Average specific density of individual cells of pure cultures of Chromatium warmingii and Chromatium vinosum were measured by isopicnic gradient centrifugation with Percoll during growth at constant illumination as a function of the increasing content of intracellular sulfur. Cell number and volume, bacteriochlorophyll a, sulfide, and sulfur were followed in the cultures along with cellular buoyant density. Poly--hydroxybutyrate was monitored at several points during growth of the cultures. The density of C. warmingii changed from 1.071 to 1.108 g cm-3 (sulfur content per cell varied from 0 to 1.71pg). C. vinosum changed its density from 1.096 to 1.160 g cm-3 (sulfur content per cell varied from 0 to 0.43 pg). Maximum sulfur content in pg of sulfur per m3 of cell volume were 0.178 for C. warmingii and 0.294 for C. vinosum. Measurement of the differences in buoyant density, volume and sulfur content before and after ethanol extraction of cells with and without intracellular sulfur, allowed tentatively to estimate the density of sulfur inside the cells as 1.219 g cm-3. Monomolecular layers of lipid extracts of microsomal, mitochondrial outer and inner membranes, and pure lipid species have been used to measure their interaction with apo- and holocytochrome c. Large differences were observed both with respect to the nature and the lipid specificity of the interaction. The initial electrostatic interaction of the hemefree precursor apocytochrome c with anionic phospholipids is followed by penetration of the protein in between the acyl chains. Apocytochrome c shows similar interactions for all anionic lipids tested. In strong contrast the holoprotein discriminates enormously between cardiolipin for which it has a high affinity and phosphatidylserine and phosphatidylinositol for which it has a much lower affinity. For these latter lipids the interaction with cytochrome c is primarily electrostatic. The cytochrome c-cardiolipin interaction shows several unique features which suggest the formation of a specific complex between the two molecules. These properties account for the preference in interaction of the apoprotein with the lipid extract of the outer mitochondrial membrane over that of the endoplasmic reticulum and the large preference of cytochrome c for the inner over that of the outer mitochondrial membrane lipid extract. Only apocytochrome c was able to induce close contacts between monolayers of the mitochondrial outer membrane lipids and vesicles of mitochondrial inner membrane lipids. Experiments with fragments of both protein and unfolding experiments with cytochrome c revealed that the differences in interaction between the two proteins are mainly due to differences in their tertiary structure and not the presence of the heme group itself. The initial unfolded structure of apocytochrome c is responsible for the high penetrative power of the protein and its ability to induce close membrane contact, whereas the folded structure of cytochrome c is responsible for the specific interaction with cardiolipin. The results are discussed in the light of the apocytochrome c import process in mitochondria and suggest that lipid-protein interactions contribute to targeting the precursor toward mitochondria and are important for its translocation across the outer mitochondrial membrane and the final localization of cytochrome c toward the outside of the inner mitochondrial membrane.  相似文献

The presence and distribution of fibronectin and actin in breast fibroadenoma cells have been investigated in frozen sections and primocultures of the same samples, by means of indirect immunofluorescence techniques. In the tissue cells, epithelial cells were negative whereas myoepithelial cells were positive with the two antibodies. Moreover, fibronectin was mainly distributed in basal lamina of ducts and ductules whereas actin appeared to label stroma cells. In primocultures obtained from the same samples of fibroadenomas, the labelled patterns of fibronectin and actin were not really distinguishable between cells; the two protein distributions were quite usual. Our data on human breast fibroadenoma cells are in agreement with the concepts of cellular adaptation or selection in culture and emphasize the difficulty in correlating results of "in vitro" and "in vivo" studies.  相似文献

Plant–animal mutualistic networks are interaction webs consisting of two sets of entities, plant and animal species, whose evolutionary dynamics are deeply influenced by the outcomes of the interactions, yielding a diverse array of coevolutionary processes. These networks are two‐mode networks sharing many common properties with others such as food webs, social, and abiotic networks. Here we describe generalized patterns in the topology of 29 plant–pollinator and 24 plant–frugivore networks in natural communities. Scale‐free properties have been described for a number of biological, social, and abiotic networks; in contrast, most of the plant–animal mutualistic networks (65.6%) show species connectivity distributions (number of links per species) with a power‐law regime but decaying as a marked cut‐off, i.e. truncated power‐law or broad‐scale networks and few (22.2%) show scale‐invariance. We hypothesize that plant–animal mutualistic networks follow a build‐up process similar to complex abiotic nets, based on the preferential attachment of species. However, constraints in the addition of links such as morphological mismatching or phenological uncoupling between mutualistic partners, restrict the number of interactions established, causing deviations from scale‐invariance. This reveals generalized topological patterns characteristic of self‐organized complex systems. Relative to scale‐invariant networks, such constraints may confer higher robustness to the loss of keystone species that are the backbone of these webs.  相似文献

