms appear to be genetically distinct and respond differently depending on soil pH and fertility conditions.  相似文献