Some primitive meteorites are carbon-rich objects containing a variety of organic molecules that constitute a valuable record of organic chemical evolution in the universe prior to the appearance of microorganisms. Families of compounds include hydrocarbons, alcohols, aldehydes, ketones, carboxylic acids, amino acids, amines, amides, heterocycles, phosphonic acids, sulfonic acids, sugar-related compounds and poorly defined high-molecular weight macromolecules. A variety of environments are required in order to explain this organic inventory, including interstellar processes, gas-grain reactions operating in the solar nebula, and hydrothermal alteration of parent bodies. Most likely, substantial amounts of such organic materials were delivered to the Earth via a late accretion, thereby providing organic compounds important for the emergence of life itself, or that served as a feedstock for further chemical evolution. This review discusses the organic content of primitive meteorites and their relevance to the build up of biomolecules.  相似文献

Although temporary cyst stages are common in dinoflagellates,their role remains unclear. Every year Alexandrium taylori (Dinophyceae)forms dense patches (106 cells l-1) along La Fosca beach (Spain,northwest Mediterranean), which last for 2 months (July, August).One of the characteristics of the life history of A. tayloriis the shift from a vegetative motile stage to non-motile temporarycysts. Here we present the temporal changes in the abundanceof temporary cysts in sediments and their in situ encystmentand excystment rates. The in situ encystment rate of temporarycysts from the water column to the sediment ranged from 1.8x 106 to 4.4 x 106 cysts m-2 day-1, whereas the excystment ratewas between 0.9 x 106 to 2.7 x 106 cysts m-2 day-1 during thebloom period. Some of the temporary cysts in the sediment tookmore than 1 day to produce vegetative cells and remained viablefor at least 4 days. We propose that temporary cyst formationin this species is a tool for reducing population losses. Theproduction of temporary cysts can be an advantage since partof the population is stored in the sediments.  相似文献

In this issue of Neuron, Goodchild et al. show that torsinA knockout and knockin mice have similar phenotypes, which suggests that the mutant torsinA allele causes disease because it has decreased function. Mycobacterium tuberculosis (M. tb) pathogenesis involves the interaction between the mycobacterial cell envelope and host macrophage, a process mediated, in part, by binding of the mannose caps of M. tb lipoarabinomannan (ManLAM) to the macrophage mannose receptor (MR). A presumed critical step in the biosynthesis of ManLAM, and other mannose-containing glycoconjugates, is the conversion of mannose-6-phosphate to mannose-1-phosphate, by a phosphomannomutase (PMM), to produce GDP-mannose, the primary mannose-donor in mycobacteria. We have identified four M. tb H37Rv genes with similarity to known PMMs. Using in vivo complementation of PMM and phosphoglucomutase (PGM) deficient strains of Pseudomonas aeruginosa, and an in vitro enzyme assay, we have identified both PMM and PGM activity from one of these genes, Rv3257c (MtmanB). MtmanB overexpression in M. smegmatis produced increased levels of LAM, lipomannan, and phosphatidylinositol mannosides (PIMs) compared with control strains and led to a 13.3 +/- 3.9-fold greater association of mycobacteria with human macrophages, in a mannan-inhibitable fashion. This increased association was mediated by the overproduction of higher order PIMs that possess mannose cap structures. We conclude that MtmanB encodes a functional PMM involved in the biosynthesis of mannosylated lipoglycans that participate in the association of mycobacteria with macrophage phagocytic receptors.  相似文献

The present study was aimed (1) to investigate the effect of cholesterol and fat enriched diets on the development of steatohepatitis in apolipoprotein E-knockout mice, and (2) to study the chronological relationships between the development of hepatic alterations, hypercholesterolemia and atherosclerotic lesions in this experimental model. The study consisted of two protocols. Protocol 1 was used in 90 mice subdivided in groups of 18. For 10 weeks, each group was given a diet with different fat and cholesterol contents. Protocol 2 was used in 42 mice, subdivided in four groups. Each group was given a diet enriched with cholesterol and palm oil and they were sacrificed at 8, 13, 18 and 24 weeks of age. Results were as following. (1) Mice given high fat/high cholesterol diets developed an impairment of liver histology consisting of fat accumulation, macrophage proliferation, and inflammation. (2) These effects were modulated by the type of fat: olive oil was mainly associated with macrovesicular steatosis and cholesterol plus palm oil with severe steatohepatitis. (3) There was a chronological and quantitative relationship between liver impairment and the formation of atheromatous lesions. We conclude that apolipoprotein E-knockout mice may be a useful model for investigating the mechanisms of diet-induced steatohepatitis. (Mol Cell Biochem 268: 53–58, 2008)  相似文献
